Abstract

Lymphatic filariasis is a serious public health problem in llie world including in Indonesia. The number of endemic areas increased with high incident and chronic patients. In urban areas, the disease is caused by filarial nematodes, called Wucheria bancrofi, and is spread through the bite of infected mosquitoes, especially the species of'Culex q u i n q u e f a s c i a t u s Say. Host-parasite relationship remains unknown, as well as the geographical differences of the vector. The objective of this research was to determine the genetic variability of mosquitoe vector Cx. q u i n q u e f a s c i a t u s f r o m t w o f i l a r i a s i s e n d e m i c a r ea s , P e k a l o n g a n D i s t r i c t . Mosquitoes were collected indoors and outdoors by using "man landing biting" method andsweapnet. T h e D N A e x t r a c t i o n from e a c h s i n g l e f e m a l e m o s q u i t o w a s p e r f o r m e d , a n d t h e n t h e D N A w a s u s e d as a P G R t e m p l a t e w i t h o l i g o n u c l e o t i d e R A P D p r i m e r O P A 1 1 . T h e P G R p r o d u c t s w e r e a n a l y z e d b y e l e c t r o p h o r e s i s i n 1 % ( w / v ) a g a r o s e g e l . T h e r e s u l t s h o w e d 15 l o c i w i t h 1 0 0 % p o l y m o r p h i s m .

L y m p h a t i c filariasis i s a d i sease c a u s e d b y filarial i n f e c t i o n o n l y m p h a t i c v e s s e l , t h a t m a y d u e t o n e m a t o d e s i n f e c t i o n o f Wuchereria bancrofti, Brugia malayi, o r Brugia timori, a n d w i l l t r a n s m i t t h r o u g h t h e b i t i n g o f i n f e c t e d m o s q u i t o e s . T h i s d i sease i s o n e o f s e r i o u s p r o b l e m i n p u b l i c h e a l t h , because o f e c o n o m i c l o s s a n d p r o d u c t i v i t y [ 1 , 2 ] . I t has r e p o r t e d t h a t t h e r e i s 1 1 9 m i l l i o n cases w i t h l y m p h a t i c filariasis, a n d m o r e t h a n 9 0 % o f cases c a u s e d b y fV. Bancrofti i n f e c t i o n [ 3 ] . L y m p h a t i c filariasis has b e e n t a r g e t e d f o r e l i m i n a t i o n b y t h e y e a r 2 0 2 0 b y W o r l d H e a l t h O r g a n i z a t i o n [ 4 ] .

I n I n d o n e s i a , t h i s d i s e a s e h a s f o u n d i n m o r e t h a n 3 0 p r o v i n c e s , i n c l u d i n g 2 3 1 d i s t r i c t s t h a t h a v e c o n f i r m e d as e n d e m i c ?r?a§. A s r e p o r t e d i n 2 0 0 5 t h a t m o r e t h a n 2 0 m i l l i o n p e o p l e l i v e i n e n d e m i c a reas , a n d 8 ,243 c h r o n i c p a t i e n t w a s d e t e c t e d [ 4 ] . I t w i l l a s s u m e d t h a t i n t h e fiiture t h e e n d e m i c a r e a a n d t h e i n c i d e n t w i l l i n c r e a s e d .

T h e d i s t r i b u t i o n o f t h e s e t h r e e n e m a t o d e s w a s v a r y a n d d e p e n d o n m o s q u i t o e s spec ie s , b i o n o m i c a n d g e o g r a p h i c d i f f e r e n c e s . T h e r e i s 6 8 s p e c i e s o f m o s q u i t o e s s e r v e as v e c t o r f o r W.bancrofti , from t h e g e n u s Culex, Aedes, Anopheles d a n Mansonia. [ 5 ] . Culex quinquefasciatus i s a c o s m o p o l i t m o s q u i t o i n h u m a n h a b i t a t i o n s . U n t i l n o w , h o s t - p a r a s i t e r e l a t i o n s h i p r e m a i n s u n k n o w n , a s w e l l a s t h e g e o g r a p h i c a l d i f f e r e n c e s o f t h e v e c t o r . T h e r e i s l a c k i n f o r m a t i o n i n t h e g e n e t i c h e t e r o g e n i c i t i e s o f Cx. quinquefasciatus [ 7 ] , e s p e c i a l l y i n I n d o n e s i a . T h e r e f o r e , t h e o b j e c t i v e o f t h i s s t u d y w a s t o d e t e r m i n e t h e g e n e t i c v a r i a b i l i t y o f Cx. quiruiuefasciatus p o p u l a t i o n from t w o filariasis e n d e m i c a r e a s i n P e k a l o n g a n D i s t r i c t

I n 2 0 0 7 t h e r e w a s 3 . 9 % o f m f - r a t e w i t h 4 filarial c h r o n i c p a t i e n t a t S i m b a n g K u l o n v i l l a g e B u a r a n s u b d i s t r i c t a n d has b e e n f o l l o w e d b y m a s s d r u g t r e a t m e n t , a n d i t has s i g n i f i c a n t e f f e c t i n r e d u c i n g m f - r a t e , b e c o m e s 2 . 6 % i n 2 0 0 9 [ 8 ] . F u r t h e r n i o r e , i t h a s a l s o r e p o r t e d t h a t m / r a t e w a s 2 . 3 5 % a t P a s i r s a r i v i l l a g e w i t h 1 7 p e r s o n h a v e c l i n i c a l s i g n a n d 1 p e r s o n w i t h filarial c h r o n i c [ 9 ] .

T e c h n i q u e has b e e n i m p r o v e i n t h e s t u d y o f p a r a s i t e r e l a t i o n s h i p w i t h h o s t a n d t h e e n v i r o n m e n t S i m i l a r s t u d y h a v e d o n e i n I n d i a a n d o t h e r c o u n t r i e s [ 7 , 1 7 , 1 8 ] . I t i n d i c a t e d tha t i n t h e fiiture t h e r e w i l l a l o t o f s t u d y s h o u l d be d o n e i n I n d o n e s i a , b e c a u s e o f t h e g e o g r a p h i c a l l y d i f f e r e n c e s o f d i s t r i b u t i n g o f m o s q u i t o e s a n d pa ras i t e . A s w e k n o w , t h a t I n d o n e s i a has 3 spec ie s o f filarial p a r a s i t e W.bancofti, Brugia malayi, a n d Brugia timori , a n d has d i f f e r e n t m o s q u i t o v e c t o r s p e c i e s [ l , 2 , 1 9 ] .
