FOR SEVERAL YEARS NOW, I HAVE BEEN INVESTING MY TIME WITH THE FOUNDATION OF STARS. AT 14, I WAS DIAGNOSED WITH ACUTE LYMPHOBLASTIC LEUKEMIA AND I WOULD SAY THAT IT IS ONLY THANKS TO SCIENTIFIC ADVANCES THAT I’M HEALTHY TODAY. THANKS TO EACH AND EVERY ONE OF YOU BECAUSE EVERY GESTURE, HOWEVER SMALL, MAKES A DIFFERENCE AND HELPS A STAR TWINKLE BRIGHTER. Daniela, Spokesperson for the Children

ANNUAL REPORT 5

JUST A FEW OF THE ACHIEVEMENTS IN PEDIATRIC RESEARCH SINCE THE BEGINNING OF THE FOUNDATION:

PROGRESS MADE WITH YOUR SUPPORT:

In 2010, Dr. Guy Rouleau from CHU Sainte-Justine garnered world- wide recognition for his discovery of mutations in genetic predisposition to autism

and schizophrenia. Among his great achievements is the DISCOVERY OF MORE THAN 20 GENES RESPONSIBLE FOR DISEASES, and the discovery of new mutational mechanisms.

In June 2010, the team of researchers from the Montreal Children’s Hospital made a genetic breakthrough, recognized by L’Actualité Magazine as one

of the 35 most important inventions: it is now possible to TRACK ANY GENETIC DISEASE IN RECORD TIME with a powerful and reliable gene sequencing technique.

In April 2016, researchers at the Montreal Children’s Hospital also discovered

an experimental treatment to REDUCE THE SIZE OF THE PEDIATRIC BRAIN TUMOURS. This treatment is less invasive than conventional care and also avoids the side effects of radiotherapy and chemotherapy.

Over the last 45 years, the average survival age of children with cystic fi brosis

has increased from 5 TO 47 YEARS.

Only 10 years ago, the average survival rate of patients with brain cancer

was less than 1 year but, thanks to recent scientific breakthroughs, THIS HAS INCREASED TO 18 MONTHS. Some patients can even receive a prognosis of 3 to 5 years.

In 30 years, the survival rate of children with acute lymphoblastic leukemia rose

from 20% TO ALMOST 85%.

In 20 years, the number of Sudden Infant Death Syndrome (SIDS deaths) has

FALLEN BY MORE THAN HALF.

Over the last 10 years, the evaluation and treatment of children with viral

bronchiolitis have greatly improved, which SIGNIFICANTLY REDUCES THE RISK OF COMPLICATIONS.

Research has allowed doctors to save the lives of children born prematurely,

including those weighing just 500g, and ensure their LONG-TERM QUALITY OF LIFE.

DID YOU KNOW that...

ANNUAL REPORT 6

It is with great pride and confi dence that I present the 2015-2016 Foundation of Stars Annual Report.

To address current challenges in philanthropy, we looked at this year as a turning point to stand out and better pursue our mission of excellence, and to focus on more promising research projects.

Message from the CHAIRMAN OF THE BOARD

Jacques Dubuc

OUR COLLABORATION WITH EXPERT COMMITTEES ENSURES GREATER OBJECTIVITY WHEN SELECTING PROJECTS. THEREFORE, IT IS THE WELL-BEING OF SICK CHILDREN THAT EXEMPLIFIES OUR PARAMOUNT CONCERN.

Our goal remains the same: to raise funds for research into children’s diseases and allow new researchers to fl ourish, offering sick children and their families a better life. That is the soul and the purpose of the Foundation of Stars.

Looking to the future, the Foundation of Stars continues to work with the Fonds de recherche du Québec – Santé (FRQS). Together, we offer training grants and career bursaries in pediatric research investing up to $500,000 each to fund these scholarships in the upcoming years; a major and essential contribution of more than a million dollars for research into children’s diseases in Québec.

In addition to these funds, we are developing other collaborative, large-scale projects that will allow the Foundation of Stars to provide optimized support for pediatric research by being there where the need is greatest. Our collaboration with expert committees ensures greater objectivity when selecting projects. Therefore, it is the well-being of sick children that exemplifi es our paramount concern.

Through these partnerships, we are getting closer every day to our ultimate goal: to provide children a future free of diseases and suffering.

I would like to thank the members of the Board who are leaving us this year and I want to acknowledge their invaluable contribution to the Foundation of Stars: Danielle Gauthier, Martin Godbout, Martine Grégoire and Stella Riggi. I also want to extend a warm welcome to new members who have joined us: Alain Auclair, Yves Léveillé, Robert Racine and Dr. Guy Rouleau.

I also hereby express my gratitude to our passionate staff – our most important resource, our loyal volunteers, our essential institutional and corporate partners, our Governors, our dedicated Board of Directors and our President and CEO.

Finally, a special thank you to all of you, our generous donors, benefactors and partners, who offer us the support that assures the vitality of our mission.

ANNUAL REPORT 7

STILL SO MUCH TO DO

Each year, hundreds of children die from untreatable diseases; it is an unspeakable injustice. These children who cannot defeat their diseases are our daily inspiration.

However, advancements in science and the speed at which technology is evolving feed our hope and allow no more excuses: why delay the development of treatments and cures against diseases that pediatric medicine has not yet found?

We live in a world where imminent discoveries are about to save children’s lives, a world where seeing a child suffer is no longer tolerable.

But government subsidies are decreasing and are not enough. To continue to advance and achieve concrete results, research on childhood diseases needs your support.

We are very grateful to all our benefactors and donors who ensure that the Foundation of Stars remains the largest non-governmental source of funding for research into children’s diseases.

I sincerely thank our remarkable team for working every day with passion and determination to help improve the lives of sick children.

I would also like to highlight the work of our dedicated volunteers for the many hours they have devoted to our cause and thank our generous partners for their essential contribution. Without them, it would be impossible to continue our work.

Finally, I would like to thank our researchers without whom the mission of the Foundation of Stars could not exist, and I warmly thank our Board of Directors for their outstanding contribution to the development of the Foundation.

Message from the PRESIDENT AND CEO

Josée Saint-Pierre

WE LIVE IN A WORLD WHERE IMMINENT DISCOVERIES ARE

ABOUT TO SAVE CHILDREN’S LIVES, A WORLD WHERE SEEING A CHILD

SUFFER IS NO LONGER TOLERABLE.

Six years ago, when Maëlle was just 7 years old, and was already diagnosed with mild autism, she was further struck down by Myasthenia Gravis, an auto-immune neuromuscular disease rarely seen in children. After several relapses, hospitalizations, hundreds of treatments in hematology-oncology and immunology, after infections of all sorts and risky operations, Maëlle is still with us at 13.

It is through the development of biomedical engineering and the discovery of new anti-cancer medications that we can treat certain auto-immune diseases, including the one Maëlle suffers from, that we still have the pleasure of seeing her grow.

Being struck by an incurable illness means that you never know what tomorrow holds. We feel a certain urgency to live each day to its fullest and to be close to our loved ones. Maëlle is now in Grade 8 at High School. Later, she wants to be a painter and to work fi nding shelter for abandoned animals.

SAMUEL, A COURAGEOUS YOUNG ARTIST

Childhood is short and precious. When a child has to sacrifi ce a part of his or her childhood to fi ght against a disease, we all feel the horrible injustice of the situation.

In May 2011, when he was only 5 years old, doctors discovered that Samuel was suffering from a brain tumour. Seven months later, he tragically lost sight in his right eye. Since then, doctors have performed surgery on his brain 6 times, carried out 4 chemotherapy regimens including an experimental one, without actually countering the tumour growth.

In January 2016, Samuel began a 5th chemotherapy protocol and is hoping for success.

Because Samuel’s disease is very diffi cult to treat, we know that the road to recovery is still long but our main hopes are grounded on research.

Doctors and researchers still have a lot of work to do to understand how brain tumours form and to fi nd the best treatment for each child under their care. This is why it is vital that people continue to support the Foundation of Stars.

A STAR Samuel

• 6 brain surgeries

• 2 surgeries for the installation of a catheter for receiving chemotherapy

• 5 chemotherapy regimens, including 2 experimental ones

• Multiple exams and medical appointments

Even if Samuel was worried about the surgeries, he knew and understood that he had no choice and that they could perhaps stop his unbearable headaches – which they did!

MY BIGGEST DREAM IS THAT ONE DAY MY TUMOUR DISAPPEARS AND ALL SICK

CHILDREN GET HEALTHY. Samuel, 10 years.

ANNUAL REPORT 10

A STAR Liam

IN 98% OF CASES BOTH PARENTS CARRY THE DEFECTIVE GENE, AS DOES 1 IN 40 PEOPLE.

On September 13, 2015, our little warrior celebrated his 3rd birthday! Dad, Mom and a lot of people were, and are, very proud of Liam!

When he was 5 months old, he was struck by Spinal Muscular Atrophy Type 1 (SMA), the most severe version of this type of disease. SMA can occur at birth or, more often, between 3 and 6 months of age. Most doctors must then sadly inform parents that their children with SMA Type I will probably not live more than 2 years. It is actually quite exceptional if some children live longer than that (about 80% of them die before their 1st birthday).

It took 3 ½ years for us to be at peace with the idea of cutting his hair. Hair is a kind of Biomarker and evidence of time passing, of life, of living; and it is a very symbolic moment for our family. We also kept the lock of hair of the fi rst haircut of Liam’s big brother Malou. It is a nostalgic thing that we do, but it took on a new dimension with the gloomy prognosis that hovered over Liam.

Not having enough time left to see our child with long hair, afraid that this might be the last time we cut his hair, we want to have the memory of long hair as long as possible in case the inevitable happens. Fear that between two shaves, Liam will no longer be able to touch and pull my beard ... in short, one small step for Liam, a big one for Dad.

Finally, 3 months later, his hair is quite long and we will once again collect the cuttings this summer and many more in the future!

SMA Type 1 is a genetic disease, and in 98% of cases both parents carry the defective gene, as does 1 in 40 people. After a few weeks or months of life, children are weakened (“hypotonic”) because their motor-function muscles are affected. They cannot stand up, have diffi culty rolling over, and cannot sit up without help. Children can hold onto objects at fi rst, as muscle damage starts with the root of the limbs (hips for legs and the shoulders for arms), then onto the extremities (hands and feet). Those affected by this disorder are perfectly normal, often with brilliant minds, they are quite lively, like to communicate and are very conscious about the world around them.

We fi ght for the right treatments for Liam. So far, his condition is stable and he has made incredible progress considering the circumstances. We are so proud of our little boy and we fi ght tirelessly to bring awareness to the disease that is attacking him. We even created a website (www.letliamlive.com) and are active on social networks to share our daily struggles.

Please follow us on our social networks and thank you for supporting pediatric research.

Liam’s Parents, Emmanuelle and Yan.

As for other events in addition to corporate events, the Foundation of Stars had the opportunity to rely on the support of faithful partners year after year. By raising money through various fundraising activities, they ensure the future of pediatric research and offer real hope for healing sick children. Thank you!

CHANTAL LACROIX AND JIMMY SÉVIGNY’S MOI POUR TOI CHALLENGE

Chantal Lacroix and Jimmy Sévigny organized a challenge that brought people together on September 26th and 27th, 2015. Participants ran from Montreal to Québec to support pediatric research!

It was a transformational and unparallelled motivational experience that Chantal Lacroix and Jimmy Sévigny, stars of the popular show Maigrir pour Gagner, offered to the Québec population: the Challenge SOS Santé - Moi pour Toi!

In teams of 8, almost 150 great runners answered the call and ran a relay between Montreal and Québec to support the Foundation of Stars. Running to fi nd the path to better health, to overcome diseases, to run for someone important to them, to support sick children: there were all kinds of motivation that allowed each runner to keep running despite tough times.

Most of the runners will remember this challenge and the public who cheered them up along the 5 stages of the relay: Montreal, Berthierville, Trois-Rivières, Portneuf and Québec. The Foundation of Stars is very proud to have been able to rely on these generous people. Through their continuous efforts, they managed to raise more than $80,000! A great achievement in a very short time frame!

Today, autism affects 1 child out of 88 and it touches 5 times more boys than girls. A rapid increase in the frequency of autism suggests the contribution of environmental factors, in addition to already known genetic risk factors (20-30%). More research on maternal infections will provide new evidence of the role of common and modifi able environmental factors infl uencing autism pathophysiology during pregnancy. The results of the project could pave the way to create new methods for preventive treatment of autism, or at least reduce its severity.

VANESSA BAO

McGill UniversityPh.D. Student

Vanessa’s research project focuses on the way individuals with Autism Spectrum Disorders (ASD) integrate (combine) information that they see and hear. Researchers have thought that people with ASD may have a more diffi cult time putting pieces of information together than people without ASD. The goal of her project is to help fi gure out if there is a difference in the way sensory information is combined in ASD.

Since 2015, the scope of research projects as well as leading researchers were selected by the FRQS (Fonds de recherche du Québec-Santé) and they have benefi ted from funding equally from the FRSQ and the Foundation of Stars to advance research on childhood diseases.

We live in a world where imminent discoveries are about to save children’s lives. For this reason, we want to keep on providing greater support to the best students and researchers, and support the most innovative projects. All this is done to increase the scale of our action and, furthermore, the range of support from our benefactors geared towards the same goal: to provide children with a better and healthier future.

Type 1 Diabetes is an increasingly common disorder that can affect several systems of the body including kidney, eye, cardiovascular and neurological functions. As well, children with Type 1 Diabetes can experience frequent fl uctuations in blood glucose levels affecting multiple aspects of cognition. Even if these effects are subtle, they may put some children with Type 1 Diabetes at an academic disadvantage. The broad aim of the research is to better understand the impact of Type 1 Diabetes on children’s school experiences and academic outcomes. The overarching goal is to improve the health and school experiences of children living with Type 1 Diabetes.

In the fi eld of pediatrics, nasal ventilatory support is used more and more often in respiratory failure situations to avoid complications related to ventilation through an endotracheal tube. The insuffl ations of positive pressure gas in the airways may cause an air passage into the esophagus and gastric distension, a source of Gastroesophageal Refl ux Disease (GERD) and sometimes major complications. The general objective of this research is to carry on the investigation of the effects of nasal ventilatory support on GERD.

Cerebral Palsy (CP) is a common condition in pediatric rehabilitation and more than half of the sufferers cannot walk without help.We believe that children who have the most diffi cult motor adaptations to walk following the completion of a demanding physical activity (tiring) will be those with the lowest level of physical activity and who feel useless during physical activities. This hypothesis has not been clearly evaluated in previous studies in the population and CP will be more fully explored in this study.

Hemophilia is a rare genetic anomaly that increases the normal time it takes for blood to coagulate. This can cause bleeding in the joints and lead to damage and complications such as limited range of motion, severe arthritis and chronic pain. Although the disease is incurable for now, prevention of bleeding is administered through an injection treatment of concentrated coagulation factors as well as engaging in recommended sports activities and avoiding unsuitable sports (contact and speed sports). The study aims to understand what could infl uence young people to ignore the recommendations. The ultimate goal is to support patients and their families so that they have a good quality of life, in the long-term.

Asthma is the most common chronic disease in children, affecting 10% of Canadian children. Scientifi c literature suggests that early signs of chronic respiratory dysfunction as adults are present in the fi rst years of life. Considered together, the benefi cial effect of asthma control medications, the evolution of the disease, and the use and cost of health care could provide the fi rst solid evidence of the long-term impact of asthma control drugs taken during the early years of childhood. Such a conclusion could seriously challenge the current health management of affected preschoolers who are rarely studied despite their high morbidity rates.

Among children, depression contributes to impairments at school, peer relationships and family functioning (Rutter, 2006). Exposure to maternal stress is one factor that has been linked to an increase in symptoms of depression among children (Oberlander et al., 2010). Moreover, Negative Emotionality (NE), a temperamental trait characterized by sadness, fearfulness and stress reactivity, has also been associated with childhood depression (Hyde et al., 2008). The objective of my research is to build on these fi ndings to determine if the relationship between early childhood NE and childhood depression is independent from or infl uenced by maternal depression and genotype.

Proliferative retinopathy is a leading cause of Retinopathy of Prematurity (ROP), i.e. blindness affecting premature infants, and Diabetic Retinopathy (DR), i.e. diabetes affecting the working population. Persistent or acute damage on the retina blocks its normal development and initiates blood vessels degeneration, resulting in hypoxia, lack of oxygen and nutrition delivered to the retina. Current dogma claims that hypoxia is the main factor of excessive and misguided revascularization. However, we question if energy defi ciency would play a critical role in revascularization since the retina is a highly energy-demanding tissue.

Laurence’s study is to characterize the immune abnormalities of children with ENI (Exposed but Non-Infected). HIV is a virus that can be transmitted from mother to child during pregnancy, childbirth or through breastfeeding. Fortunately, the administration of antiretroviral drugs to the mother during pregnancy and the development of a preventive therapy in newborns can prevent transmission of the virus to the child. However, regardless of these treatments, there is a growing number of children who have ENI despite the absence of HIV infection, and these children show more concerning immune characteristics.

When babies are born prematurely, their lungs are still immature and fragile and they cannot support breathing on their own. As such, the vast majority of babies are connected to a machine that artifi cially breathes for them. However, it is very diffi cult to determine the best time to disconnect these newborns from the machine with the tools currently available.

Our research group is interested in exploring the utility of analyzing biomedical signals such as heart and breathing rates to better predict whether these babies are ready to be disconnected from the machine. We aim to build a clinical database and cardiorespiratory signals from more than 200 major premature babies. The goal is to fi nd the best combination of clinical markers and cardiorespiratory measures that would more accurately predict whether the baby is ready to be disconnected from the machine or not.

Children with Borderline Intellectual Functioning (BIF) are children whose cognitive functioning falls below the average IQ. Yet, they do not qualify for an intellectual disability. These children often suffer academically, and they also have greater rates of diffi culties in emotional health and functioning. However, very little research has been conducted on effectively delivering interventions to these children. This project examines the perception of children with BIF of their own mental health and functioning, and how this perception might differ from that of their parents. It also examines whether self-awareness of diffi culties is associated with motivation to receive treatment intervention amongst children.

LEAH WRIGHT

Concordia UniversityMaster’s Student

Despite mounting evidence that stress during childhood predicts adult health outcomes, there is no existing systematic literature review and meta-analysis of studies on the association of stress and cortisol in childhood.

Leah’s plan is to elucidate the possible timing and manner through which stress becomes biologically embedded during childhood. This is an essential preliminary step in the establishment of salient developmental periods of vulnerability, and this line of research has great potential to inform interdisciplinary research fi elds, especially epigenetics.

ANNUAL REPORT 26

WE STRONGLY BELIEVE THAT CHILDRENWHO HAVE LIVED OR ARE LIVING THROUGH A DIFFICULT FIGHT AGAINST ILLNESS HAVE THE RIGHT TO FORESEE THEIR FUTURE WITHOUT HAVING TO CONTINUOUSLY WORRY ABOUT THEIR HEALTH.

Therefore, 70 million dollars have been collected to fi nance distinguished pediatrics researchers working on major children’s diseases research projects. Since 2014, the Board of Directors keeps on adjusting its strategic approaches to ensure the excellence of funding pediatric research.

Firstly, the Foundation of Stars maintains its partnership with the Fonds de recherche du Québec – Santé (FRQS) in order to choose the best research projects based on a scientifi c evaluation by Expert Commitees (from Québec, Canada and worldwide). Secondly, the Foundation of Stars continues to support the next generation of researchers in order to promote innovation and the development of promising, results-driven projects.

FINANCIAL Highlights

FUNDS DISTRIBUTION FOR PEDIATRIC RESEARCH

Since autumn 2014, and as shown in the table below, part of the funds collected during the years 2014-2015 and 2015-2016 were invested in collaboration with the FRQS. 15 students – whose projects are described in this Annual Report have received scholarships for highly promising research projects. Thus, since 2014, the Foundation of Stars and the FRQS have each committed to a maximum of $500,000 per year to co-fi nance training grants and career awards, representing an investment of more than a million dollars to advance research on childhood diseases. Other funds are also forecasted for 2017.

We are also developing other types of collaborations with reseachers from major academic institutions and pediatric hospitals in order to support clinical research that will directly affect the patients. These future collaborations will allow quicker results and will impact pediatric research. Therefore, we are continuing to focus on excellence to provide a better future for sick children.

We want to pursue our commitment of providing more substantial support to the best students and researchers and, by so doing, support the most innovative research projects.
