Abstract:

A ualitative risk assessent as carried out on 'R()PR'as*6 (t as !ound that

there ere several ha&ards hich could result in an unintended $enetic seuenceor novel $enetic co"ination and that the pro"a"ilities o! these occurrin$ ere

ver+ hi$h6 (t as also !ound that several iproveents associated ith the 'as*

technolo$+ ere capa"le o! controllin$/iti$atin$ these risks7 "ut the+ had not

+et pro$ressed to the point here 'as* as suita"le !or in vivo therapeutic

applications or idescale release to the environent6 (ntentional ha&ards that

Introduction:

'R()PR'as* is a relativel+ ne "reakthrou$h in "iotechnolo$+ that has led to a

sall la"s revolution via the ease o! $enoic anipulation6 (nitiall+ discovered

"+ (shino et al6 in 1*7 clustered regularly interspaced palindromic repeats 

:'R()PRs;7 ere descri"ed as a set o! 2*"p repeats interspaced "+ 32"p

seuences :spacers; located in the $enoe o! Escherichia Coli. The !ail+ o! 'as

:CRISPR-associated; proteins< !ound in over 40= o! >acteria seuences and

around *0= o! Archaea7 as descri"ed soon a!ter :,ojica et al6 2000;6 Throu$h

stud+ o! their putative !unction as endonucleases it as discovered that the+

ere capa"le o! actin$ as an innate adaptive !or o! >acterial iunit+ :,ojica

et al67 2005< Pourcel et al67 2005;7 and in 200 ,arra?ni et al6 discovered that the32nt @spacers direct 'as nuclease activit+ a$ainst .BA< con?rin$ a h+pothesis
