MILLIPLEX MAP Endocrine PanelsThe endocrine system is made up of small, ductless, and highly vascular organs

that secrete hormones, extracellular signaling molecules that regulate a variety

of functions, including growth and development, tissue function, metabolism

and even mood. Typical endocrine glands include the pituitary, thyroid and

adrenal glands. However, endocrine hormones secreted from the gut and

adipose tissue also play a critical role in regulating metabolism. Disruption of

hormone production, release or function, resulting in a hypo- or hyperfunction of

endocrine glands, is responsible for a variety of diseases: type 1 and 2 diabetes,

obesity, hypothyroidism, acromegaly and cancer.

Assay Conditions

• Amylin (active) and Amylin (total) cannot be plexed

together.

• This kit may be used for the analysis of either active

amylin or total amylin with any or all of the analytes in

the panel in serum, plasma, other body fluids, and

cell/tissue extract or culture samples.

• This is an overnight assay requiring 25 µL sample size.

• The assay requires a 1:3 dilution in serum or plasma

using assay buffer.

• Users will need to determine the appropriate dilution

when using cell or tissue culture supernatants.

MILLIPLEX MAP Human and Rat/Mouse IGF-1 Single PlexesInsulin-like Growth Factor (IGF-1) is a polypeptide endocrine

hormone similar in structure to insulin. Stimulated by growth

hormone and produced primarily in the liver, it binds to the

IGF-1 receptor with a high affinity and to the insulin receptor

with a much lower affinity. The tyrosine kinase IGF-1 receptor

is a potent activator of the Akt signaling pathway, which drives

a variety of processes including cell growth and division and

apoptosis inhibition. Because approximately 98% of IGF-1 is

always bound to one of six binding proteins (IGF-BP), extraction

may be required to measure extracellular protein concentrations.

Increased levels of IGF-1 have been correlated with an increased

risk of cancer. In addition, while involved in reducing hemoglobin

A1C levels and daily insulin consumption in types 1 and 2

diabetes, these increased levels have also been implicated in the

development of diabetic retinopathy. Drugs have been developed

as replacement therapy for severe primary IGF-1 deficiency, a

group of diseases in which patients do not respond to growth

hormone therapy.

MILLIPLEX MAP Pituitary Multiplex PanelsCalled the “master gland” because it controls many other endocrine glands, the

pituitary gland secretes a number of key hormones that play important roles

in the regulation of metabolism, growth, and reproduction. For example, these

hormones are responsible for the stimulation of: the adrenal gland (ACTH),

thyroid gland (TSH), ovaries and testes (FSH, LH) and breast milk production

(prolactin), as well as blood pressure and the amount of water excreted by

the kidneys. In turn, the pituitary gland is connected to and controlled by

the hypothalamus, which determines the amount of stimulation needed by the

pituitary’s target glands by detection of their hormone levels.

MILLIPLEX MAP Stress Hormone Multiplex PanelsStress is defined as the disruption of homeostasis through physical and/or

psychological stimuli (stressors). Physiologically the neuroendocrine system,

which includes the hypothalamus, pituitary and adrenal glands, plays a critical

role in response to stress. Activated by corticotropin-releasing hormone (CRH)

and arginine-vasopressin (AV), the pituitary gland releases ACTH which results

in the secretion of cortisol / corticosterone and other glucocorticoids from the

adrenal glands. These corticoids involve the whole body in the stress response

and are finally involved in the termination of the stress response by a negative

feedback loop to the hypothalamus.

MILLIPLEX MAP Thyroid Multiplex and Single Plex PanelsTriiodothyronine (T3) and thyroxine (T4) are key hormones secreted from the

thyroid gland with a wide-range of biological effects, including the metabolism

of carbohydrates, fatty acids, and proteins. In addition, these hormones play a

vital role in the development of the central nervous and skeletal systems. The

syntheses and release of T3 and T4 are regulated by Thyroid-Stimulating Hormone

(TSH), which is secreted from the anterior pituitary gland. Together, these thyroid

hormones play critical roles in the regulation of energy balance and mental and

physical development. According to some studies, the simultaneous measurement

of T3, T4 and TSH is of greater value than individual measurements, especially

when monitoring disease relapse or therapeutic response.

Luminex xMAP Technology

1 Luminex uses proprietary techniques to internally color-code microspheres with two fluorescent dyes. Through precise concentrations of these dyes, 100 distinctly colored bead sets can be created, each of which is coated with a specific capture antibody.

3 The microspheres are allowed to pass rapidly through a laser, which excites the internal dyes marking the microsphere set. A second laser excites PE, the fluorescent dye on the reporter molecule.

2 After an analyte from a test sample is captured by the bead, a biotinylated detection antibody is introduced. The reaction mixture is then incubated with Streptavidin PE conjugate, the reporter molecule, to complete the reaction on the surface of each microsphere.
