Guide to importing images from ImageJ/FIJI to Photoshop and Illustrator
1) Open image in FIJ and split stack into images

2) Change LUT to greys (NB: you are not interested in changing colour in PS you can keep colour LUT is you like)
3) Make sure images are RGB Colour
4) Save your Files as TIFFs
5) Open Photoshop (PS), then from file menu choose scripts>Load Files into Stack…
6) Choose Browse and locate your TIFF files you just savedand click Open and then OK.
7) This will load all of your images into separate layers
8) Make 3 new folders by clicking on
the folder icon 3 times
9) Drag each image into a folder Folder Icon
10) You can now ad as many adjustment layers as you like
to each image. Normally Levels and Hue/Saturation are the only ones you will
need.
NB: Adjustment layers are great as they provide non-
destructive editing abilities and allow you to quickly turn off
and on your changes.

10) Click on the adjustment layer and a Properties panel should appear. You can use
this to play and alter theimage as you want.
11) To colour an image click the colourize box, then move
Saturation to 100%And Lightness down to -30 to
-40%Move the colour slider to your
desired colour.
12) NB: currently you will only see the top layer image. To make a composite overlay image select each Folder Group
and then from the dropdown menu select Screen.Repeat for other Group/s
13) You should now have an image that looks like this
14) To copy all the layers as a single merged image to Adobe Illustrator, first select all (cmd+A)
14) Then choose Copy Merged from Edit menu (or shift+cmd+C).
You can then paste (cmd+V) this Image into your Illustrator File
15) Optional: If you want to copy each coloured or grey scaled layer individually, just hide the layers your not
interested in. Then repeat Step 14.
Only the green layer now visible
16) For Grey scale image, just hide the Hue/Saturation adjustment layer
