Intro to Micro…Scopes Lab 1/2/2017

Directions: You will need to get a stamp/initials from me to show you understand how to use and care for a microscope prior to the end of class. Everyone will need to get a stamp or you will not be able to utilize microscopes for any upcoming labs. DO NOT SKIP AROUND IN THIS PACKET. FAILURE TO GET SIGNATURES IN ORDER WILL LEAD TO A “0” ON THIS PACKET.

Part I: Setting up the Microscope

1. Read Care of the Microscope to learn how to properly care for a microscope.

Care of the Microscopes

· The microscopes are precision instruments that require proper care. Always carry the microscope with two hands. One on the arm of the microscope and the other on the base

· Keep the microscope away from the edge of the table; nothing should be on the lab table but the microscope and your lab sheets.

· Avoid tilting the microscope when using.

· The lenses of the microscope are quite costly so if they are dirty please contact me so I can give you proper lens paper to clean them, never use anything but lens paper.

· When you are finished always set the microscope back to SCAN power and make sure the stage is lowered all the way down.

· The Revolving Nosepiece has three powers:

· Scan: the lowest power

· Remember when you are looking for your object it should be on SCAN, once you find it then use the stage clips to hold your slide in place.

· Once you have your specimen in your FIELD OF VIEW you can then begin to change magnifications for better observations.

· You can also adjust the amount of light by using the microscope’s diaphragm

Parts of a Microscope

Once you have studied the diagram call me over so we can discuss how well you know the parts of the microscope

Now follow steps 1-3:

1. Place the printed out letter “e” on the slide with a cover slip over the top.

2. View the “e” under the different magnifications. Remember you cannot use the COARSE ADJUSTMENT knob when looking at the HIGH POWER magnifications.

a. Does it look the same as on the slide?

b. If so how? If not what’s different?

3. Now move the “e” to your left.

a. What does it look like in the microscope?

__________________________________________________________________________________________

b. Did it move the same direction?

__________________________________________________________________________________________

c. If not which direction did it move?

Now let’s prepare a wet mount

1. Using the eyedropper place ONE DROP of water on a clean slide

2. Hold the coverslip at a 45o angle with the bottom edge of the slide touching both the slide and the drop of water. Then very slowly lower the coverslip. (This will help to eliminate the chance of bubbles, which will make it hard to see your specimens)

Once you have been successful call me over so I can sign your sheet and you can move on

Now follow steps below:

1. Locate a specimen on SCAN power and use the STAGE CLIPS to hold it into place. Sketch your observation at 100X magnification (LOW) then sketch your specimen again at 400 X (HIGH) magnifications. Label and record any observations.

2. Repeat steps for at least two more specimens.

Turn your packet in by the end of class.
