Welcome to the 2015 Showhouse for a Cure. WBCS was founded in 1998 as an all-volunteer 501 (C) (3) charitable organization. With the dedication of thousands of volunteers and the generous support of the community, WBCS has raised over $5.45 million dollars for early-stage breast cancer and prostate cancer research at the Medical College of Wisconsin. This is an amazing feat, and a true testament to the spirit of the greater Mil-waukee community.

Five days before Christmas in 2002, Tacoma, Washington newspaper pub-lisher Elizabeth “Betsy” Brenner received unwelcome news—a diagnosis of breast cancer. Betsy recalls, “You could have knocked me over. The last thing I had time for was cancer. I was 48 years old, and I couldn’t remember the state I was living in when I got my last mammo-gram; much less, when it took place.” She celebrated New Year’s Eve 2003 recover-ing from a needle biopsy, followed three weeks later by a mastectomy, then che-motherapy and reconstructive surgery.
Now, fast forward to April 2013. Betsy, who had relocated to Milwaukee in 2005 as president and publisher of The Journal Sentinel, received more bad news. Her cancer, extensive ductal carcinoma in situ (DCIS), had recurred. She faced this challenge with mixed feel-ings. “A recurrence is clearly more serious and forces some ‘life deci-sions’ on patients and families. In another way, though, it wasn’t as daunting as the first time. I knew what to expect. I knew about the fatigue and the reaction to medication. I knew my hair would grow back. So, with some experience behind me, it was easier to face and get through it.”
Fortunately, the years intervening Betsy’s first and second diag-noses brought new research and treatment options. Now, she says, “The drug combinations that I’ve been given are all relatively new, seem to be extremely effective—and are all a result of research done and advanced thanks to investments like the type made by WBCS.”
Betsy’s career as a high profile newspaper executive means having to fit treatment into a busy work and travel sched-ule. Husband Steve Ostrofsky, a retired newspaper executive,
“… has been an amazing caregiver. He’s attended every doctor visit; been there for every treatment, and kept me going with good humor—even when nothing is funny. Friends and colleagues have all stepped up with encouragement and support. I’m very lucky to have had all of them on my side.”
In addition to serving on the As-sociated Press and Newspaper Associa-tion of America industry boards, Betsy makes time to give back to the commu-nity. She serves as a Medical College of Wisconsin trustee and is on the boards of Children’s Hospital of Wisconsin and Boys & Girls Clubs of Greater Milwau-kee. She is a past board member and president of mentoring organization ABCD and served as honorary chair of the 2014 local Susan G. Komen walk.
Betsy sums up her cancer journey by observing, “Nobody wishes a cancer diagnosis on anyone. It’s hard. It’s really scary, and with it you realize you’re mortal—per-haps for the first time. But it’s there. We learn to live with it and live through it. Contrary to the image of the ‘courageous cancer survivor’, I can’t say I’ve ever felt particularly brave about this. I’d much rather never have had cancer. But, I’m doing what I have to do because I want to live a quality life for as long as I can—and I’m fortunate to have an incredible team of family, doctors, friends and colleagues to support me as I do. That’s what I’d wish for anyone facing the same challenge.”

Research for a CureWBCS, Inc. invested $350,000 in research in
2014, with a cumulative sum of $5.45 million in seed funding. Over time, this has enabled researchers to leverage our support into an additional $43.7 million in competitive external research awards, a $6.1 million increase over 2013. Ming You, MD, PhD, MCW Can-cer Center Director, describes our all-volunteer organi-zation’s $5.45 million in research support since 1998 as having a “transformational” impact on breast cancer and prostate cancer.
Dr. You elaborates, “Contributions from WBCS are so powerful because the impact goes far beyond the initial gift. WBCS leadership has the insight to require that donations support only the best, most promising and most innovative cancer research. This type of in-vestment always comes back exponentially in the form of new federal funding dollars, high-impact publica-tions, and new ideas in cancer research.”
The Center uses WBCS awards as pilot grants to allow researchers to develop the substantive data needed to apply for National Institutes of Health (NIH) and other external and national funding. Austerity at the federal level for research con-tinues to weigh heavily against chances for even the most promis-ing approved research protocols to get funded. This troublesome phenomenon affects both young and seasoned investigators and can deter young scientists from pursuing a research career. Dr. You explains, “Without WBCS, many promising projects simply would not be a reality today. We are talking about research that has developed into the newest ideas in cancer prevention, detec-tion and treatment.”
The largest annual adult cancer research donor to the Center, WBCS leads the way with philanthropic support for early-stage breast cancer and prostate cancer research at the cellular level.
One of the most exciting developments for 2015 is the award of the WBCS Endowed Breast Cancer Research Professorship. The Chair was given to new recruit Hallgeir Rui, MD, PhD, who comes to MCW from Kimmel Cancer Center at Thomas Jeffer-son University. Dr. Rui has a strong history of successful breast cancer research, including prestigious publications, NCI and Ko-men Foundation funding, leadership of multidisciplinary teams
and mentoring of junior faculty, students and staff. His spouse, Marja Nevalainen, MD, PhD, also will be joining the MCW Can-cer Center. Her current research focus is prostate cancer, an area WBCS also supports.
In addition, two 2014 research projects were awarded sec-ond-year funding of a two-year, $100,000 grant: Weiguo Cui, PhD, “Harnessing Dual-Specific T Cells in Breast Cancer Im-munotherapy” and Vera Tarakanova, PhD, “Regulation of Innate Immune Responses by Breast Cancer Therapy.”
To ensure research we support is directed toward the most promising and competitive science, all two-year research ap-plications we fund are subject to peer scrutiny and review both initially and again before funding is continued for a second year. WBCS takes very seriously the importance of stewardship and accountability for the generous community and corporate sup-port entrusted to our mission.2015 Report to WBCS on the Large Center Grant
Carol Williams, PhD, has provided an update on the 2012 WBCS $100,000 Large Center Grant in Breast Cancer Research. This award supports a group of Cancer Center scientists and clinician scientists who are sharing innovative ideas and novel techniques to develop new ways to halt the growth of breast tu-mors and inhibit metastasis. WBCS support is allowing these researchers to gather the experimental data required to apply to the National Cancer Institute (NCI) for a Program Project Grant (PPG), as well as for individual grants to further expand their breast cancer research.
Dr. Williams reports there is an expansion of the WBCS-supported group of investigators who are working together to advance breast cancer research at MCW. Several of these in-vestigators are experts in advanced scientific techniques that are being used to develop better strategies to predict and treat breast cancer. Dr. Michael Flister, a new Assistant Professor in the Department of Physiology, is developing methods to identify

Research for a Cure continued
genetic risks that contribute to breast cancer development. Dr. Amit Joshi, a new Associate Professor in the Department of Radiology, is developing techniques to track drugs in the body to ensure that these drugs attack only breast cancer cells without hurting normal cells.
The addition of these researchers has strengthened the consortium of multi-disciplinary investigators working together to identify an interrelated, synergistic proposal to submit to the NCI for large institutional funding.WBCS Scientific Review Committee
The WBCS Scientific Review Committee (SRC) meets annually to review grant proposals and recommend approval and funding by the WBCS board of directors. Dr. Ravi Misra, PhD, Dean of the Graduate School, chairs the MCW peer scientist review committee that reviews and ranks all proposals before they are presented to the SRC for further evaluation and discussion. The purpose of the SRC, in partnership with the MCW Cancer Center, is to provide oversight and focus to the allocation of WBCS funds provided by the annual Showhouse for a Cure and its supporting events.

Last year nearly 40,000 breast cancer-associated deaths oc-curred, of which many are related to acquired drug resistance in recurrent tumors. Malignant tumors contain cancer cells as well as non-cancerous cells. These non-cancerous cells help to make up the tumor microenvironment (TMe). They are important in the development of breast cancer progression and metastasis, and the development of cancer resistance to chemotherapies. Current breast cancer research focuses on the cancerous breast tumor
cells alone, missing determinants of cancer risk in the TMe. we propose to examine the role of the TMe in breast cancer acquired resistance to chemotherapeutic drugs in triple negative breast cancer, which is breast cancer that lacks estrogen and progester-one receptors. This type of breast cancer is particularly aggressive and currently lacks targeted treatments. our goal is to improve pa-tient treatment and therapeutic outcomes for triple negative breast cancer by identifying new therapeutic targets in the TMe.
DNA encodes information, organized as genes, which can give rise to an rNA intermediate, which in turn gives rise to proteins. Prior to protein synthesis these rNA intermediates undergo a process known as rNA splicing that allows for the formation of multiple proteins from a single gene. It is well documented that rNA splicing of some genes can go awry in cancer. one such gene important for cancer is SmgGDS, which comes in two splice forms, one of which is primarily involved in breast cancer and is
a target for developing cancer therapies. however, nothing is known about what regulates SmgGDS splicing decisions in can-cer cells. we seek to gain a basic understanding of how the Smg-GDS splicing decision is made with the goal of uncovering novel “druggable” targets that will lead to therapies to reduce the level of the cancer-promoting SmgGDS splice form.
Prostate cancer initially occurs when cells in the prostate grow uncontrollably, from which it spreads to other organs, especially the bones. Statistically, it affects 1 in 6 American men. Initially, pros-tate cancer relies on androgen signals to sustain its growth, and providing effective therapy relies on androgen depletion. As pros-tate cancer progresses it can develop into a much more aggres-sive androgen independent form that is refractory to androgen depleting therapy. understanding the mechanism of this switch is
important for developing effective targeted therapies to prostate cancer. we have identified a kinase protein termed TPL-2 that ap-pears to be a regulator of the switch from androgen-dependent to independent forms of prostate cancer. This proposal will use a transgenic TPL-2 animal model recently developed by the ap-plicant to address the in vivo role of TPL-2 in invasiveness and metastasis of prostate cancer in androgen independent cancer to identify potential areas for future targeted therapies.

WBCS fulfilled its commitment to MCW to fund an endowed chair in 2014. Soon, a search committee was formed and recruitment begun to seek an outstanding candidate for the WBCS endowed chair. As a result of the search, not one, but two key scientists, Hallgeir Rui, MD, PhD and Marja Nevalainen, MD, PhD, will be joining the Medical College of
Wisconsin Cancer Center (MCWCC) on July 1, 2015.Dr. Rui, Professor and Vice Chair of Cancer Biology and
Scientific Director of the Jefferson Breast Care Center at the Kimmel Cancer Center at Thomas Jefferson University in Philadelphia, has been named the Wisconsin Breast Cancer Showhouse (WBCS) Endowed Chair in Basic Breast Cancer Re-search and Professor of Pathology at MCW.
Dr. Rui has a strong history of successful breast cancer research, leadership of multidisciplinary teams and mentoring of junior faculty, students and staff. He is the principal investigator of an NCI R01 and an NCI R21 and has made a series of contribu-tions to increase understanding of the molecular mechanisms of breast cancer. Dr. Rui leads an inter-institutional consortium of multidisciplinary investigators to classify malignant breast tumors based on the expression of druggable target proteins for improved personalized cancer care, an effort initiated by a $6.7 million, five-year Komen Promise Grant. Since 2006, Dr. Rui has directed the Pathology Translational Research Core Facility, which provides basic and advanced histotechnology and tissue procurement services for more than 50 principal investigators.
In his new position, Dr. Rui will build on existing MCW ex-pertise and research activities to assemble a core interdisciplinary
team of researchers who will conduct a wide spectrum of breast cancer research. Dr. Rui will serve as a WBCS spokesperson, participate in fund raising efforts, and build strong connections with the community and potential donors.
Additionally, Dr. Rui will serve as the MCWCC’s Associ-ate Director of Basic Science. Dr. Rui’s experience as a research program leader and a shared resource director at an NCI-desig-nated cancer center is expected to provide the ideal background for this new role. As Associate Director of Basic Science, Dr. Rui will be responsible for leadership of the basic science research pro-grams, Cancer Biology and Tumor Progression and Metastasis, and oversight of all Cancer Center shared resources.
Dr. Nevalainen, Associate Professor of Cancer Biology and Associate Director for Education and Training at the Kimmel Cancer Center at Thomas Jefferson University in Philadelphia, has been named to the new leadership position of MCWCC Associate Director of Education. She also will serve as Professor of Pathol-ogy at MCW. Dr. Nevalainen’s current research focus is prostate cancer, working to develop effective pharmacological therapies for advanced cases. She has been NCI funded since 2005 and is the principal investigator on an NCI R01 and R21. She has received multiple grants from the Department of Defense Prostate Can-cer Program and American Cancer Society, including a Research Scholar Grant. In addition to multiple awards and honors, in December 2014, Dr. Nevalainen was awarded the 2014 Sidney Kimmel Cancer Center Innovator of the Year Award. Prostate cancer is also a focus of WBCS in early-stage research support.
WBCS welcomes Dr. Rui as the WBCS Endowed Chair in Basic Breast Cancer Research. Breast cancer affects 1 in 8 individuals. Prostate cancer affects 1 in 6 men. We look forward to work-ing with both Dr. Rui and Dr. Nevalainen to support further advancements and ultimately cures for these devastating diseases.

Fighter Award in 2014 to recognize the tireless efforts of an indi-vidual or entity dedicated to ending breast cancer and prostate cancer. This award may be given annually in memory of past WBCS board member and founding Tee Up for a Cure chair Christine Hamilton Foote (March 12, 1950-February 5, 2014).
Christy fought cancer with grace, a quiet dignity, and always with hope. Despite the challenges she faced over many years, Christy continued to raise funds for research and work for cures right up to the end of her life. She will continue to be an inspira-tion to those fighting cancer and to those who are seeking cures.

In 2001, the last remaining single vacant lot in Shorewood was sold, and in 2004, a spectacular house was con-structed on it. Located at 3965 N. Lake Drive, the Tuscan-style house has been chosen as the 2015 Wisconsin Breast Cancer Showhouse Inc.’s 18th Anniver-sary Showhouse for a Cure.
Deep River Partners, Ltd., a Milwaukee architectural and interior design firm, designed the asymmetri-cal structure, combining the old world construction techniques of northern Italy with spectacular views of Lake Michigan, Atwater Park, and Shore-wood’s Plensa sculpture. The two-story Tuscan-style thirteen-room house features four bedrooms, three and one-half bathrooms, two fireplaces, a sunroom, and a lower level recreation space with a wine cellar within 5,762 square feet of living space.
The stucco façade is enhanced by a rusticated pattern of flag-shaped stone along the base recalling the look of Ital-ian farmhouses and villas. Contrast is provided by using cedar for the arched wood columns, the paired block roof brackets, and the balcony posts. Resting next to the window-framed quar-ter-sawn white oak arched entrance door is a 100 year-old antique iron pot from the Gan’su area of China. Of special interest are the small decorative squares which cleverly mimic a belt course found above the semicircular windows. Balconies with wrought-iron balustrades overlooking the garden as well as a front entrance
tower with clerestory windows complete the overall appearance of a timeless Tuscan Villa.
The front-facing terrace and gardens are surrounded by river birch trees, a dry-fit Lannon stone wall, and low shrubbery pro-viding privacy from the public zone. Emerald arborvitae which mimic northern Italy’s cypress trees, are strategically placed on the terrace to provide a sense of order and formality. The soothing water feature includes a 200-plus year-old hand-carved granite pot with a traditional Chinese floral pattern. The surrounding stone provides a balancing element to the landscape dominated by earthiness. A split-faced Lannon stone walkway leads to a dwarf Sargent crabapple tree hosting white spring flowers and red fall berries. The ancient statue at the entrance to the rear en-tryway is over 400 years old and is a Chinese symbol for good luck and protection. The symbol also represents the many facets of life.
The entrance door with its window surround and four carved teak corbels on the vestibule wall sets the stage for dramatic high-lights of the interior. Dominating the foyer is an intricately-carved teak water fountain from Bali, designed by the internationally-re-nowned tropical garden artist, Made Wijaya. Richly-grained wide wood planking draws one into the adjacent formal dining room. There your eye is drawn to the hand-crafted chandelier, repur-posed from an antique French fireplace grate and hanging from four decoratively painted Mediterranean tiles.
The state-of-art kitchen, with its large green-house window, blends the outdoors with the indoors by combining the warmth of a fireplace, the wood flooring, and custom-built cabinets with the alluring view of a private garden-oasis featuring one more sculp-ture created by Made Wijaya. The adjacent room’s two-main walls display ornately-carved Balinese teak wood panels.

Horizontal cedar hand-hewn beams polished with brie wax, Caribbean pine wide-planked hand-scrapped flooring, and arched windows and doors invite one into the wide gracious living room. The stunning views of the Tuscan landscaping and Lake Michi-gan on the east side of the room vie for attention with the elegant floor-to-ceiling Trumeau Louis XV style limestone fireplace on the west side of the room.
An adjoining sunroom provides a less formal space with its patterned terra cotta tile floor reclaimed from France. Pleasant views of the covered patio through a continuous band of arched windows and doors are among the room’s focal points.
A handsome curved wrought-iron staircase leads to a landing which is highlighted by a dramatic Indonesian chandelier carved of teak wood. Also, noteworthy in the landing area are the heavy antique copper doors, which were discovered in a salvage facility in Illinois, and hand polished to their present glowing patina.
On the upper level of the home, the master bedroom creates the serene atmosphere of an Italian villa with outstanding vistas of Lake Michigan and a tiled open-air private terrace. The distinc-tive doors leading into the dressing room are 200 year-old yu’mu wood courtyard doors found in Beijing. The lady’s antique stone-topped dressing table with the small Kalista sink is also made of yu’mu originating in China.
The adjoining master bathroom creates the open light atmo-sphere of a Roman bath with its soaring height, clerestory win-dows, decoratively tiled floor and walls, and a wood-paneled tray ceiling adding dramatic interest. A 100 year-old Chinese buffet has been repurposed into a wash basin.
The lower level serves as a recreational space which features a full bath, den and wine cellar.
Characteristic of Tuscan-style homes, the Showhouse is stra-tegically positioned on the lot to create an extension of the garden space and a connection to the sky, water and land beyond.
The 180-degree panorama of Lake Michigan from the house is enhanced by Jaume Plensa’s sculpture, “Spillover II”, resting on the bluff at Atwater Park.
Known and highly respected as a public space artist, the internationally- renowned Spanish sculptor’s piece was chosen by the Shorewood Public Art Committee for its transparency and statement. An anonymous donation to the Committee funded the purchase and the Shorewood Foundation covered incidental costs and continues to fund its long-term maintenance. “Spillover II” is eight feet tall and lit from the inside at night. It features a seated person hugging their knees and is constructed of stainless steel letters combined randomly as homage to the single letter creating words, ideas, text and culture. Metaphorically it is about society – a single person is nothing, but in collaboration with others become a society, a country, a universe all together as one. The sculpture was unveiled and installed on a cement base in September, 2010 with Jaume Plensa in attendance.
Atwater Park was established in 1910 by the city of East Milwaukee at the east end of what was then Atwater Road, now Capitol Drive. The name was changed to Shorewood in 1917.
It was as wide as the Atwater roadway with a wooden stair-case down the south end of the bluff. The Village of Shorewood purchased land for expansion to the north in 1929 and additional land one year later. Black locust trees were planted to stabilize the bluff and harp lamps were installed along switchback trails in 1932. As part of the 1938 Works Progress Administration (WPA) to employ Depression-era unemployed, jetties and a bathing pavilion were constructed on the beach. Using one-half million dollars of Federal money, the pavilion boasted of a roof-top deck for daytime picnicking and nighttime dancing.
In 1967, two cable cars were installed from the park to the beach. The Village initiated a contest to name the cars. Able Cable and Twinkle Tow were the winning names. The cost for a one-way trip was ten cents, but the cars were inoperable more often than operational. No one in the Milwaukee area knew the complicated mechanics of the German-made cars and the Village removed them in 1977. Ten years later, the beyond-repair bathing pavilion was demolished and new steps to the beach were built. A bronze, iron, copper and steel fountain was installed in the park in 1969. It remained until the Plensa sculpture was erected and is now in storage.
Intriguing playground equipment designed for both kids and adults and an expanded pathway to the beach have enhanced the park and beach recently. Repair of the jetties and a contemplative area are long-range plans for the park and beach.
Visitors to the Wisconsin Breast Cancer’s 18th Anniversary Showhouse will be awed by the architectural beauty of this Tuscan-style designed house, terrace, and the museum-quality artifacts displayed inside and outside.
