LABORATORIUM VOOR KLINISCHE DIAGNOSTIEK N.V.

Preface

Dear Colleagues, You are now holding our price list for July 2019 to June 2020. As always, it is full of innovations and helpful for clinical practice.

Our services: We believe we have an impressive port-folio. Laboklin has been working in the field of vete-rinary laboratory medicine for 30 years, and we have remained in touch with our field. What test is best, which parameters are most useful in clinical practice? These are the questions that guide us in compiling our portfolio.

Microbiology: We have been working according to the CLSI/EUCAST guidelines for many years, species specific and with profiles that focus on specific types of clinical changes.Clinical chemistry: Efficient profiles, practice specific profiles, many parameters that are difficult to find else-where, e.g. erythropoietin.Immunology: We use modern technologies such as flow cytometry to characterise tumours.Pathology: State of the art with information on margins for tumours, grading of various tumours, immunohistology, tissue profiles – all from an expe-rienced team and at high speed.Genetics: Consistently up to date portfolio and breed specific profiles; collaborations with universities ensure that interpretations are science based.

Our business terms: We can invoice the practice on a monthly basis. If the invoice is sent to the practice, we offer a discount depending on the revenue volume. We are able to invoice the owner for genetic testing, if they have signed the submission form and we have received

a valid address - no worries for your practice! (Owners are invoiced with a factor 1.4 plus 21% Dutch VAT) We continue to offer free sampling and shipping materials. Our courier service for sample pick-up and transport throughout most regions in Europe makes working with us easy. Results can be ob-tained online via our website or our app, send via email or fax. The data can be imported into practice software programs. We are also available by phone: Monday to Friday 08:00-19:00, Saturday 09:00-13:00.

What makes us special? We are still independent – a laboratory from veterinarians for our colleagues in clinical practice, without connections to investor groups, chains, or other ties. Our focus is not maxi-mizing profit, it is optimizing quality. Research is part of our daily work – proven by our many conference presentations and publications.

Quality: The quality you have a right to expect. At LABOKLIN, we have been meeting this expectation for many years with our consistent accreditation according to DIN EN ISO/IEC 17025:2005. This means that all of the tests and procedures we do are described in detail, carried out with internal controls, and evaluated by the experts of the German accreditation body (DAkkS). Our employees take part in regular training courses in order to fulfil the demands we make on ourselves.

Best regards from the laboratory, Dr Elisabeth MüllerCEO LABOKLIN

LABORATORIUM VOOR KLINISCHE DIAGNOSTIEK N.V.

Dysbiosis Profile• Diarrhoea, flatulence, chronic inflammatory bowl

diseases, eczema, and food intolerances are often associated with changes in the intestinal flora.

• Evaluation of the intestinal microbiome offers new options for classification. The parasitological and mycological tests included as well as the detec-tion of the inflammatory markers calprotectin and alpha-1 antitrypsin help to define possible causes of the clinical signs.

Serotonin levels• In dogs, behavioural problems like hyperaggression

or increased anxiety can be caused by reduced serotonin levels.

• Measuring serotonin supports the diagnosis and treatment evaluation in dogs with behavioural issues and offers additional treatment options in the form of food supplements or drugs.

Food allergens• The new allergen profile „Food Allergens Exotic“

offers analysis of IgE antibodies against 15 addi-tional allergens for dogs and cats. It also includes allergens that are new in animal food production such as insect proteins.

• The use of innovative microarray technology means that only a small sample volume is needed for many allergy profiles, making it possible to pre-pare full profiles even from small volume samples.

BRAF in dogs• An oncogenic mutation of the BRAF gene

V595E has been reported in the literature, and by LABOKLIN studies, to be present in most of canine bladder and prostate carcinomas.

• LABOKLIN offers molecular detection of this mutation in the following samples: urine sediment, cytological smears, and formalin fixed biopsies.

Differentiation of neuropathogenic EHV-1 variants• Two different EHV-1 variants have been

described in horses. They are differentiated by a nucleotide exchange in the DNA polymerase gene and are associated with different levels of neuropathogenicity.

• LABOKLIN differentiates the two variants in all EHV-1 positive cases by PCR automatically and at no additional cost.

Quantitative PCR• In many cases, evaluating the number of patho-

gens present by PCR is relevant for treatment and prognosis. We therefore offer a quantitative PCR for the following pathogens:

Quantitative PCR Species Sample

Leishmania dog EDTA blood/bone marrow

Filaria dog EDTA blood

Parvovirus dog Faeces

Distemper virus dog Conjunctival swab

Coronavirus cat Faeces

Mycobacterium avium complex (MAC) sensitivity in Miniature Schnauzers• This test determines a genetic predisposition

and therefore increased susceptibility to MAC (Mycobacterium avium complex) in Miniature Schnauzers. LABOKLIN is the only laboratory that uses the protocols shared by the research group.

Genetic blood group determination in cats• Until recently, blood group determination in

Ragdolls, Turkish Angoras, and Siberians was limited. Based on our own in-house research, we now have a test method that clears up previous inconsistencies between serology and genetics. Our test offers the highest possible reliability for all cat breeds.

LABORATORIUM VOOR KLINISCHE DIAGNOSTIEK N.V.

Preanalytics Blood, plasma and serum samplesThe expiration dates of sample tubes should be checked regularly. Tubes with expired dates should no longer be used. It is important to add the correct amount of sample material to the tubes when collecting your sample. Blood should be allowed to flow slowly down the side of the tube and the sample should be shaken gently immediately after collection. Blood collected for serum or plasma should be centrifuged in a timely manner and the plasma or serum should be pipetted into a separate tube. Blood smears should be made directly after blood collection and kept and sent at room temperature.

The following parameters are sensitive to how the sample is collected, stored and transported:• Serum (Li-heparin plasma to some degree):

AP, heat stable AP, ß-HBS, calcium, chloride,CRP, Cu, Fe, fructosamine, bile acids, K, lipase,Mg, Mn, Na, NEFA, phenobarbital, anorganicphosphate, Se, T4, TSH, Zn

• Only serum: Aspergillus Ab, progesterone, Yersi-nia Ab, ionised calcium (note air-tight packaging)

• Serum, cooled: Insulin, erythropoietin, IGF-1,relaxin, LH, PTH, AMH, serotonin, troponin I,inhibine B, CPSE, vitamin A, D3, E

• Serum, frozen: Cold chain must not be broken:fT4 quantification with equilibrium dialysis

• EDTA plasma, frozen:ACTH, PTH-rP, aldosterone, BNP

• EDTA blood: Blood count, lead, blood group,Coombs test, cyclosporine, glutathione peroxida-se, immune status, Knott test, leukaemia profile, leukaemia classification, heavy metal screening,PARR, thrombocyte Ab

• EDTA blood frozen: vitamin B1, B2, B6• Citrate plasma:

PT, PTT, TT, D-dimer, von von Willebrand Ag(cooled)

• Citrate blood: Thromboelastography• NaF sample: Glucose, lactate• Urine: Admixture of blood changes the results of

the U-P/C; the cortisol-creatinine ratio cannot beevaluated in samples that contain blood.

Medication: Many drugs influence the concentrations of clinical chemistry, endocrinological, serological, and haema-tological parameters. Corticosteroids: Cortisol, all antibody detections, T4, fT4, T3, AP, PTH, CRP, SAA, blood count.

Histology• Skin: Submit a sufficient number of representa-

tive samples. Avoid collection artefacts: Electro-coagulation, ripping, squashing, autolysis adver-sely affect the analysis. Ideal sample size: 0.5-1.0cm, punch biopsies skin: 0.6 cm

• Tumours: The evaluation of the margins requirese.g. submission of all of the tissue (possibly incl.tumour bed biopsies). Formalin fixed 10%.Clinical history needed (fill out pathology sub-mission form). Results usually in three workingdays(Monday-Friday). Exception: unfixed samp-les, samples that have to be decalcified, cases inwhich special stains are necessary.

CytologySmears following air drying for evaluation of cells, especially tumour cells and inflammatory cells. Staining: Diff-Quick®, unstained samples can also be submitted. Special stains (e.g. for fungi, mycobacteria, copper) available. Results usually on the same day.

1: Formalin container 2: Serum tube 3: EDTA tube

4: Citrate tube 5: NaFB tube

Polymerase chain reaction (PCR) Pathogens that cause viraemia, parasitaemia, or bacteraemia can be directly detected in EDTA blood during this phase. At least 0.2 ml is necessary. Swabs should be submitted without medium (excep-tion: CEM profiles, which requires swabs with Amies transport medium). Pathogen detection in faeces requires a sample amount about the size of a hazel-nut. As some pathogens (e.g. coronavirus, Tritricho-monas foetus) are only shed intermittently, we recom-mend collecting faeces over 3 days. Additional sample materials, e.g. tissues, urine, and punctate should be submitted in sterile, uncoated sample tubes. Fixatives like formalin are not appropriate. The samples do not generally need to be cooled. The material can be stored in the refrigerator at 2-8 °C until it is submitted. Repeated freeze/thaw cycles should be avoided.

LABORATORIUM VOOR KLINISCHE DIAGNOSTIEK N.V.

Genetic TestingEDTA blood (about 1 ml) can be used for molecular detection of hereditary diseases, parentage analy-sis, or for the genetic dermination of coat colour or blood group. Alternatively, swabs of the oral mucosa, so-called buccal swabs, can be used in dogs and cats. Two swabs without transport medium should be submitted per animal. For DNA profiling or parentage analysis, we always recommend submitting a blood sample. In horses, about 20 hair roots from the mane or tail can be used for DNA preparation.

Microbiological / parasitological testingIt is important to label all samples and to include the animal species and sample location, since it may be necessary to use additional special plates in some cases.

• Swabs for bacteriological testing (aerobic and anaerobic) should be submitted in transport medium. Samples from localisations such as abscesses should be tested for anaerobes as well as aerobes. Some bacteria (e.g. chlamydia, mycoplasma) are detected by PCR. For these, a swab without transport medium should be submitted. Mycological testing can be done from samples with and without transport medium.

• Fluids can be submitted in sterile tubes or aswab of the fluid can be taken. Whether a swabwith or without medium is used depends on whattesting should be done. A uricult can be used forurine.

• Blood should be submitted in blood cultureflasks. Sampling should be done while the animalhas a fever, 2-3 flasks at different time points ifpossible. Use one aerobe and one anaerobe flaskwith bouillon.

• Hair & crusts should be collected from the edgeof the lesion, with roots. The samples should besubmitted in clean paper bags or tubes. Testingfor ectoparasites, bacterial, or fungal pathogens,or PCRs are possible.

• Faeces should be submitted in faecal tubes.Faecal swabs can only be used for bacteriology, mycology, or PCR. Various other faecal testing,e.g. parasitology, requires faeces, not just a faecalswab.

The following tests are time critical:< 48 h after sampling: blood count, FACS, immune status, thromboelastography, urinalysis incl. sediment< 72 h after sampling: thrombocyte antibodies

Sample storage time:EDTA and citrate blood and urine are stored for 7 days. Serum and plasma samples are stored frozen for 14 days, swabs for 14 days, faecal samples for 7 days, DNA samples (genetic tests) for 5 years.
