FOREWORD

Second International Seminar on Pharmaceutical Sciences and Technology 2016 (2nd ISPST 2016) is a continuation from first event on 2014. As one of the international event, the scientific content of 2nd ISPST2016 is truly multi-disciplinary: it covers all aspects of pharmaceutical sciences and technology starts from pharmaceutical technology, pharmaceutical analysis, pharmacology, food analysis, and natural product. Through sharing and networking, 2nd ISPST 2016 will provide an opportunity for researchers, practitioners and educators to exchange research evidence, practical experiences and innovative ideas on issues related to pharmaceuticals sciences. We realize that this event is only a small effort to contribute exchanges in researches and experiences, therefore more collaborative works and continuous relationship between researchers are still needed. 2nd ISPST2016 has received 186 abstracts addressing different scientific topics related to pharmaceutical sciences and technology. The scientific program is composed of 5 keynote lecturers, 5 plenary lectures from 10 lecturer, and 186 oral and poster presentations. The rich and diverse scientific contents represent many exciting achievements and also point to new challenges. On behalf of the organizing committee, I would like to extend a warm welcome to all delegates to Universitas Padjadjaran, Jatinangor, West Java, Indonesia. The organizers thank Universitas Padjadjaran, PT Biofarma, PT Cendo Pharmaceutical Industry, PT Horiba, and PT Tannach, PT Kimia Farma and off course Faculty of Pharmacy for generous support to this international conference. It is a great pleasure that The Second International Seminar On Pharmaceutical Science And Technology 2016 (2nd ISPST 2016) organized by Department of Pharmaceutical Analysis and Medicinal Chemistry and Department of Pharmaceutical and Formulation Technology,Faculty of Pharmacy, Universitas Padjadjaran being held on November 24th 2016. This seminar is a media for researchers, students, industries, and practitioners to share ideas and latest information on Pharmaceutical sciences and technologies. The second ISPST will focused on advances material design, process and technology for improving pharmaceutical product quality. This seminar will serve as a venue for researcher, professional, and students which have interest in the area of pharmaceutical sciences and its related fields to build many fruitful collaboration for their own research project and will also enrich collaborations activity in education, research and community service of Faculty of Pharmacy Universitas Padjadjaran. Honorable Head of the National Agency of Drug and Food Control of Indonesia or the representative, Dean of Faculty of Pharmacy Universitas Padjadjaran Distinguished guests, invited speakers, and all participants of the Second International Seminar on Pharmaceutical Science and Technology 2016. First of all, please allow me to extend my warmest welcome to you at the 2nd ISPST 2016 this day here at our Jatinangor campus. We are very proud to present this great event that facilitates findings in all pharmaceutical areas, including basic and applied sciences related to it. Pharmaceutical science is a wide discipline discussing both analytical and manufacturing aspects, and has been rapidly growing since the last decades. Novel findings in good manufacturing process to meet patient’s compliance are needed to solve the growth of new diseases. Based on the above, Faculty of pharmacy of Universitas Padjadjaran organize this seminar. The seminar is a continuation of the first International Seminar on Pharmaceutical Sciences and Technology (ISPST) which had been held in 2014. This year, the seminar has a theme Advances Material Design, Process and Technology for Improving Pharmaceutical Products Quality. Dr. Ir. Penny Kusumastuti Lukito, MCP is a Head Of National Agency Of Drug And Food Control (NADFC) Republic of Indonesia start from 2016. She was born at Jakarta on November 9th 1963. She was gained her Ph.D on Environmental Technique and Minor in Urban and Regional Planning at University of Wisconsin-Madison, Wisconsin, USA, on June 2000. She was received her master degree in City Planning (MCP) at Massachusetts Institute of Techonology (MIT), Cambridge, Massachusetts, USA.

Roland Bodmeier is a Professor of Pharmaceutical Technology at the College of Pharmacy at the Freie Universität Berlin in Germany. He studied Pharmacy at the Ludwig-Maximilians Universität München, Germany, (1977-1982) received a Ph.D. in Pharmaceutics at the University of Texas at Austin (1986) and a Dr. rer. nat. habil from the University of Regensburg (1993). He served as an Assistant/ tenured Associate Professor at the College of Pharmacy at the University of Texas at Austin (1986-1994) before he moved to his current position. He was a visiting professor at the Upjohn Company (1991) and at the University Henri Poincare in Nancy, France (1994, 1996). His research focuses on innovative drug delivery systems with controlled drug release (peroral systems, biodegradable implants/microparticles, microencapsulation, multiparticulate dosage forms) and has resulted in more than 170 publications, book chapters and patents/-applications.

NANOCARRIERS FOR DERMAL DRUG DELIVERY

ABSTRACT The successful treatment of diseases requires that the drug is transported efficiently to the site, where it is required for treatment. Mostly systemic drug delivery is used, however, for various diseases, local delivery is an alternative to systemic delivery because of a localized drug effect with minimized side effects. Local delivery routes includes dermal delivery and classically, this has been achieved with traditional dosage forms like ointments, creams, gels, etc. Advanced drug delivery systems for the skin include nanocarriers for delivery into the upper skin and hair follicles. This presentation will discuss the preparation and in vitro characterization of various nanocarrier systems. The nanocarriers include nanocrystals prepared by nanomilling, lipid nanoparticles prepared by high pressure homogenization and polymer nanoparticles prepared by solvent evaporation method. The model drugs include dexamethasone and tacrolimus. The carriers are investigated as a function of various formulation and process variables and are characterized with respect to drug loading, particle size, physical state of drug, drug release, cell culture toxicity and with various in vitro skin models. Kyung Teak. Oh, PhD is an Assistant Professor at Nanobiopharmaceutics Lab, Chung-Ang University, College of Pharmacy, Seoul, Korea from 2008 until now. He was received his Ph.D. from Department of Pharmaceutical Sciences, University Nebraska Medical Center, Omaha, NE, USA (2006). His research focuses on preformulation, formulation and preclinical study for new medicine candidates and incrementally modified drugs; development of synthetic drug, imaging agnet, and gene delivery targeting strategies for cancer therapy; development of nansized carriers for small and macromolecules using polymers ; and development of polymeric formulations for anticancer drugs and proteins with triggering release mechanisms.

DEVELOPMENT OF POLYMERIC PLATFORMS FOR NANOMEDICINES

ABSTRACT

Many molecules were discovered and developed for therapeutic agents. However, their

potential is severely compromised by the unfavorable properties of molecules and significant

delivery and targeting obstacles that prevail in vivo. Therefore, effective drug delivery systems to

improve the physicochemical properties and to deliver drug to targeting sites are required for the

successful development of new developed drugs.

Amongst versatile drug carriers, polymeric platforms were developed for pharmaceutical

applications with various advantages. We have developed a nanomedicine based on several block

copolymers to escalate therapeutic efficacy of anticancer agent by enhanced targeting to cancers.

The synthesized copolymers were used to deliver the anticancer agents, genes, and imaging agents.

Furthermore, the polymeric nanoplatforms showed several favorable properties in terms of particle

size, narrow distribution, high drug loading capacity, etc.

In this seminar, we will discuss about incrementally modified drug, nanomedicines,

polymeric based drug vehicles, and preclinic study using a nanomedicine developed in our lab,

Nanobiopharmaceutics lab.

Yasushi Arano, Ph.D is a Professor in Department of Moleculer Imaging and Radiotherapy, Chiba University, Chiba, Japan. He received M.S and Ph.D in Pharmaceutical Sciences at Kyoto University, 1973-1977. He had work experience as Associate Professor in Kyoto University, 1996-1999; as a Research Associate/Assistant Professor in Kyoto University, 1982-1996, Research Fellow in Harvard Medical School, 1988-1989. He has over 100 publications from Scopus. He has expertise and research interest in Radiochemistry, Radiopharmaceuticals, and Molecular Imaging.

MOLECULAR DESIGN OF RADIOLABELED ANTIBODY FRAGMENTS FOR MOLECULAR IMAGING AND TARGETED RADIONUCLIDE THERAPY OF TUMOR

ABSTRACT

Yttrium-90 and iodine-131-labeled monoclonal antibodies against CD20 have shown significant

efficacies for the treatment of non-Hodgikin’s lymphoma patients. In contrast to hematologic malignancies, however, treatment of solid tumors with radiolabeled antibodies has been found ineffective, since antigens are well protected by poor vascular supply, elevated interstitial pressure and a relatively long transport distances in the tumor interstitium. These factors are amplified as the tumor growth. Therefore, radiolabeled antibodies should be optimized to enhance the tumor uptake and retention in the solid tumor while minimizing the exposure of non-target tissues.

Antibody size affects the rates of antibody clearance from circulation and penetration into tumors. Radiolabeled intact antibodies eliminate slowly from circulation, which causes increased radiation dose to bone marrow and dose-limiting hematologic toxicity. Intact antibodies also exhibit limited diffusion distance from blood vessels, which limits penetration into tumors. Since the unfavorable pharmacokinetics of radiolabeled intact antibodies is caused by the presence of effector regions that are not required for specific binding with antigens, antibody constructs such as Fab and single chain Fv fragments have been used as vehicles to deliver radiation to tumors. Although promising results have been documented with radiolabeled antibody constructs in preclinical and clinical studies, the use of the antibody constructs raises questions about their potential renal toxicity, especially when they are labeled with metallic radionuclides. Thus, radiolabeled antibody constructs would become much more useful for imaging and treatment of solid tumors if the undesirable renal radioactivity could be reduced without impairing the radioactivity levels in tumors.

In this paper, our recent results to reduce the renal radioactivity levels of radiolabeled antibody fragments using enzyme-cleavable linkages will be presented. Prof. Habibah A Wahab (School of Pharmaceutical Science, University Sains Malaysia). A graduate in BSc in Science and Practice of Pharmacy from Liverpool John Moore University, Habibah A Wahab obtained her PhD in Pharmaceutical Technology from King’s College London, University of London in 1999. In 1999, Habibah joined Universiti Sains Malaysia, as a lecturer at the School of Pharmaceutical Sciences where she found a research group “Pharmaceutical Design and Simulation (PhD)” which focuses on research on drug discovery especially those utilizing structural bioinformatics and computer aided drug design approaches. In 2010 Habibah was promoted as a full professor in the School of Pharmaceutical Sciences, Universiti Sains Malaysia.

DESIGN & DISCOVERY OF POTENTIAL ANTI-DENGUE COMPOUNDS TARGETING THE ENZYME NS2/NS3 PROTEASE

ABSTRACT

The global incidence of dengue increases dramatically in these few decades. Many studies have been attempted to overcome this mosquito-borne disease, however, there is neither vaccine nor drug approved by Food Drug Administration (FDA) to treat dengue patients. In this talk, I would like to share some of our work in the design and discovery of potential anti-dengue compounds targeting the enzyme NS2B/NS3 protease, one of the major protein targets in dengue drug discovery. Using our previous virtual screening on National Cancer (NCI) database, we modified a hit compound bearing thioguanine scaffold into a series of 35 compounds for synthesis and bioactivity evaluation. One compound, MH018, demonstrated high inhibition against protease and DV2 virus with IC50 of 28

M and 11 M, respectively, at non-toxic concentration. In another related work, we undertook design of potential NS2B/NS3 protease inhibitor based on the virtual screening results of Natural product Discovery System (NADI) as well peptides based on the protease natural substrates. The design utilizes pharmacophore modeling, molecular docking and molecular dynamics simulation, which results will be shared in this communication.

SENSORS FOR MONITORING OF PHARMACEUTICAL PRODUCT QUALITY

ABSTRACT

This paper deals with the application of optical sensors as a versatile tool for monitoring of

pharmaceutical products quality. As product quality monitoring devices, the sensor could be employed for monitoring of the quality of raw material, processed and the product itself, as well as during their transportastion, distribution and storage. Here, two approaches could be applied, i.e. “reagentless approach”, where the infra red based optical sensor is used, and the “reagent approach”, where they consist of the integration of reagent or bioreagent into optical sensor system. The regentless approach is based on infra red spectra (NIR & MIR) and analysis using chemometrics to monitor and distinguish the quality of pharmaceutical products, starting from raw material untill the product distribution and storage as on-line monitoring.

The reagent approach usually called chemical sensor and biosensor that can be used for monitoring of pharmaceutical products. The current progress is rely on the use of immobilized reagent or bioreagent on paper or plastic for low-cost optical (bio)chemical sensors. The paper based sensor has been developed as platform for antioxidant and polyphenol sensors for herbal products qaulity. Others have been used for monitoring of amoxicillin quality in syrup, ethanol content in syrup and cosmetics. Thus, the optical sensor system is small, disposable, easy to use and suitable for quality monitoring applications of pharmaceutical products. This kind of sensor system is attractive and user friendly device for use in quality monitoring purposes, in the field, or as a low-cost alternative to more-advanced technologies already used in pharmaceutical product quality monitoring.

ABSTRACT

Molecularly imprinted polymer (MIP) gives amazing selective molecular recognition ability and can be as good as natural enzymes and antibodies. One of the most frequently used applications of MIPs is a selective separation and detection of pharmaceutical compounds in biological for therapeutic drug monitoring. MIPs based on solid phase extraction has very good capacities on capture selectively and concentrate of the targeting compound from real samples. The research will be covered a few works on the development of molecularly imprinted polymer based on solid phase extraction in selective recognition of pharmaceutical compound in biological samples. The methods we are using consist of three main stages which are measuring monomer-template interaction in solution, polymer synthesis, and evaluation of the polymer performances. Based on the results on a few compounds (sulfonylurea, ibuprofen, thyroxine hormones), we can conclude that molecularly imprinted polymer has great performances on a selective capture of a compound from biological interest. ABSTRACT Purple sweet potato is potential to be developed as an anti-diabetic drug because it can lower fasting blood sugar levels, triacylglycerol, and cholesterol in patients with diabetes type 2. The present study aimed to develope a standardized purple sweet potato dried extract and tablets containing the dried extract. In this work, the dry extract was obtained by adding various adsorbent into an ethanolic extract and then the dried extract of sweet potato was evaluated for its anthocyanin, total flavonoid content, solubility, particle size, the percentage of fines, flowability, compressibility and moisture content. Tablets containing the dried extract were tested for their average weight, friability, hardness, disintegration time and dissolution. The total flavonoid and anthocyanin contents in the dried extract were 0.46 and 0.20 % w/w, respectively. The dried extract presented consistent physicochemical properties and allowed the tableting process by direct compression. Tablet properties were fulfilled the Indonesian Farmacope specifications. The procedures described in this study is useful to prepare sweet potato dried extract with good properties for the development of a high-quality herbal medicine.
