1. Contamination Contamination of RNA samples by reagents, enzymes, or genomic DNA is the most common frustration experienced by researchers working with RNA in their workflow.

• Ethanol carry-over contamination as a result of poor column design is also a common source of contamination which can disrupt reverse transcriptase during RT-PCR and cause errors in absorbency readings.

• Genomic DNA contaminants are particularly frustrating as they can lead to errors in quantification in real-time PCR and disrupt the construction of cDNA libraries.

2. Elimination of Carry-Over Contaminants The NB RNA Miniprep Kit utilizes an unique flat bottom design on our RNA Spin Column which eliminates the creation of a 'dead corner,' or area of a spin column which traps ethanol during centrifugation. This design is unique to NB and greatly reduces the chance of carry-over ethanol contamination which negatively effects many sensitive downstream applications such as RT-PCR. 3. Removal of Genomic DNA Contaminants Pre-Clean Columns, included in our NB RNA MIniprep Kit, provide an efficient means to remove genomic DNA contaminants and other large particles from a RNA sample with a quick, 2 minute spin on a tabletop centrifuge. Many RNA kits on the market will not provide a column for this purpose relying solely on in-column digestion for genomic DNA removal. 4. Trizol Extraction Vs. Spin Column Purification One of the most common methods of RNA isolation is the use Trizol. While certainly effective, this method also has several drawbacks. It requires several steps of tedious extractions, and requires the use of toxic chemicals which are hazardous to health. The extracted RNA samples may not be consistent in purity when extracted from different sources and thus, may not be suitable for comparative analysis. In contrast, the advantage of using Spin Column Purification procedure is its convenience. Clean, high-quality total RNA is just a few spins on a tabletop centrifuge away. It does not use highly hazardous compounds. RNA samples with consistent purity can be obtained from a variety of sample sources, which are suitable for quantitative analysis. In the end, the choice of method for RNA Isolation lies with the researcher and his/her downstream applications.
