Electrical Hazards

The Agilent SPS 4 contains electrical circuits, devices, and components operating at dangerous voltages. Contact with these circuits, devices and components can cause death, serious injury, or painful electrical shock.

To reduce the risks of electrical shock, this equipment employs a three- wire electrical cord and plug to ground the equipment. To use this safety feature:

• Ensure the matching wall outlet receptacle is properly wired and grounded correctly.

• Never use a three- wire to two- wire plug adapter.

• Never use a two wire extension cord or a non- grounded multiple outlet receptacle strip.

• Do not use any power cord other than that supplied with this equipment.

• The power cord set that is shipped with this equipment cannot be used with other products.

• Position the autosampler so that it is easy to access the mains power on the wall outlet or to remove the power cord from the autosampler or wall outlet.

• Only Agilent- trained, Agilent- qualified, or Agilent- approved customer support representatives may open panels or covers that are secured by screws. Removal of such panels or covers can expose parts that involve the risk of electric shock or personal injury.

This equipment is rated Equipment Class I.

NOTE

Application of the wrong supply voltage can create a fire hazard and a potentially serious shock hazard, and can seriously damage the autosampler.

Do not connect the autosampler to the mains power supply until all checks and settings have been completed.

Replace or repair faulty or frayed insulation on power cords.

The autosampler must always be connected to protective earth through the power

WARNINGcord. Position the autosampler so that it is easy to access the mains power switch on the wall outlet or to remove the power cord from the autosampler or wall outlet.

Observe the following precautions when using your autosampler in conjunction with flammable liquids.

To reduce the risk of fire:

• Ensure the following autosampler parts do not contact the surface of the fluid or have parts near the surface of the fluid:

• parts that consume power

• parts that produce heat

• parts that conduct heat from other areas in any form

• Ensure no parts in or near the fluid surface are capable of igniting a flammable liquid.

• Operators must adhere to standard laboratory practices for the safe handling of flammable liquids.

Other Precautions

Use of the autosampler in any manner other than that specified by Agilent may impair the protection provided by the equipment.

Use of the autosampler may involve materials, solvents and solutions that are flammable, corrosive, toxic or otherwise hazardous.

Careless, improper, or unskilled use of materials, solvents and solutions can create explosions, fire, toxicity and other hazards that can result in death, serious personal injury and damage to equipment and property.

Always ensure that laboratory safety practices governing the use, handling and disposal of such materials are strictly observed. These safety practices should include the wearing of appropriate safety clothing and safety glasses.

Components of the autosampler move automatically and at speed.

Keep clear of the probe arm assembly during operation to prevent injury.

A protective cover is available to prevent accidental contact with the probe arm assembly.

CE Compliance

Your autosampler has been designed to comply with the requirements of the Electromagnetic Compatibility (EMC) Directive and the Machinery Directive (commonly referred to as the MD) of the European Union. Agilent has confirmed that each product complies with the relevant Directives by testing a prototype against the prescribed EN (European Norm) standards.

Proof that a product complies with these directives is indicated by the CE symbol on the rear of the product, and the documentation that accompanies the product containing a copy of the Declaration of Conformity. The Declaration of Conformity (DoC) is the legal declaration by Agilent that the product complies with the directives listed above, and shows the EN standards to which the product was tested to demonstrate compliance.

This equipment meets the following IEC classifications:

Group 1 ISM equipment: group 1 contains all ISM equipment in which there is intentionally generated and/or used conductively coupled radio- frequency energy which is necessary for the internal functioning of the equipment itself.

Class A equipment is equipment suitable for use in all establishments other than domestic and those directly connected to a low voltage power supply network which supplies buildings used for domestic purposes.

This device complies with the requirements of CISPR11, Group 1, Class A as professional radiation equipment. Therefore, there may be potential difficulties in ensuring electromagnetic compatibility in other environments, due to conducted as well as radiated disturbances.

The Agilent SPS 4 autosampler, represents the state- of- the- art in autosampler design and is the latest generation of autosamplers from Agilent. The Agilent SPS 4 is a random access, single probe autosampler, providing front- end automation for several Agilent instruments.

The Agilent SPS 4 can accommodate a variety of fixtures, including a standards rack, four sample racks, a wash reservoir, a sample probe and other accessories.

Depending on your application, there is a choice of options. Note that “diameters” in the following list mean outside diameters (OD):

• Standards Rack:34 well rack (12 wells for 29 mm diameter tubes + 22 wells for 17 mm diameter tubes)5 well rack (for 61 mm diameter bottles)

• Flat Pack Sample Racks: 21 position rack for 30 mm diameter tubes24 position rack for 25 mm diameter tubes40 position rack for 20 mm diameter tubes60 position rack for 17 mm diameter tubes90 position rack for 13 mm diameter tubes

Agilent SPS 4 Autos

The Agilent SPS 4 has 3 axes of movement for the sample probe — longitudinal (X- axis), rotary or theta (T- axis) and up and down (Z- axis). Figure 1 shows the front view of the Agilent SPS 4.

1 Spill Tray

The spill tray sits on the bottom base bars of the Agilent SPS 4, and catches any spills.

2 Rack Location Mat

The rack location mat sits on top of the spill tray, and positions the sample racks, standards rack and fixed wash reservoir.

3 Standards Rack

The standards rack is located in the center of the rack location mat. There are 2 standard rack options.

• 34 well (12 wells for 29 mm OD tubes + 22 wells for 17 mm OD tubes)

• 5 well (for 61 mm OD bottles)

4 Sample Racks

The sample racks are located on both (left and right) sides of the standards rack. The rack location mat has a maximum of four sample rack positions — two rack positions on either side of the standards rack position.

• 21 position rack for 30 mm OD tubes

• 24 position rack for 25 mm OD tubes

• 40 position rack for 20 mm OD tubes

• 60 position rack for 17 mm OD tubes

• 90 position rack for 13 mm OD tubes

5 Microtiter Plate Adapter

The microtiter plate adapter is used to mount four 96- well microtiter plates on the rack location mat. It occupies two sample rack positions.

6 Fixed Wash Reservoir

The fixed wash reservoir is located at the rear of the rack location mat. It is supplied with tubing used to connect the wash reservoir to the rinse source and the waste container.

7 Probe Arm

The probe arm moves the sample probe on 3 axes — longitudinal (X- axis), rotary or theta (T- axis) and up and down (Z- axis).

8 Sample Probe

The sample probe is mounted on the probe arm.

9 Power/Error Indicator

The power/error indicator is located on the front panel. The LEDs are green to indicate power is on and the autosampler is functioning correctly. The indicator lights change to red when an error occurs in the equipment. There are 4 error indicators and these illuminate in different combinations to indicate a specific error condition. Rear View

Figure 2 shows the rear view of the Agilent SPS 4.

1 Power Switch

The power switch turns on/off the Agilent SPS 4. I: Power is ON.

O: Power is OFF.

When the Agilent SPS 4 is turned on, the initialization sequence runs to check the basic functions and set the probe position. If there are no errors, the power/error indicator turns green.

The output connector from the AC adapter is connected to this socket.

3 Host Communications Port (USB)

The host communications port connects the Agilent SPS 4 to a computer controlling your Agilent instrument or in some cases, to the instrument directly, using the furnished standard USB Type A to B cable. The Agilent SPS 4 will appear as a virtual COM port under Windows.

4 Auxiliary Communications Port (AUX)

The auxiliary communications port provides an RS485 interface, but is not used in current Agilent software.

5 3- channel Peristaltic Pump

The peristaltic pump is located in the center on the rear panel. Maximum two channels of three- channel pump are used to pump the rinse solution to the fixed wash reservoir. One channel can also be used to pump the used rinse solution to the waste container (optional). This peristaltic pump may also be called a wash pump.

The wash pump speed is adjustable using the instrument control software.

6 Purge Input

The purge input port is not currently used.

7 Cable Ducting

The cable ducting is used to hold the USB cable and the power cable.

8 Wash Pump Tubing Ducting

A ducting plate that extends vertically down the back of the autosampler for routing the tubing from the wash pump to the fixed wash reservoir. Clips are used to secure the wash pump tubing in place. The same clips are also used to anchor the sample line from the sample probe to an optimal length.

Agilent SPS 4 Autos

The Agilent SPS 4 is designed to automate the presentation of liquid samples, drawn from tubes held in racks loaded on the rack location mat, to the instrument for analysis. The Agilent SPS 4 is controlled through software commands sent via a USB interface.

The Agilent SPS 4 should only be used within the stated specifications.

The autosampler should be placed on a bench with a height between 790 mm (31.1") and 910 mm (35.8") for optimal user ergonomics. A footprint of 600 mm × 363 mm (23.6" × 14.3") is required. The bench should be flat with no more than 3 mm (7/64”) variation. If a sampler cover kit is fitted, additional space should be allowed on the side where the exhaust hose ducting is located.

The autosampler should be located in close proximity to your instrument so as to reduce the delay caused by the length of capillary tubing connecting to the autosampler.

Inspection and Unpacking

Unpacking and Lifting

Keep the shipping box and packaging. If you need to return the autosampler during the

NOTEwarranty period, it is a requirement of the warranty that the equipment is repacked in the original shipping box. For instructions on packing the equipment, refer to the Packing/Unpacking Guide.

Inspect all items for damage. Any damage or missing items should be reported to

CAUTIONAgilent immediately. Cite the part number and serial number of your autosampler.

Danger to hands and feet. When unpacked the autosampler is heavy. To avoid injury

The unit should be lifted/moved from the positions shown in Figure 4. One person should be at the front and one person should be at the rear of the unit. Bend at the knees if lifting from a low position.

A Protective Cover is available as an optional accessory. It is used to protect:

• samples from drafts, dust and airborne contamination

• the operating environment and/or the autosampler from the fumes or solvent vapors arising from exposed liquids

• the operator from possible injury caused by the movement of the probe arm or sharp edged sample probe

• the operator from sample fumes

It is made from transparent plastic panels provided in any easy- to- fit kit form. When fitted, the protective cover creates a closed environment and takes up no additional bench space. The autosampler working area is accessed through a sliding door that can lock in a raised position to allow for easy access to individual samples or racks.

When you install the autosampler with the optional Cover Kit, install the Cover Kit before

NOTEinstalling the autosampler. For instructions on installing the Cover Kit, refer to “Installing the Cover Kit” on Page 82.

It is recommended that the autosampler is fitted with a Cover kit when handling

Fitting the Spill Tray

WARNINGacidic solutions (samples, standards or rinse solutions), In this case, the fumes must be extracted continuously and at all times through an exhaust hose, which is connected to one of the Cover’s side panels.

Ensure that the autosampler does not have a cover kit installed if working with organic solvents.

To prevent you and the instrument from earthquake damage, it is recommended to

The autosampler has a spill tray underneath the rack location mat to contain any excess liquid that occurs as a result of an accidental spillage, for example, from a dropped sample rack or maybe an unanticipated overflow of the wash reservoir.

Place the spill tray flat on the deepest section located between the base bars. It should be fitted with the front edge oriented as shown in Figure 5.

It is recommended that any ingress of liquid into the spill tray is cleaned up immediately after a spill has occurred. To access the spill tray for cleaning, the sample racks, the standards rack and the fixed wash reservoir (together with associated tubing) need to be removed from the rack location mat (refer to “Installing the Rack Location Mat” on Page 36) before it can be lifted off the spill tray to access any contained liquid.

The autosampler has a separate rack location mat which sits on top of the spill tray and positions the sample racks (or a rack adapter for 96- well microtiter plates), standards rack and fixed wash reservoir.

To fit the rack location mat:

1 Ensure that the spill tray is correctly fitted onto the autosampler base bars.

2 One end of the rack location mat has a "dovetail" feature as shown in Figures 6 and 7. Orient the rack location mat so the dovetail feature is on the left hand side.

3 Ensure that the slot for the fixed wash reservoir is located at the rear of the unit when installing the rack location mat.

4 Position the rack location mat over the spill tray and align the dovetail feature in the mat with the dovetail feature in the left end frame before pressing down firmly.

5 Check that the rack location mat fits firmly in place on the spill tray by trying to move it from side to side and front to back. There should not be any movement when fitted correctly.

6 If the rack location mat does not fit correctly even after completing the steps above, contact your local Agilent office or representative.

Installing the Wash Reservoir

The fixed wash reservoir is supplied as a kit with the inlet and outlet nipples fitted along with some lengths of inlet (2.5 mm ID) tubing and outlet (5.0 mm ID) tubing. For the single port fixed wash reservoir, rinse solution flows up through the smaller wash chamber and overflows into the larger drain chamber to waste. For the dual port fixed wash reservoir, there are two wash chambers and a common drain chamber.

The fixed wash reservoir is mounted at the rear of the rack location mat (see Figure 6).

To install the fixed wash reservoir:

1 Put the base of the fixed wash reservoir into the locating hole and twist it clockwise 60 degrees so it locks into position. The nipples on the fixed wash reservoir should be facing toward the rear of the

autosampler (see Figure 8).

2 Ensure that the fixed wash reservoir is seated correctly by visually checking that it is both perpendicular and firmly attached to the rack location mat.

3 The larger (5 mm) nipple is the outlet nipple. Attach the 5 mm ID tubing and place the other end of the tubing (drain line) into a suitable waste container.

If a rear autosampler cover panel is fitted, the drain line needs to be guided through the oval rubber grommet in the bottom of the rear cover (see Figure 9).

Air locks in the drain line may cause rinse solution in the fixed wash reservoir to back up

NOTEand overflow. To prevent air locks, the drain line should fall freely and drop vertically into the waste container. The end of the drain line should be suspended above the liquid level in the waste container.

4 The 2.5 mm tubing is attached to the inlet (lower) nipple which, in turn, is connected to the outlet tubing from the wash pump. For the dual port fixed wash reservoir, there are two inlet nipples which will need to be connected to tubing from 2 separate channels of the wash pump.

If a rear autosampler cover panel is fitted, the inlet tubing from the wash pump needs to be guided through the oval rubber grommet in the bottom of the rear cover (see Figure 9).

5 Re- fit the rack location mat to the unit.

Installing the Standards Rack

NOTE• There is no leakage between the main body and the connector of the

fixed wash reservoir.

• The drain is not blocked by rinse and sample solution chemistry.

The standards racks are located in the center of the rack location mat by placing the rack’s feet into the holes in the rack location mat. There are 2 standard rack options as shown in Figure 10. Either may be supplied with the equipment depending on your application.

• 34 well rack (12 wells for 29 mm OD tubes + 22 wells for 17 mm OD tubes)

• 5 well rack (for 61 mm OD bottles)

To install the standards rack:

1 Locate the holes for the standards rack in the rack location mat.

2 Align the feet from the standards rack with the holes in the rack location mat. Press down on the rack to ensure it is correctly located and the bottom of the standards rack is sitting flush with the rack location mat.

Installing the Sample Racks

The sample racks are located on both (left and right) sides of the standards rack by mating the rack’s feet with the holes in the rack location mat. The rack location mat has maximum four sample rack positions — two rack positions on the either side of the standards rack position. The following rack options are supported.

• 21 position rack for 30 mm OD tubes

• 24 position rack for 25 mm OD tubes

• 40 position rack for 20 mm OD tubes

• 60 position rack for 17 mm OD tubes

• 90 position rack for 13 mm OD tubes

To install the sample racks:

1 Locate the holes for the sample racks in the rack location mat.

2 Align the feet from each sample rack with the holes in the rack location mat. Press down on the rack to ensure it is correctly located and the bottom of the sample rack is sitting flush with the rack location mat.

To ensure the sample racks are seated correctly in the rack location mat, it is recommended

NOTEthat you use the sample racks supplied with the autosampler. Sample racks from alternative suppliers may not fit correctly.

It is important to ensure that the vials sit vertically within the sample rack. If a vial sits

CAUTIONat an angle in the rack, the probe may hit the side of the vial as it enters or leaves the vial. This can affect the alignment of the probe and/or damage the vial and the probe, which might lead to loss of sample.

If there is a gap between the diameters of the vial and the rack well, the vials may not sit vertically, resulting in the probe hitting the edge of a vial. Ensure that the selected vials fit the rack correctly.For example, a 60 position rack needs vials that OD is within 16.1 mm to 16.5 mm at a height of 60 mm. If the diameter is less than or equal to 16.1 mm, you may be able to use them with optional rack overlays.

Always switch off the power to the autosampler before installing or removing the

To fit the sample probe:

1 Turn the autosampler off.

2 Manually rotate the probe arm so it faces the front of the autosampler for easy access.

3 Undo the mount nut from the probe mount block on the sample probe and slide the nut off the sample probe.

4 Place the mount nut from the sample probe on top of the accessories plate with the recessed edge facing up and the holes approximately aligned.

5 Move the Z- axis slide down stopping just above mount nut sitting on the accessories plate.

6 Slip the tip of the sample probe into the hole in the middle of the Z- axis slide, through the mount nut and the accessories plate until the probe mount block is sitting in the z- axis slide.

7 Slide the mount nut up onto the probe mount block and tighten to secure the sample probe to the Z- axis slide.

8 Route the sample line from the sample probe to the left around the probe arm housing. When using a carbon fiber sample probe, do not run the sample line through the hook (or tube restraint) at the top of the Z- axis slide. Connect the cable tie to the eyelet on the underside of the probe arm cover and leave a loop of approximately 10 mm. Thread the sample line through the loop in the cable tie as shown in Figure 14.

Do not use the tube restraint for pre- formed sample probes. It is only

9 Feed the sample line through the rubber grommet in the middle of the wash pump tubing ducting and then slot the sample line into the tube anchor clip between the colored markers on the tubing as shown in Figure 14. The tube markers on the sample line define the optimal sample length of sample line from the sample probe to the exit point of the autosampler.The following lists the color of the tube markers on the sample probe tubes.

• Red: Sample probe kit, 0.25 mm ID

• Yellow: Sample probe kit, 0.5 mm ID

• Green: Sample probe kit, 0.8 mm ID

• Blue: Sample probe kit, 1.0 mm ID

• Black: Sample probe kit, 1.2 mm ID

• Red: Sample probe to suit microtiter plate

10 Connect the sample line to the input of your instrument's sample introduction system, adjusting the length from the exit point as required.

Fitting the Wash Pump Tubing

CAUTIONfreely in all axes. If it is too tight, it will restrict the probe arm and may cause a movement failure. If it is too loose, the probe arm may become tangled in the tubing as it moves.

A wash pump is fitted on the rear of the autosampler and is used to supply fresh rinse solution to the fixed wash reservoir. The wash pump is a three channel peristaltic pump with four rollers and an individual pressure plate for each channel. One and two channels of the wash pump are used to feed rinse solution into the single and dual port fixed wash reservoirs respectively. The third channel can also be used to pump the used rinse solution to the waste container (optional).

The wash pump is controlled by the instrument control software and rotates clockwise.

The wash pump accommodates standard 3 bridged pump tubing which is available in a range of internal diameters for delivering different flow rates. The optimal distance between the bridges on the pump tubing is 82–85 mm.

Wash pump tubing is available in different tube materials so ensure that you select a tube

NOTEmaterial that is compatible with the rinse solution. If you are not sure which material to use, contact your Agilent representative for a recommendation.

When a pump channel is not in use, release the occlusion arm on the channel and remove the tubing from the guides to extend the life of the tubing.

Two Drain Arrangements

The waste rinse solution drains from the wash reservoir by means of a gravity drain, which is the standard arrangement for draining the wash reservoir. If preferred, you can create a pumped drain arrangement. This section introduces the gravity drain arrangement and the pumped drain arrangement.

Gravity Drain Arrangement

In a gravity drain arrangement, the rinse solution moves through a channel (and two channels for dual port wash reservoir) of the peristaltic pump to the inlet of the wash reservoir, as shown in Figure 16. It then drains out from the outlet of the wash reservoir into the waste container by means of a gravity drain.

1 Release the pressure plate by disengaging the occlusion arm.

2 Fit one of the outside bridges on the pump tubing into the appropriate left tube clamping block.

3 Feed the tubing around the rotor and fit the center tube bridge into the right tube clamping block. Ensure there are no twists or kinks in the tubing after loading as this will adversely affect tube life.

4 Reposition the pressure plate over the rotor and fix by slotting the occlusion arm back into the track slot.

5 Route the outlet of the wash pump tubing through the tube anchor clips to the delivery line or input of the fixed wash reservoir.

6 Route the inlet line of the wash pump tubing through the tube anchor clip(s) and connect this to the suction line from the rinse solution reservoir.

• Keep delivery and suction lines as short as possible.

Pressure Plate Adjustment

NOTE• Run at a slow speed when pumping viscous fluids.

• Use the largest bore tube running at a slow speed for the longest tube life.

• It is recommended to release the occlusion arm when the pump is not in use to extend the life of the pump tubing.

The position of the occlusion spring on each channel of the wash pump is set in the factory to optimize the tension on the pressure plate when pump tubing is fitted to the wash pump. It is possible to adjust the tension on the pressure plate on a specific channel (only if the wash pump can be turned on via the instrument control software) using the following procedure:

1 Fit a new pump tube to the specific wash pump channel as described on Page 47.

2 Connect the inlet line to the wash reservoir and place the outlet line into a beaker.

3 Loosen the occlusion screw with a 5.5 mm (7/32”) wrench until there is no pressure on the pump tubing.

4 Turn the wash pump on using the instrument control software.

5 While the wash pump is running, tighten the occlusion screw until rinse solution starts to flow.

6 Tighten the screw a further half turn.

Connecting the Power and the Communications Cable

Before connecting any part of the autosampler to the mains power supply, check you have the correct power supply. The power requirements are given in “Power” on Page 22.

Application of the wrong supply voltage can create a fire hazard, or a potentially

WARNINGserious shock hazard, and can seriously damage the instrument and any attached ancillary equipment. Do not connect the instrument to the mains power until you have ensured that the mains power supply from the outlets in your laboratory is within the range stated in “Power” on Page 22.

The rear panel of the autosampler contains the peristaltic pump, purge input, the power socket and switch, and a USB port for communication between the autosampler and the computer that is running instrument control software, as shown in Figure 2 on Page 19.

To connect the power:

1 Ensure that the power switch is turned off.

2 Connect the barrel output connector of the AC adapter to the power supply input on the rear panel of the autosampler.

3 Connect the input plug of the AC adapter to a suitable mains power outlet.

Ensure the autosampler is always connected to the protective grounding of the mains supply.

Do not switch on the autosampler until all checks and settings for the operation have been completed.

Host Communications Port (USB)

The host communications port (USB) is used to connect the autosampler to the computer controlling your instrument.

To connect your autosampler:

1 Plug one end of the supplied USB cable into the host communications port.

2 Connect the other end of the USB cable to a USB port on your computer or instrument.

3 Fix the USB cable and the power cable using the cable ducting on the rear column of the autosampler.

Auxiliary Communications Port

Confirming the Operation

NOTEThe Auxiliary communications port is not used in current Agilent software.

When the autosampler is turned on it will go through initialization tests and set the probe position. If the initialization process is not successful, refer to the troubleshooting information that is provided in “Initialization Problems” on Page 68.

Ensure that the probe arm’s movement is not interfered with during operation/initialization.

During the initialization sequence:

1 The probe moves upwards on the Z- axis and stops.

2 The probe arm moves in the opposite X- axis direction to which the probe arm is facing and homes against an end frame.

3 The X- axis may move to clear the end frame. The probe arm rotates counter- clockwise on the Theta- axis and stops.

4 The X- axis homes against the left end frame.

5 The probe arm moves to the four corners of the rack location mat.

6 The probe arm stops above the fixed wash reservoir.

Setting up the Instrument Controlling Software for SPS 4

You need to set up your instrument control software in order to set parameters such as the rack configuration, the communication port, probe depth, and so on. For instructions on how to install your instrument software, see the associated documentation or the instrument Help.

Ensure that the autosampler can be controlled by the instrument software when setup is complete.

When lifting the autosampler:

• Ensure the power is off and the power and USB cables are disconnected.

• Remove any accessories fitted to the rack location mat such as sample racks, standards rack, the fixed wash reservoir and any associated tubing.

• Remove the clear side covers (if fitted).

• It is advisable to lift the autosampler with two people positioning their hands through the end frames at either end as shown in Figure 18.

Before starting the autosampler, carefully read Chapter 1, "Safety Practices and Hazards" and ensure that the laboratory is set up according to the details specified.

To start up the autosampler:

1 Ensure the autosampler is connected via a three- pin, grounded power cord of the AC adapter to a three- pin, grounded power outlet.

2 Ensure that the wash reservoir is properly connected.

3 Turn on the power using the Power Switch on the rear panel of the autosampler.

The autosampler performs an initialization sequence on startup. The power/error indicator is illuminated green when the initialization sequence is completed correctly.

The initialization sequence moves the probe arm through the four corners of the rack

NOTElocation mat and finishes with the probe repositioned above the fixed wash reservoir. This process takes about 30 seconds.

If an error occurs during the initialization sequence, refer to Chapter 6, "Troubleshooting".

4 Adjust the pressure plates of the wash pump until the desired rinse solution flow rate is achieved. Refer to “Pressure Plate Adjustment” on Page 48.

5 Purge air from the wash system (wash reservoir and flow path) by placing the rinse solution uptake tubing in the rinse solution source and running the rinse solution through the wash reservoir.

Ensure there are no air bubbles visible in the rinse uptake tubing before you start using the autosampler.

To shutdown the autosampler, complete the following steps:

1 Drain the wash system by removing the rinse solution uptake tubing from the rinse solution source. Let the wash pump run until all solution drains from the tube attached to the wash reservoir outlet.

If you use a rinse solution other than deionized water, flush the wash reservoir with deionized water before shutting down the autosampler. For more information, see the following section, “Flushing the Wash Reservoir and Flow Path” on Page 55.

2 Release the pressure plate on the wash pump.

Releasing the pressure plate decreases wear on the pump tubing.

3 Turn off the autosampler power switch.

Flushing the Wash Reservoir and Flow Path

Generally, you can operate the autosampler without flushing the wash system (wash reservoir and flow path). Under normal circumstances, you can simply drain the wash system prior to shutting down the autosampler. However, you need to flush the wash reservoir and flow path under two circumstances:

• during initial startup of the autosampler after installation

• following the use of strong bases, acids, or organic solvents as rinse agents

Flushing the wash system during initial startup of the autosampler removes any contaminants that could cause interference during sample analysis. Flushing the wash system after using strong rinse agents prevents degradation and failure of the flow path components.

To flush the wash reservoir and flow path, complete the following steps:

1 Insert the rinse uptake tubing into a deionized water source.

If you are flushing the wash system during initial startup, first use a 2% nitric acid solution

Once the wash system has been flushed, you can proceed with the sampling sequence or drain the wash system as part of the shutdown procedure.

All operations of the autosampler can be controlled by commands from the instrument control software. For details on how the software interface interacts with the autosampler, consult the relevant section of the manuals or instrument Help.

Routine Maintenance

The following parts of the instrument require routine maintenance.

Daily

• Confirm that the capillary tubing from the probe to the nebulizer is secure and kink free.

• Rinse the probe and transfer tubing by pumping rinse solution or distilled water at the end of analysis.

• Clean up any spills and wipe the rack location mat and the probe arm with a damp cloth. Refer to “Cleaning” on Page 58 for further details.

• Check the probe and capillary tubing for blockages.

Weekly

• Check the wash pump flow rates, and replace the pump tubing if necessary. Alternatively, move the tubing so that the other set of bridges are secured in the peristaltic pump.

Cleaning

The exterior surfaces of the autosampler, spill tray, rack location mat and accessories should be kept clean. All cleaning should be done with a soft, lint- free cloth. If necessary, this cloth can be dampened with water or a mild detergent.

WARNING The probe arm of the autosampler can move without warning that may pose a potential hazard due to its moving parts. Always switch off the power to the autosampler before cleaning the autosampler.

CAUTION Clean any sample spills immediately to preserve the life of the instrument.

If necessary, the probe can be removed from the autosampler and placed in an ultrasonic bath for cleaning.

Replacing the Wash Pump Tubing

Routine maintenance of the autosampler includes replacement of the wash pump tubing. Because of the operating nature of the wash pump, the tubing will probably be the most frequently replaced item on the Agilent SPS 4 autosampler. If you use strong bases, acids, or solvents as rinsing agents, the tubing may break down rapidly.

The optimal distance between the bridges on the pump tubing is 82–85 mm.

To replace the wash pump tubing, complete the following steps:

1 Shutdown and unplug the autosampler.

2 Release the pressure plate by disengaging the occlusion arm.

3 Remove the old tubing, and replace the pump tubing.

Carefully pull or cut the old tubing to remove it.Replace the new tubing carefully. Damage can result if you apply too much force.

4 Install new tubing following the instructions in “Loading Wash Pump Tubing” on Page 47.

CAUTION Do not use organic solvents or abrasive cleaning agents. This will help to preserve the plastic components and painted surfaces.

NOTE A rinse solution should always be pumped through the autosampler at the end of a run.

The Agilent SPS 4 is equipped with diagnostic tools such as power/error indicator and initialization sequence on startup to assist in identifying the problems.

Power/Error Indicator: Provides useful information on the error state of the

autosampler.

Initialization Sequence: Tests that the probe arm functions in all its movement

axes by moving through the standard and sample positions for the currently loaded mat and rack settings.

To identify your problem:

• If the autosampler appears to have no power and the power/error indicator is not illuminated, refer to “Power Supply Problems” on Page 65.

• If the power/error indicator is not lit or the LEDs are lit RED, refer to “Operating State Problems” on Page 65. For the error state indicated by the power/error indicator, refer to “Error Indicator Codes” on Page 66.

• If the autosampler fails in the initialization sequence, refer to “Initialization Problems” on Page 68.

• If the initialization sequence is successful but the autosampler has a malfunction in the probing operation, refer to “Probing Problems” on Page 69.

Power Supply Problems

If the Agilent SPS 4 appears to have no power and the power/error indicator is not illuminated:

1 Check that the power cable is connected to the power supply and that it is turned on.

2 Visually check the Agilent SPS 4 and power lead for any electrical damage.

3 If the Agilent SPS 4 still fails to initialize contact your Agilent customer support representative.

Operating State Problems

The power/error indicator is used to identify the operating state of the Agilent SPS 4:

• If it is lit GREEN then the Autosampler has power. It is reasonable to assume that the autosampler is working correctly.

• If it is not lit, then turn off the power at the power switch on the rear panel of the autosampler. Turn the power off at the power point. Check that the power cable is correctly seated into the power supply. Check the power plug from the power supply is inserted correctly into the power supply inlet on the rear panel of the autosampler. Turn the power on at the power point. Turn the power to the autosampler back on at the power switch.

• If the autosampler initializes but the power/error indicator does not illuminate then the indicator is potentially faulty. Contact your authorized Agilent representative.

• If the autosampler does not initialize and the power/error indicator does not illuminate then it is possible that either the power supply or main board may be faulty. Contact your authorized Agilent representative.

Error Indicator Codes

The power/error indicator on the front panel is a line of four LEDs. These LEDs illuminate RED when an error has occurred with the Agilent SPS 4. The red illumination of these LEDs indicates a binary error code to assist you in identifying the possible cause of the incorrect functioning of the unit. Other more extensive error reporting is available via the software commands.

Table 7 shows the binary error codes by the power/error indicator and error descriptions. In the table, E1 means the leftmost LED, E4 means the rightmost LED, 0 means LED off, G means LED (green) on, and R means LED (red) on.

If an error occurs, a combination of the LEDs show the binary error code for the fault and an error message is sent to the instrument control software. For example, Error 7 = 0RRR is a theta- axis position error.

Initialization Problems

The initialization sequence is a powerful self- diagnostic tool. It runs immediately after the autosampler is powered on and tests the functionality of the autosampler. The probe arm goes through a sequence of movements and stops above the fixed wash reservoir position on the rack location mat. If there is a problem with the autosampler, then it will likely manifest itself at some stage during the initialization sequence.

The correct initialization sequence can be seen on Page 50.

If the autosampler fails to complete initialization, the power/error indicator illuminates to display the error condition. See Table 7 for the error indicator codes.

Check the Agilent SPS 4 by doing the following:

1 Turn the power off and move each axis manually until they stop and determine if there is any mechanical obstruction preventing full travel.

2 If no mechanical obstruction can be identified turn the unit back on and allow it to attempt the initialization process again.

3 If unsuccessful, contact your Agilent customer service representative.

Probing Problems

Probe Arm Jam

Agilent SPS 4 Autos

A probe arm jam is indicated when the Agilent SPS 4 cannot complete its instructed move. This could be caused by:

• The probe arm hitting an obstacle or the sample line is restricted while moving into position.

• The moving components have excessive friction.

• The position of the probe arm has been moved without being driven by the autosampler (for example, when bumped by a user).

If this happens, check the autosampler by doing the following:

1 Turn the power off and move each axis manually until they stop and determine if there is any mechanical obstruction preventing full travel.

2 If no mechanical obstruction can be identified turn the unit back on and allow it to attempt the initialization sequence again.

3 If unsuccessful contact your Agilent customer support representative.

When the Sample Probe Hits an Object

If the sample probe hits an object like a sample rack or sample vial, or it misses the wash position in the fixed wash reservoir, it may generate error condition 13 (refer to Table 1) and the autosampler will stop. Check the following:

1 Check that the probe is straight and mounted tightly to the Z- axis slide.

2 The probe should pass through the center of the accessories plate at the bottom of the probe arm (see Figure 13).

3 Check that the spill tray and rack location mat are fitted correctly (see Chapter 3, "Installation" for instructions).

4 Check that sample tubes sit vertically in the sample rack and are not leaning to one side. If the sample tubes are not suitable for use with the sample rack, change them to the correct diameter tubes to suit the sample rack or change to the correct sample rack size.

5 Check that the fixed wash reservoir is correctly mounted.

6 Check that the rack configuration is correct.

If all accessories are fitted correctly, reset the unit and attempt to enter the fixed wash reservoir again. If unsuccessful, perform user calibration. User calibration makes minor adjustments to the centering of the sample probe on the Agilent SPS 4.For information on user calibration, refer to Chapter 7, "Running the Calibration Wizard".

If you performed the calibration procedure but the problem remains unresolved, contact your Agilent customer support representative.

Communication Problems

1 Check that the computer and the autosampler have the same communication settings.

2 Disconnect the USB cable and initialize the autosampler to ensure that it is in a working state. To do this, turn off the power switch, wait 10 seconds and then turn the power switch on. If the Agilent SPS 4 completes the initialization sequence with no error indicators, then this usually indicates that the autosampler is working fine.

3 Reconnect the USB cable and ensure it is firmly seated at both ends.

4 Using the instrument control software, send a command to the Agilent SPS 4. If the Agilent SPS 4 responds when the command is sent, this means it has received the command from the computer. If unsuccessful, contact your authorized Agilent representative for further instructions.

The sample probe can be displaced from the center after hitting a sample vial or the sides of the fixed wash reservoir. User calibration allows for minor adjustments to center the sample probe. This chapter provides instructions on performing user calibration for the Agilent SPS 4 autosampler using the Calibration Wizard software.

Before User Calibration

Check the following points before performing user calibration. Performing user calibration when the unit and accessories have not been setup correctly could cause a hazard.

• The spill tray is correctly fitted.

• The rack location mat is seated correctly in the spill tray and end frames.

• The sample racks are correctly engaged in the locating holes in the rack location mat and are seated flat on the rack location mat.

• A sample probe is fitted and has been mounted correctly.

• The sample probe is essentially straight.

Calibration Wizard Overview

Use the calibration wizard to adjust the centering of the sample probe. There are four calibration points, one for each corner in the movement range of the probe arm. At each calibration point, adjust the probe position.

Com Port: This area is used to connect this wizard to your autosampler. When the wizard starts, select the relevant COM port, and then click Connect. Refer to the instrument Help for more information.

Undo button: Reverts to the original calibration values and restarts the calibration process.

Reset button: Starts the calibration process.

Mat Layout: The area under the Com Port area shows the information on the rack layout specific to your autosampler when the calibration wizard has successfully connected to your autosampler.

Z Axis Position: This area is used to adjust the Z-axis position of the sample probe at each calibration point. The indicator shows the present height of the sample probe. The Up/Down bottoms move the sample probe height.

Probe Arm: This area is used to adjust the X and Y-axes positions of the sample probe at each calibration point.

“Rear Left”, “Rear Right”, “Front Left”, and “Front Right” buttons: Moves the probe to the calibration point “Rear Left”, “Rear Right”, “Front Left”, or “Front Right”. Since the general calibration process automatically moves the probe in the correct order, you do not need to use these buttons. If you want to try again after performing general calibration, we recommend moving the probe to the calibration point you want to recalibrate.

The indicators next to the “Rear Left”, “Rear Right”, “Front Left”, and “Front Right” buttons indicate the following status.

Orange: Ready to calibrate at this point.Green: Calibration successful at this point. Red: Not ready or calibration error.

Arrow buttons: Adjusts the XY position. Use the single arrow buttons to move the sample probe 0.1 mm for fine adjustments and the double arrow buttons to move it 1 mm for larger adjustments.

Capture button: Stores the calibration data at each calibration point. Press this when the probe is centered in a good position in the sample.

Performing User Calibration

To perform user calibration:

1 Ensure the autosampler power is on.

The sample racks fitted should be empty and not contain any sample vials. If a microtiter plate Adapter is fitted, it should be loaded with four 96 well microtiter plates.

2 Start the Calibration Wizard software. The Calibration Wizard window opens as shown below.

3 Select the appropriate COM port from the drop- down list, and then click Connect in the Com Port field.

If you chose the wrong one, an error message appears. If the appropriate COM port number is not listed, check the autosampler power, the USB connection, and the COM port setting in the instrument control software.

4 If the calibration wizard can connect to your autosampler successfully, the field under the Com Port field shows the information on the rack layout specific to your autosampler. Ensure that the sample rack types displayed on the screen correspond to those loaded into their respective positions on the rack location mat.

NOTE Close the instrument software before launching the Calibration Wizard.

5 Click Reset to start the calibration process.

6 The button labeled “Rear Left” changes to orange and the sample probe moves to point “Rear Left” above the far left sample well in the sample rack or microtiter plate in that position. The sample probe is at the top of the Z- Axis stroke (i.e., the Z- axis position is zero).

To adjust the probe position in point “Rear Left”:

a Adjust the sample probe height to the top of the sample well using the Up/Down buttons.

b Adjust the XY position to the middle of the well using the arrow buttons.

c When the position of the probe is centered in the sample well, click Capture.The indicator next to the “Rear Left” button changes from orange to green.

NOTE During the calibration process, it is important that you do not click the Capture button while moving to the different calibration points. If you have adjusted the position to be too far away from the theoretical XY position at any point during the calibration, an error message is displayed to instruct you to try again.

10 When all four points have been captured, a confirmation message is displayed asking if you want to write the new calibration values.

• Click Yes to write the calibration values.The autosampler is adjusted using the new calibration value.

• Click No if you want to retry. The calibration process is restarted.

11 After calibration has been successfully completed, check the positioning of the sample probe by clicking on each corner position button, and driving the Z- Axis position down. Confirm that the sample probe is positioned in the center of each well.

Error List

On running the calibration wizard software, you may encounter any error listed below:

• Serial Port Error

Example: Autosampler did not respond.

A serial port error occurs if the software cannot communicate with the autosampler. Check that the power is on, and no error lights are illuminated. Also confirm the USB connection and the COM port setting in the instrument control software. If the problem still remains unresolved, contact your Agilent customer support representative.

• Envelope Error

Example: Probe reached the maximum Y limit.

An envelope error occurs if you try to move the sample probe outside its motion envelope limits. If the well position is outside the motion envelope, contact your Agilent customer support representative.

• Capture Failure

Example: Probe is too far from starting point.

A capture failure occurs if the point you are trying to capture is too far away from the expected position. Check that the sample probe positions above the correct well position, and the sample racks you are using match the description in the Calibration Wizard. If the problem still remains unresolved, contact your Agilent customer support representative.

• Autosampler Error

Example: Move Z failed with ERR(8).

An autosampler error occurs if the arm has a collision, such as if the probe hits the sample rack. If this happens, click the Reset button. The autosampler will reset, and the calibration process can be continued. If the problem still remains unresolved, contact your Agilent customer support representative.

• Skipping Calibration

Example: None of the positions were adjusted. Skipping calibration.

If all four captured positions are within 0.5 mm of their original position, the existing calibration is deemed to be accurate, so the calibration does not be adjusted. In this case, any problems you are having are unlikely to be caused by alignment. Contact your Agilent customer support representative.

The Autosampler Cover covers an Agilent SPS 4 autosampler and can be used to:

• Help protect the operator and the laboratory environment from sample vapors.

• Protect the samples from drafts, dust and possible airborne contamination.

• Protect the autosampler from corrosive vapors arising from handling acidic samples, standards or rinse solution.

Use gloves to avoid fingerprints on the clear panels.

Fitting the Side Covers

1 Fit the exhaust hose connection into the side cover with the hole by rotating the end with the bayonet fitting clockwise as shown below.

2 Take the side cover with the exhaust hose connection fitted and fit to the side of the autosampler that suits connection to an exhaust hose. The side cover is pushed into the recesses of the end frame from the inside and slides down to lock in place.

3 Fit the other side cover (blank) to the other end of the autosampler using the same process.

Fitting the Front Cover

1 Position the front cover with the handle facing outwards and the bottom fold facing inwards. Guide the front cover into the slots at the top of the end frames as shown below.

2 Supporting the front cover underneath, gently guide the front cover down until it sits on the rack location mat.

Door Operation

Agilent SPS 4 Autos

The door (front cover) has two positions:

1 In the Closed position, the door creates an enclosure around the samples.

2 In the Open position, sample racks and standard racks/tubes can be loaded or unloaded.

Dropping the front cover can cause liquid to splash from sample tubes which could

The door is held in the open position by a step in the end frame slots.

To change the sample probe or access the fixed wash reservoir, it is recommended to first remove the front cover. Slide the front cover up until the handle is almost touching the chassis and then flex it slightly forward to remove.

Connections

General

There is an entry/exit point to the fixed wash reservoir through a rubber grommet on the rear cover.

Exhaust Ducting

On the side cover there is a ducting connection for connecting exhaust ducting. The hole will suit ducting with an internal diameter of 50 mm (1.97”) used in conjunction with a cable tie or hose clamp.

Care and Maintenance

• Do not use abrasive cleaners or strong solvents to clean the cover as this will reduce the clarity of the cover.

• Use a soft cloth and household window cleaner to clean off fingerprints on the cover.

• If using volatile chemicals ensure the cover is adequately evacuated at all times.

It is strongly recommended that an autosampler cover is fitted to the autosampler

WARNINGwhen it is used with samples, standards or rinse solution(s) that release corrosive fumes in any concentration. Whenever such liquids are present, the atmospheric contents of the sample cover must be extracted continuously at a minimum rate of 350 liters per minute. Corrosive samples and standards should be removed from the autosampler once analysis is completed.

Failure to provide adequate extraction at all times will potentially concentrate the

To avoid damaging rinse lines, select materials with appropriate chemical resistance for your application, particularly if you use a rinse solution other than deionized water.

For your reference, the standard supplied rinse lines contain the following materials:
