Semen, collected from bull are kept in 37 °C
Then mixed with Stock -A solution
The temperature is gradualy minimized into 4°C within 1.5 hours Along with it, Stock B solutin is also cooled into 4°C
Then 1/2 B solution is mixed with the semen in 4°C
After 30 minutes the rest 1/2 B solution is mixed at 4°C
Microscopic test of sperm viability
Filling & sealing of straw
Then the temperature is gradually minimized into -140°C in 4-5 hours & kept for 20 minutes
Finally the straws are transferred into liquid nitrogen having -196°C temperature
