THE ELIMINATION OF BIOFILM EMBEDDED BACTERIA BY ENHANCING SENSITIVITY OF CERTAIN ANTIBIOTICS WITH COPPER

Aim: Biofilms that are extracellular polysaccharides and produced by biofilm producer bacteria adhere bacteria on abiotic surfaces, ca-use transition of planktonic bacteria to sessile form.1 Antimicrobial and antibiofilm alternative solutions are needed to discover, instead antibiotics of which sole high dose and frequent usage can cause re-sistance to many antibiotics. It has been accepted that metals have antimicrobial activity. Abiotic surfaces such as orthopedic implants that are impregnated with copper can prevent colonization of biofilm producer pathogens, and can detach biofilms produced on implants. In this study, the effects of combination treatment of copper and am-picillin that is basic antibiotic frequently used against planktonic ba-cteria and biofilm embedded bacteria adhered on prosthetic implant were studied.  Materials & Methods: Minimum inhibitory concentrations of bi-ofilm embedded bacteria (bMIC) and Fractional inhibitory concentra-tions (FIC) indexes of combination treatments of ampicillin as a β-la-ctam and copper against biofilm producer clinical isolates. SA1, SA2, and SA3, SA4 isolates are MSSA (methicillin sensitive Staphylococcus aureus) and MRSA (methicillin resistance Staphylococcus aureus co-lonized on implant were determined according to Clinical Laboratory Standards Institute (CLSI), respectively.2 Results and Discussion: bMIC of copper and ampicillin against SA1, SA2, SA3, SA4 were 0.256 and 8.192, 0.128 and 4.096, 0.512 and 4.096, 1.024 and 8.19, respectively. This study demonstrated that the activities of copper with ampicillin were synergistic with the FIC in-dexes of 0.50, 0.39, 0.48 and 0.32 against SA1, SA2, SA3, and SA4, res-pectively. Conclusion: Indwelling medical devices such as orthopedic wires, prosthetics can be impregnated by β-lactams and copper to overco-me colonization and production of matured biofilm on indwelling de-vices, consequently, implant associated infections.
