This new development based on cell culture and hydroponics provides a simple, one-step answer to the problem of formulating clear AHA - ceramide systems for advanced skin care. Ceramide RW is a biologically engineered ceramide complex identical to human skin ceramides, and L- (+)-lactic acid is produced from hydroponically grown tomatoes, where total control over nutrients and root temperatures optimizes lactic acid content. The two are brought together to form ALPHA-CERAMIDEIN - your one- step answer to the formulation of advanced skin care systems.

Background information

ALPHA HYDROXY ACIDS……….

Facial cosmetics based on sour milk have been used since ancient Egyptian times, a period of some 5000 years. The main active ingredients are believed to be the alpha-hydroxy acids, predominantly lactic acid and its derivatives. Alpha-hydroxy acids (Ahas) are naturally occurs substances isolated from a variety of sources, such as sour milk, sugar cane, tomatoes and other fruit. They perform a number of functions at the level of the stratum corneum on the skin’s surface. There are many different kinds of alpha-hydroxy acids, and some out- performs others - even low usage levels. Intensive research has identified lactic acids, particularly the L-form as one of the most effective forms of AHA. Perhaps not surprisingly, this occurs naturally in the body. Campo’s L-lactic acid is isolated from tomatoes cultivated by hydroponics using root temperature control to optimize yields of L-lactic acid.

Ceramide(s) are found in human skin and comprise of 43-46% of the stratum corneum where they play a major role as a water retention barrier. They include sphingosine and fatty acids, and are generally linked with sugars to form structures called glycosphingolipids. Six kinds of ceramides have been identified in the human skin. Clinical studies of the application of the materials on the forearms of ten healthy male volunteers confirm the augmentation of water-retention, cell-to-cell adhesion, and in the prevention of barrier changes. Further clinical studies confirms that human ceramide levels decrease with increasing age, especially on the face, and in one third of individuals studied, on the hands as well. Campo’s Ceramide RW is a new complex of biologically engineered material, identical to human skin ceramide and is a spin-off from the P34 cdc2 (gene expression) research work on cellular signal transudation, especially in the area of skin cancer proliferation and down-regulation activities of certain natural products; conducted by Kampoyaki Research, Singapore.

AHA + CERAMIDES = ALPHA CERAMIDEIN

We have considered the skin as a structure akin to bricks and mortar. The bricks are the stratum corneum cells of the surface skin, and the mortar the intercellular cement that holds the cells together. As skin ages, cells lose their shape, inter-cellular material loosens its grip and the cells slide apart. This gives the skin a slack appearance, it loses its tone and fine lines and wrinkles begin to appear. Alpha-hydroxy acids work to restore the youthful appearance of the skin. However, they can only complete part of the job. They need the help of ceramides that act like a molecular mortar to reinforce the newer, smaller cells. Since Elizabeth Arden’s “Alpha-ceramide” 4 steps hit the market in the summer of the 1994, many cosmetic raw material suppliers have been trying to produce a clear, colorless, ceramide liquid based that is soluble in ethanol and water. These efforts have been unsuccessful except for a couple of vegetable ceramide powder types with poor stability. These turn into cloudy opaque liquids from which it is virtually impossible to formulate a clear, colorless, generic to the Arden product.

The answer is Campo Alpha -Ceramidein!!!!!

It has now become possible to super-charge AHA with ceramides, the moisture-enhancing lipids that support and hydrate young skin. Ceramides help to reinforce the skin’s moisture barrier and generally improve the skin’s condition creating a strengthened barrier upon which the AHA complex can work gently and effectively. The L-lactic acid complex works progressively as part of the alpha-ceramides unique step-by-step system and in this way, all skin types slowly adjust and acclimatize themselves to produce optimal results without the possibility of irritation.

Alpha-Ceramidein is a power-packed system to combat fine lines and wrinkles with L-lactic acid isolated from tomatoes and natural alpha-ceramides produced by cell culture.

OBJECTIVE:

To evaluate the test article for irritancy potential utilizing the MATREX in vitro toxicity testing system.

INTRODUCTION:

TESTSKIN and MATREX are sophisticated in vitro systems. Developed in Organogenesis Inc. of Cambridge, Massachusetts, they closely mimic human skin in structure and function. The Living Dermal Matrex (LDM) consists of a three-dimensional construct comprised of living cells in a collagen matrix. Nutrition is provided through the base via a permeable membrane, leaving the surface open to the atmosphere. This makes an ideal system for applying a variety of materials, including liquids, powders, oils, gels and creams. The Living Skin Equivalent (LSE) has all the features previously described, plus the formation of an actual epidermis complete with stratum corneum.

TESTSKIN and MATREX, when used with the recommended cell metabolism assay can quickly provide toxicological profiles. The procedure involves a solubilized, reactive tetrazolium salt (MTT), which is metabolized by the mitochondria of living cells and converted to a purple formazan dye. The color intensity of the skin replica extract, measured photometrically, correlates directly with its viability. When measured against controls, values ranging from 0% to 100% (plus or minus approximately 20%) can be calculated for each dose of an applied substance.

METHOD:

The appropriate dilutions of test sample and control articles were applied to MATREX. After the appropriate exposure period, the articles were rinsed from the MATREX surfaces. MTT (tetrazolium salt) assay medium was utilized in order to quantify cell metabolism. At the end of the staining period, excised portions of each MATREX were immersed in acidified isopropanol, which extracted the converted MTT from tissue samples. A Dynatech MR 4000 Automatic Microplate Reader was used to determine the absorbance of each extract at 570 nm. With the absorbance of a negative control defined as 100%, the percent absorbencies of the test and control articles were determined. The percentages listed below directly correlate with the cell metabolism in the MATREX samples.

HISTORICAL IN VITRO RESULTS: Propylene glycol has historically been categorized as virtually non-irritating when tested using the Draize irritation methodologies. Morpholine has been categorized as moderately irritating when tested in the same manner. DISCUSSIONS: The sponsor-submitted sample elicited in vitro results comparable to those recorded for propylene glycol.

CONCLUSION: The results indicate that the sponsor-submitted product has virtually no irritation potential, under the conditions of this test.

CONCLUSIONS

The test article Campo Alpha-Ceramidein was assessed for its potential to induce gene mutations according to the plate incorporation test (experiment I) and the pre-incubation test (experiment II) using Salmonella typhimurium strains TA 1553, TA 1537, TA 100 and TA 102. The assay was performed in two independent experiments both with and without liver microsomal activation. Each concentration, including the controls, was tested in triplicate. The test article was tested at the following concentrations: 33.3; 100.0; 333.3; 1000.0; 2500.0; and 5000.0 ug/plate

No toxic effects occurred in the test groups with and without metabolic activation in experiment I and II in all strains used. The plates incubated with the test article showed normal background growth up to 5000.0 ug/plate with and without S9 mix in all strains used. No substantial increases in revertant colony numbers of any of the five tester strains were observed following treatment with Campo Alpha Ceramidein at any dose level, either in the presence or absence of metabolic activation (S9 mix). There was also no tendency of higher mutation rates with increasing concentrations in the range below the generally acknowledged border of significance. A slight decrease (0.001%) in revertant colony numbers was observed in strain TA 102 at 333.3 and 1000.0 ug/plate in experiment I in the presence of metabolic activation. However, this effect is considered not to be relevant since it could not be reproduced in the normally more sensitive pre-incubation assay. Appropriate reference mutagens were used as positive controls and showed a distinct increase in induced revertant colonies. In conclusion, it can be stated that during the described mutagenicity test and under the experimental conditions reported, the test article did not induce point mutations by base pair changes of frameshifts in the genome of the strains used.

AHA - Ceramide complex for clear formulations

Alpha-Ceramidein a skin care ingredient has been developed by Campo Research and Development of Singapore. It is said to be an answer to the problem of formulating clear AHA-Ceramide systems for advanced skin care.

Facial cosmetics based on sour milk have been used since ancient Egyptian times. The main active ingredients are believed to be the alpha-hydroxy acid (Ahas) predominantly lactic acid and its derivatives. Ahas are naturally occurs substances isolated from a variety of sources, such as sour milk, sugar cane, tomatoes and other fruits. They perform a number of functions at the level of stratum corneum on the skin’s surface. There are many different kinds of AHA, and Campo says that some out-perform others. Intensive research has identified lactic acid, particularly the L- isomer, as one of the most effective forms. Campo’s L-lactic is isolated from tomatoes cultivated by hydroponics. Ceramides are found in the human skin and comprise 43-46% of the stratum corneum where they play a major role as a water retention barrier.

They include sphingosines and fatty acids, and are generally linked with sugars to form structures called glycosphingolipids. Six kinds of ceramides have now been identified in human skin. Campo’s Ceramide RW is a new complex of biologically engineered material, truly identical to human skin ceramides. It is a spin-off from the p34 cde2 (gene expression) re, especially on skin cancer proliferation and down regulation activities of some natural products, which has been carried out by Kampoyaki Research, Singapore. Since Elizabeth Arden’s Alpha-ceramide 4 steps hit the market in the summer of 1994, many cosmetics raw materials suppliers have been trying to produce a clear, colorless ceramide liquid base that is soluble in ethanol and water. These efforts have been unsuccessful except for a couple of vegetable ceramide powder types with poor stability. These turn into cloudy liquids from which it is virtually impossible to formulate a clear, colorless product.
