Aminoacil-tRNA sintetasas
• Une los aa al correcto tRNA
• 20 enzimas diferentes
• Requiere ATP

Reconstitution of the eIF4E/GST-4G1f/Pab1p Complex(A) Reconstitution with Pab1p immobilized on poly(A)-Sepharose resin. Pab1p bound to poly(A)-Sepharose resin was incubated with the indicated GST-4G1f proteins and eIF4E. Bound proteins were eluted with buffer containing SDS, resolved by SDS–PAGE, and visualized by Western analysis with the appropriate antibodies.(B) Reconstitution with eIF4E immobilized on the cap-analog resin 7mGDP-agarose. eIF4E bound to the resin was incubated with the indicted GST-4G1f proteins and Pab1p/poly(A)125 complexes. Bound proteins were analyzed as in (A).

If the dsRNA is incubated with GST-4G1f-459, Pab1p, and eIF4E (D), most of thedsRNA is linear although some circles are observed (lower left corner). In thepresence of GST-4G1f, Pab1p, and eIF4E (E and F), much of the dsRNA is in acircular conformation with a protein complex positioned where the ends of the RNAmeet. Differences in the apparent width of the RNA result from variation in the tipand imaging force used to generate each image

CODON INICIACION
Pu X X A U G Pu C C Pu C C A U G G
-3 +1 +4 -3 +1 +4

2 proteínas diferentes == N-terminal
== secuencias enteras

The autoradiograms show [35S]Met-labeled proteins synthesized in vitrofrom capped mRNAs that encodechloramphenicol acetyltransferase.(A) Initiation at AUG#1 generates anN-terminally extended protein,labeled preCAT. A suboptimal contextaround AUG#1 allows some ribosomesto scan past that site and initiateinstead at AUG#2. This leakyscanning produces a shorter protein,labeled CAT. (B) All mRNAs have asuboptimal context (U in position −3)around the first AUG codon and,except for the control in lane 1, amoderately stable base-pairedstructure between the first andsecond AUGs. The only variable is thedistance (n) between AUG#1 and thebase of the hairpin structure. Whenproperly positioned, the downstreambase-paired structure apparentlysuppresses leaky scanning (lane 4). Afull description of these constructsand the adjustments required for therabbit
