A photodiode array based spectrophotometer providesa nearly instantaneous measurement of the entireUV-Visible spectrum. For some applications and methods, this optical configuration offers tremendousadvantages in data acquisition and sample throughputcompared to a traditional monochromator-based spectrophotometer.
Diode array instruments use a reverse opticalgeometry where light of all wavelengths is passedthrough the sample. The sample absorbs some characteristic wavelengths based on its composition.Wavelengths of light not absorbed by the sample aretransmitted to the detection system. The transmittedlight passes through an entrance slit and is then dispersed onto a photodiode array by a grating.Simultaneous detection at all wavelengths throughoutthe UV and visible regions of the spectrum allowsnearly instantaneous display of an absorption spectrum.
Traditional, monochromator-based systems measurescan speed in nanometers per minute because only a single wavelength is measured before the mono-chromator must be moved and the next wavelengthmeasured. In contrast, acquiring data in a singlemeasurement allows an equivalent scan speed of54,600 nm/min.
Further, reverse-optical systems have no movingparts; therefore they provide superior wavelengthaccuracy and repeatability. These instruments alsoneed much less maintenance and offer much longerinterval between qualifications.
