Plan prezentacji
![Page 4: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/4.jpg)
Obiekt badań
• Jak uorganizowane jest ludzkie mtDNA
• Jego powiązania przestrzenne z aparatami transkrypcyjnym, translacyjnym, translokacyjnym i degradacji RNA
• Zachowanie mtDNA podczas podziału mitochondriów.

![Page 6: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/6.jpg)
• ~16.6 kb
• 22 tRNA molecules
• 2 rRNAs
• 13 mRNAs
• Both strands encode transcripts
• Dense packing
Human mtDNA – organisation
![Page 7: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/7.jpg)
Primary mitochondrial transcripts
• Policistronic transcripts
• Genes encoded by mtDNA lack regulatory sequences
• Common promotor region for transcripts from both strands
• In case of ATP6/8 and ND4/4L ORFs of two proteins are partially encoded by the same DNA sequence in different reading frame
Human mtDNA – information expression

mtDNA uorganizowane jest w foci po ~8 cząsteczek. Foci związane są z błoną mitochondrialną.

Ogólny schemat idei integracji funkcji
![Page 10: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/10.jpg)
Dlaczego to takie istotne?
Współskładanie kompleksów mitochondrialnych zależy od białek mitochondrialnych i jądrowych
![Page 11: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/11.jpg)
Współwystępowanie foci mtDNA i nowopowstałych transkryptów

S6 - białko rybosomalne z rybosomu cytoplazmatycznego
NAC – chaperon cytoplazmatyczny uczestniczący w translokacji białka przez błony mt
Tom22 – kompleks translokacyjny zewnętrznej błony
![Page 13: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/13.jpg)
• przesuwanie obrazów względem siebie
• dla każdej pozycji liczymy współczynnik korelacji Pearsona (-1, 1)
-1 korelacja ujemna
0 brak korelacji
1 korelacja dodatnia
• obserwujemy rozkład współczynnika w zależności od dystansu przesunięcia
W jaki sposób rzetelnie analizuje się takie obrazy? - propozycja

![Page 17: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/17.jpg)
Wnioski
• mtDNA uorganizowane jest w foci po 6-10 cząsteczek
• foci występują w pobliżu miejsc podczepienia mt do szkieletu MT
• podziały mt odbywają się w pobliżu foci, ale nigdy nie dzielą foci
• foci występują w miejscach, gdzie lokalnie zmniejszona jest ilość kompleksów oddechowych, co może chronić mtDNA przed wolnymi rodnikami
![Page 18: The functional organization of mitochondrial genomes in human cells](https://reader030.vdocuments.site/reader030/viewer/2022032605/56812b9b550346895d8fbd53/html5/thumbnails/18.jpg)
Wnioski jeszcze istotniejsze...
