By live cell imaging we seek to contribute to the understan-ding of the organization and dynamics of cells under various physiological and ecological conditions. In addition to con-ventional stereo and light microscopy, we apply ultraviolet and video microscopy combined with fluorescence tech-niques. Hence, we are able to observe small structures in living cells below conventional light microscope resolution that so far have only been visible in the electron microscope. In addition, we describe and analyse the mo-tility of cells and organelles as well as whole plants.

By correlation with electron micro-scopy and application of cryo-prepa-ration techniques, we investigate the following topics: • cytomorphology of plant cells and organelle motility

• stress reactions of plant cells

• carnivorous plants and their ecological environment

• eco-physiology of heavy metal plants and their use for re- mediation of polluted soil

Visualization of our biological objects at all levels of magni-fication is a prerequisite for teaching and for dissemination of our scientific results to the general public. In this context we produce educational films about specific techniques and biological issues.

By electron microscopy we study the ultrastructure of biolo-gical samples at various levels of complexity comprising tis-sues, cells, and their extracellular products and organelles. For this purpose we apply both scanning electron microscopy as surface-sensitive technique, and transmission electron micro-scopy as a means of visualizing the structures within cells of thin and ultrathin sections. Besides studying fine structures, we perform element analysis, imaging and mapping. Particu-lar emphasis is laid on sample preparation which comprises rapid immobilization of the living state by freezing of the samples at ambient or high pressure, and subsequent steps of cryo-processing. Increasingly, our EM studies profit from techniques aiming at the correlation with light microscopic observations. Our interest is focused on: • structural alterations of organelles, cells, tissues, organs and organisms in the course of development and regression

• analysis of the elemental composition of cells, tissues, org- anisms and abiotic materials, especially where a merge of basic and applied research is indicated

• studies of cephalopod adhesive systems, cell-carrier con- structs and cuticular structures in arthropods

These electron microscopic studies contribute to a better understanding of function, ontogenetic pro-cesses and phylogenetic relationship of cells, tissues and organisms. They are part of world-wide efforts in maintaining the ecosystem. At the same time, they inspire and accompany applications for industry.
