CUSTOMER SPOTLIGHT TENGION - NEO-ORGAN / TISSUE DEVELOPMENT

Tengion was founded in 2003 to develop and commercialize a medical breakthrough that harnesses the regenerative power of the body and enables the regeneration of neo-organs and neo-tis-sues from the patient’s own healthy cells (autologous cells). This advance has the potential to augment, repair or replace impaired tissues and organs without the need for transplants from organ donors and without the risk of rejection from the body’s immune system. There are significant limitations to the current therapies for treating a range of tissue and organ abnormalities, and there is a clear need for approaches that are safer, more effective, and cost-efficient. Tengion is working with the Food and Drug Administration to begin clinical trials of its first product, a neo-bladder, by the end of 2006. The company has extensive data demonstrating proof of principle in large animals.Tengion’s technology has the potential to offer several major advantages over current best practice. MCO-5M & 18M APPLCIATIONS

Our MCO 5M and MCO 18M incubator are the choice of many researchers in these areas of research, but it’s important that you understand what require-ments your customer has for the tri gas incubator they are purchasing.

Typically, our MCO 18M will reach 2% O2 levels within 20 minutes of a door closing, for some re-search, this is not fast enough and we will need to modify the incubator to meet their requirements. We do this by adding larger solenoids and heavy duty tubing to allow the inlet pressures to be increased. Please specify this requirement with customer service when quoting this model.

The cost of this modification is $500.

NiTrOgEN USagEMany customers who purchase tri gas incubators for low oxygen consumption usage are surprised at the amount of nitrogen that these incubators consume.In cases where they are used in IVF applica-tions, the typical oxygen setting is from 5% to 5.5%. Since room air is approximately 20% oxygen, the incubator has to inject nitrogen to displace the oxygen in the chamber. This takes a lot of nitrogen, often as much as a tank per day. For Hypoxia, the consumption can become even higher.

An alternative to the big tanks of nitrogen that have to be replaced often, is to utilize a liquid nitrogen dewar to supply the nitro-gen. LN2 off gases as the liquid evaporates. By putting a regulator on the dewar, it can

supply a constant source of nitrogen often at far less cost than tanks of Nitrogen gas. If running out of liquid nitrogen is a concern, remember that our tri-gas incubators have a built-in gas switching system, so they can still keep a back-up dry nitrogen tank in the reserve side.

iN-ViTrO FErTilizaTiON (iVF) appliCaTiONS

All our incubators come with a #6 rubber plug to block the access port at the rear of the chamber. Some IVF users are concerned about the fumes produced by rubber. Although there doesn’t seem to be any adverse ef-fects, the fumes produced by rubber are considered toxic to embryos. The easy solution to this is to purchase sili-cone stoppers for these applications. These are available at the medical supply houses for a couple of dollars.

While on the subject of toxic fumes, if a gasket ever becomes loose or torn, NEVER use regular silicone caulk to seal or fix the gasket. Silicone produces acetic acid fumes which are extremely toxic to embryos. If a sealer needs to be used, use RTV type silicone and allow to cure for 48 hours before using the incubator again.

Most cultures are incubated at 5% CO2 with the balance being ambient air.

Typically, with this 95/5 mixture the O2 in the chamber will be approx.19.9% O2. This is arrived at taking 20.95% of the 95% air.This has basically been the standard formula in the clinical and research laboratory.This generally accepted method of 5% CO2 and 95% air has caused many investigators and (salespeople) to overlook the primary purpose of a laboratory incubator. To create an in vitro model of the in vivo condition of the cell culture or organism under investigation.There is much evidence that supports the theory that reduced oxygen tension in vitro increases the cellular activity of cell cultures including melanomas, embryos, macrophages, fibroblasts, and murine hybridomas.

ULT ConventionallyInsulated Freezers

SANYO MDF-Series ultra-low temperature freezers maintain internal temperatures as low as -86°C (-123°F). All models use SANYO uniquely designed compressors for ultra-low temperature applications. Manufactured with foamed-in-place insulation, they are ideally suited for use in laboratories and hospitals for long-term preservation and storage of blood, speci-mens and components, as well as materials testing.
