What is the Laminar airflow ?Q:
27
Vertical & Horizontal Laminar Flow
vsClass I
Laminar airflow exists when molecules of air move at the same speed in a parallel direction. The laminar air velocity must be in the range 0.35 to 0.55 m/sec (U.S. Federal Standard 209 A/B and British Standard 5295). This laminar flow prevents air and suspended particles from changing direction (turbulence).

HOW SHOULD BIOLOGICAL SAFETY CABINETS BE INSTALLED?
A’ Code of Practice for the Prevention of Infection in Clinical Laboratories and Post Mortem Rooms’ (The Howie report HMSO) provides recommendations on the siting of safety cabinets:
1.The length of ductwork between the cabinet and the exhaust outlet should be as short as possible.
2.The cabinet must be placed away from doors or windows which may cause draughts.
3.The cabinet must be placed in such a position that no-one walks in front of it, or behind the operator when in use.
52
24/07/59
27
4.The cabinet should not be placed in a small room (<30 cubic meters) unless it has adequate ventilation.
5.In any room containing an exhaust protective cabinet, there should not be less than 6 changes of air per hour.
6.The exhaust system should ideally be mounted on the roof of the building, but may be mounted in a wall or window.
7.The exhaust outlet should be located to avoid the effects of wind and to ensure good dispersal of exhausted air.
53
Recommendation for minimum distances for avoiding disturbance to the safety and its operator
(a) Separation of an undisturbed zone around a safety cabinet from traffic routes.
(b) Spacing when the same operator uses a safety cabinet and a bench top opposite, or where occasional traffic only is anticipated.

1. Switch on the cabinet and apply suction to the air samplers and start the spinning disc and then allow the potassium iodide to feed on to the centre of the disc.
2. Place the filter membrane from one sampler in the palladium chloride solution, within 30s to 45s the membrane will become saturated and any potassium iodide particles will become visible as brown spots. Repeat this procedure with the filter membranes from the other air samplers.
79
Test procedure
3. Examine each filter with a x 10 magnifier and count the number of brown spots on the filter membrane
4. Repeat the procedure as appropriate for the width of the aperture as described in D.2.2.
80
24/07/59
41
Calculate the number of potassium iodide particles liberated, N, using the following equation:
N = (3.1 x 107 x M)
where M is the volume of potassium iodide solution
dispersed by the aerosol generator (in ml);
3.1 x 107 is a constant derived from the droplet size, the
sampling flow rate and the speed of rotation of the disc.
81
35
Calculation of the protection factor
Then calculate a value for the protection factor, PF, separately from each filter membrane using the following equation:
PF = NV/104n
where V is the sampling flow rate n is the number of spots on the filter
membrane.

What is a HEPA filter ?Q:
85
Principle of HEPA filter
A HEPA (High Efficiency Particulate Air) filter is a fiberglass filter sealed with an epoxy resin into an aluminum frame. Each filter is tested to ensure a stated filtration efficiency (usually 99.997% or better) using either the di-octyphthalate test (U.S. Standard 209B) or the equivalent sodium flame test (BS 3928). A HEPA filter is sometimes known as an absolute filter.

Installation of Ultraviolet (UV) lamps is not permitted in biological safety cabinets at the NIH.
The NIH, CDC, NSF/ANSI, and the American Biological Safety Association all agree that ultraviolet (UV) lamps are not recommended nor are they necessary. NSF Standard 49, the industry testing standard for all biohazard cabinetry, does not provide any performance criteria for UV lighting and specifically states in Section 4.24.2 “UV lighting is not recommended in Class II (laminar flow) biohazard cabinetry.”
92
24/07/59
47
It is the current opinion of the American Biological Safety Association (ABSA) that UV lights are not recommended for use in Biological Safety Cabinetry.
Retrofitting any equipment (e.g. UV lights) into a biological safety cabinet may alter the air flow characteristics of the cabinet and could invalidate any manufacturer warranty and is not recommended.
93
The Center for Disease Control (CDC) and the National Institute of Health (NIH) agree that UV lamps are not recommended nor required in Biological Safety Cabinets (BSCs).

68
Keep Warm Facility
• Control temperature of chamber at and of cycle at 65 c
• Allow the processed media in the chamber to remain liquid
• No need of extra water bath
24/07/59
35
69
Temperature and Pressure Monitoring
• Controlling by pressure provides greater temperature stability than controlling by temperature
• Important for certain media which are sensitive to temperature overcooked.
• Typical accuracy is 0.5 c
Dynamic In-Chamber Cooling
• Two methods of cooling
• In-chamber cooling pushes air through the chamber via fine air inlet filter
• In chamber cooling can assist in drying certain load types
• Helps to speed up cooling times
• Unique feature
70
24/07/59
36
Load Sensed Process Timer
• Armour-flex probe
• Tougher than plastic or stripped-end probes
• Does not begin sterilization timer until load and drain reaches sterilizing temperature
• Does not release door until load is at safe temperature
71
• Used for Media cycles
• Speeds up cooling process
• Maintains an over-pressure in chamber
• Compressed air enters chamber through a fine air filter
• No-one else offers this as a standard feature 72
