SDA Processing

A special Statistical Deconvolution Algorithm (SDA) combinedwith patented TR-LSC background reduction provides unprece-dented sensitivity for finding the smallest peaks under low levelcounting conditions. SDA is a statistical image enhancementtechnique applied to radiochromatography. Unlike smoothing, inwhich peaks are typically broadened and sometimes completelylost by being spread into the baseline, SDA provides the ability todiscern if a “blip” on the baseline is real and should be integrat-ed and examined.

Full Connectivity for Increased Productivity

Both ProFSA and ProFSA Plus provide the versatility to connect to third party software and support devices. Full control of gradientHPLC pumps is now possible as well as connection to auto-injectors and fraction collectors. More than one HPLC station can be operated by a single PC. Data from liquid scintillation analyzers can be input either directly or via disk transfer to enable datacomparison and reporting. Homogeneous

Homogeneous flow cells with volumes from 100 µL to 5,000 µL provide the lowest backgrounds, highestsignal-to-noise ratio, and highest sensitivity of all flow detection schemes. Liquid scintillation cocktailsuch as PerkinElmer’s Ultima-Flo™ environmentally safe cocktail is mixed with the column eluate in ratiosas low as 1:1 without compromising performance. Our exclusive MCA technology eliminates interferencedue to background, chemiluminescence, and the effects of chemical quenching by spectral stripping.

Microvolume Homogeneous

Microvolume homogeneous flow cells with minimal dead volume fittings and connections are available inboth NarrowBore and MicroBore format and are recommended for FSA/LC/MS applications. NarrowBoreflow cells are available in the range 15 µL to 99 µL while for MicroBore the available range is 3 µL to 14 µL.The combination of microvolume technology with patented TR-LSC background reduction results in bestdetection, thus separating low activity sample components from background. Peak resolution is improved

five-fold which gives enhanced peak separation, elimination of peak tailing and ensures the identification of all sample components.

Heterogeneous

Heterogeneous flow cells with volumes from 50 µL to 300 µL are packed with fine grains of solid scintillatingmaterial within the coiled Teflon® tubing of the flow cell. Heterogeneous detection eliminates consumablecosts, reduces waste, and, because the sample is not mixed with liquid cocktails, allows total samplerecovery. High performance flow cells with volumes of approximately 35 µL, 100 µL, 200 µL and 400 µLoffer heterogeneous detection for applications that require high back pressure and high flow rates. Theseflow cells withstand pressures of up to 2,000 psi. The unique packed cartridge design makes cartridgereplacement easy. This may be necessary if the scintillating material becomes contaminated.

Gamma

Gamma flow cells for low and high energy gamma emitters are ideally suited for the high performanceflow scintillation analysis of samples labeled with low and high energy gamma emitting nuclides. The

Gamma-C flow cell is ideal for low energies below 70 keV such as 125I and 109Cd with counting efficiencybetter than 85% for 125I. The Gamma-B flow cell is suited for mid to high energy nuclides above 70 keVsuch as 99mTc, 131I, and 201Tl. Since the scintillator is in the window, there is no need for liquid cocktail,

thus eliminating cocktail consumption and reducing waste generation.

PET

The PET flow cell is especially suited for the analysis of radiolabeled pharmaceuticals and their metabolitesin Positron Emission Tomography (PET) applications. This flow cell provides PET centers with selective,highly sensitive detection of positron emitters such as 11C, 13N, 15O, and 18F. A unique flow cell designincorporates optically isolated bismuth germanate (BGO) scintillating windows resulting in signal-to-noiseratios unmatched by any other PET flow cell.

Homogeneous Counting—For High Sensitivityand High Efficiency

Homogeneous methods provide the lowest backgrounds, highest sensitivity, highest efficiency andthe best MDA for all isotopes. The homogeneous flow cell consists of Teflon tubing which is coiledbetween two scintillating windows. Teflon tubing is chosen because of its inertness and transparencycharacteristics to maximize light output yielding the highest counting efficiency. The scintillatingwindows form the basis of TR-LSC to achieve unprecedented low backgrounds and the highestsignal-to-noise ratio.

With homogeneous techniques, the column eluate is mixed with the liquid scintillation cocktailand passed through the flow cell. Homogeneous techniques are preferred to heterogeneous tech-niques for low energy isotopes such as 3H and molecules with high molecular weight. Display andanalyze radiolabeled samples using Spectrum Analysis to accurately set energy windows and optimize counting conditions. Quench correction curves are automatically generated to obtain themost accurate DPM results.

Heterogeneous Detection—Reduces Wasteand Recovers SampleFor applications that require total sample recovery, PerkinElmeroffers a wide range of heterogeneous flow cells. Now, you canreduce radioactive waste with cocktail-free counting for a widerange of applications.

Your Choice of Solid Scintillators

Choose from a variety of solid scintillators compatible with manyapplications. Particle sizes vary from 35 µm to 250 µm. Smallparticle sizes mean maximum light output and high countingefficiency. Choose from cerium-activated lithium glass, cerium-doped yttrium silicate, and a new advanced proprietary scintillator,SolarScint™.

High Performance Flow Cells for Normaland Micro-Volume Applications

PerkinElmer’s high performance flow cells are packed with scintillating particles for normal HPLC and micro-volume HPLC.For mass spectrometry applications where peak resolution and minimizing dead volume are important, this specially constructed flow cell provides less than 1 µL dead volume.

In addition, the glass cartridges are replaceable. All packed scintillator flow cells, whether in Teflon tubing or glass cartridges, will eventually become contaminated. Replacing cartridges is more cost effective than replacing the entire flow cell.

PerkinElmer also offers calcium fluoride solid scintillator cells through special order.

Yttrium Silicate

Yttrium silicate is probably the most efficient of the solid scintillatorsbut it is also the one most prone to contamination and it produces luminescence at low pH. Contamination problems are due to the polycrystalline nature of the material. Yttrium silicate is TR-LSCcompatible resulting in low backgrounds and high signal-to-noise ratio.

Lithium Glass

Lithium glass is the most rugged solid scintillator and is extremely resistant to contamination. It can withstand particularly rigorous decontamination procedures. It gives acceptable but lower counting efficiencies for 14C than either yttrium silicate or calcium fluoride, butis rarely used for 3H measurement. However, it is the fastest to darkadapt and the least affected by extremes of pH.

SolarScint

SolarScint’s inertness means compounds with complex matrices will notabsorb to the scintillating particles. Peak broadening, contamination andmemory effects are reduced to a minimum. With small particle sizes of35 to 60 µm, efficiency for 14C is better than 65%. SolarScint is especiallygood for inositol phosphates.

Gamma Flow Cells with the Performance of aDiscrete Gamma Counter

PerkinElmer’s Gamma-C and Gamma-B flow cells are ideally suited forhigh performance flow scintillation analysis without cocktails of sampleslabeled with low and high energy gamma emitters.

The Gamma-C flow cell incorporates europium-doped calcium fluoridescintillating windows to yield better than 85% efficiency for 125I and other“soft” gamma emitters.

The Gamma-B flow cell for mid to high energy gamma emitters such as111In, 22Na, and 99mTc uses special BGO scintillating windows. The highspecific gravity of the Gamma-B flow cell nears and sometimes exceeds theperformance of a discrete gamma counter using a 3-inch NaI detector.

Ultima-Flo™ Safer Flow Detection Cocktail LinePerkinElmer’s biodegradable cocktails for use in flow scintillationanalyzers, the Ultima-Flo family, were developed to addressthe concerns of environmental safety. These environmentallysafer cocktails satisfy all of the applications covered by theoriginal, classical solvent-based Flo-Scint™ cocktails. The highloading capacity of the Ultima-Flo cocktails means less cocktailrequired, longer residence times and higher sensitivity. Eachcocktail has low viscosity and non-gelling properties—even athigh sample loads—ensuring trouble free counting and lowerback pressures. Ultima-Flo cocktails resist quench, providinghigher counting efficiencies and more counts per peak.

Ultima-Flo MUltima-Flo M is formulated for multipurpose flow countingapplications. This safer cocktail has a high sample acceptancefor a wide range of dilute HPLC eluents, and methanol and acetonitrile gradients. Ultima-Flo M has low viscosity, uniquerapid mixing properties and is resistant to chemiluminescence.

Ultima-Flo APUltima-Flo AP accepts gradients up to 2.0 M ammonium phosphate with fast and easy mixing. Ultima-Flo AP demonstrates high counting efficiency and quench resistance for a wide variety of sample types.

Flo-Scint IIIFlo-Scint III is designed to work with methanol and acetonitrilegradients as well as moderately buffered solutions, includingphosphates. Flo-Scint III has good sample loading capacity, lowviscosity and high counting efficiency.

Flo-Scint IVFlo-Scint IV is designed to accept high molarity ammoniumformate and ammonium phosphate gradients (up to 2 M) andalso accepts concentrated salt solutions. Flo-Scint IV is resistantto chemiluminescence, has high counting efficiency, low viscosity and high sample loading capacity.
