Structure of the GLP regulation
• Test and reference items• Standard operating procedures• Performance of the study• Reporting of study results• Storage and retention of records and
materials
Scopeprinciples of GLP should apply to :
* non-clinical safety testing (human health and environment)
*test items in pharmaceuticals, pesticides, biocides, cosmetic products, food and feed additives
*test conducted in laboratory, field or greenhouse plants
-for purposes of licensing
Area of expertise
• Physico-chemical testing• Toxicity studies• Mutagenicity studies• Environmental toxicity studies on aquatic
and terrestrial organisms• Studies on behaviour in water, soil and air
bioaccumulation• Residue studies• Studies on natural ecosystems• Analytical and clinical chemistry testings• Other studies (to be specified)
GLP and ISO/IEC 17025• ISO/IEC 17025 covers many GLP
requirements• Not sufficiently covered by ISO/IEC17025

ResponsibilitiesManagement :
- Ensure that the principles of GLP are complied with
Study director :He has the full responsibilities for the overall conduct of the study and for its final report. He should:- Approve study plan and any amendments
- Ensure availability of study plan to quality assurance unit and study personnel
- Ensure that study procedure are followed
ResponsibilitiesStudy director :
- Assess and document impact of any deviations on the integrity and quality of the study
- Indicate in the final report the extend to which the study complies with GLP
- Sign and date the final report
Principal investigator :- Ensure that the delegate phases of the study are conducted in accordance with GLP- Make of declaration of GLP conformity concerning these phases
Responsibilities• More information on responsibilities of
study director and principal investigator are given in consensus documents (number 8-13)
Study personnel : - Follow the instructions in study plan and standard operation procedures- Communicate any deviation to the study director- Record raw data promptly and accurately
• Master schedule :• Up-to-date information for on-
going studies• List of archived studies
Management Tools
Remarks
Management should understand and support the application of GLPA GLP test facility can be a (sub)division in a company QA unit is not a secretariat
Principle of Quality Assurance Programme• The test facility should have a document
quality assurance programme with designated personnel, responsible directly to management
• QA Programme in form of standard operating procedure(s)
• Management should ensure that the quality assurance responsibility is performed in accordance with the principles of GLP
QA Personnel
• QA Personnel should be familiar with the tests but not involved in the conduct of the study to be inspected
• QA should be involved during validation of computerized systems
• QA personnel can exercise other activity• QA function can be outsourced (consultant)
• In this case a free access to the test facility should be ensured
QA activities• Verification of study plan :
Verify that the study plan contains the information required by the principles of GLP
• Study inspections :Verify that all studies are conducted according to GLP
• Inspection of final report :Verify that methods, procedures and observations are correctly and completely reported
• Any inspection result is to be reported in writing to management and study director
QA reports
All activities should be documented• Inspection of test facility, on-going
studies and final reports result in a documented inspection report
• Study director or management have to propose corrective action to the deviations mentioned in the report
• QA check the application of corrective action
• QA reports are finally archived separately from the studies
QA reports
• For each GLP study the QA unit should prepare and sign a statement, to be included in the final report, which specifies:• Types and dates of inspections specific
to the study • Inspected phases of the study• Dates inspection results were reported
to management and study director• The statement would also serve to
confirm that the final report reflects the raw data
QA and multi-site studies
• In the case of multi-site of studies, the responsibility of QA unit at each site should be clearly defined
• The lead QA unit is responsible for the statement in the final report
Comments
QA unit should have enough authorityQA is responsible for a strict application of GLP principlesQA should know the GLP principles and consensus documents Ideally the QA unit is the long arm of the GLP inspectors
Principles of GLP
• Facilities• Apparatus, materials and reagents • Test and reference items• Archive
Facilities
• Suitable size, construction and location to minimize disturbance
• Adequate degree of separation of the different activities
• Storage rooms separated from animal rooms
Facilities for test and reference substances
• Separate room for receipt, storage and mixing of test item
• Adequate storage condition (temperature)
• Maintain record of temperature control, movement of test and reference substances
Archiving facilities
• Ensure a secure storage and retrieval of documents, test items and specimen• More than one room my be necessary
• Archive should protect content from untimely deterioration (fire ,water,…)
• Restricted access to authorized personnel • normally restricted to archivist and
management
Apparatus
• Suitably located and of appropriate design
• Periodically inspected, cleaned, maintained and calibrated• standard operating procedure• records to be maintained (log-book)
• Computerized systems should be validated • HPLC, GC systems: simple validation• LIMS (computer network): complex
validation
Chemicals, reagents
• Labelled to indicate identity, expiry date and specific storage condition• maintain documentation on chemicals• expiry date may be extended on the
basis of documented criteria
Test and reference items• Maintain records on characterisation,
date of receipt, expiry date, quantities received and used in studies
• Identification should be documentedidentity, batch number, purity, composition
• Stability under storage and test condition should be known
• Sample for analytical purpose of all batches should be archives (except short-time studies)
• Mechanism to verify identity of test item if supplied by a sponsor (i.e. visual identification: color, aspect)
To be retained in the archives
• Study plan, raw data and final report• Test item, specimen (organs, slides)• Records of all inspection by the QA unit• Records of personnel documentation• Validation documents for computerized
systems• Historical file of all SOPs• Environmental monitoring records
(humidity, temperature)
Comments
• In the case of “stable” chemical, give reasonable expiry date
• Require from sponsor enough information on test item (batch, expiry date)
• Cross-contamination (test substance found in reference animals) is a serious problem
• Ensure traceability for every process
Coverage
SOPs should, at least, be available for:• Test and reference items• Apparatus, computerized systems,
chemicals• Biological test system• Planning of studies, reporting• Archiving• Quality assurance procedures• Elaboration, handling and revision of
SOP
Handling of SOPs
• SOPs should be revised to stay actual• Old SOPs should be eliminate from
workplace• Study personnel should understand the
content of SOP• No correction on SOP at working place
Comments
• Elaboration of SOPs need a lot of resource for a new test facility
• SOP should contain enough details to follow up the activity
• SOP should not contain too much not relevant details, to avoid deviation with the praxis
• Electronic SOPs are possible but not very convenient
• Handling of SOP should be centralised• A periodic control of SOPs is recommended
Study plan
• For each study a written plan should exist prior to the initiation of the study
• Be approved by dated signature of the study director
• Eventually be approved by management and sponsor
• Planned modification of the study in from of amendment
• Amendment to study plan have to be justified and approved by study director and maintained with the study plan
