5 mL Saturated Yeast Culture

Filter through 0.22 mm filter

Quench the cells using 2mL (a) ice cold PBS

Or (b) 80% MeOH (-80 °C)

a) Transfer filters to a tube containing 1.25 mL 80% MeOH and 1.25 mL

CHCl3

b) Add 0.5 mL ice-cold water

Separate aqueous and chloroform layers and

dry them

Sample Preparation procedure

Aim is to compare the two quenching methods using aqueous extracts

Bruker Avance III 600 MHz NMR ns = 16 sw = 16 ppm td = 32768 si = 32768 T = 298 K
