Imagineifpeoplecouldknow,inrealtime,whatpollutantstheywerebeingexposedtoandwhatthosepollutantsweredoingtotheirbodies.

AcompetitiontobuildamobiledevicethatwoulddojustthatwasannouncedJune6.

Thechallenge,issuedtotechsavvyinnovators,istocreateapersonalsensorsystem,smallenoughtobewornorcarried,thatmeasuresbothairpollutionandthephysiologicalresponsesoftheindividualcarryingit.Thosewhoacceptthechallenge,whichincludes$160,000inprizemoney,arealsorequiredtohaveaplanformakingtheresultingdataavailable,withoutrevealingindividualidentities,toresearchers,publichealthinstitutions,andotherinterestedparties.

TheMyAir,MyHealthChallenge,partoftheChallenge.govprogram,wasannouncedduringahealthtechnologyforumbyNIEHS/NTPDirectorLindaBirnbaum,Ph.D.,andGlennPaulson,Ph.D.,scienceadvisorattheU.S.EnvironmentalProtectionAgency(EPA).TheChallenge.govprogramintroducesanewvehicleforpursuinginnovation,wherebyagenciespayforresultsratherthanfundingresearch.

“Grantsfundtheproposalsthataremostlikelytosucceed,butchallengesgivetheprizetosomeonewhohasalreadyachievedsuccess,”saidBirnbaum.“Thesechallengesleadtoattractingoutsideinvestmenttoanareathat’sbeenafederalpriority.”

Whileasuccessfulsensorwillbevaluabletoindividualsandspecificcommunitiesconcernedaboutpollutants,thecollectivedatawillbeimportanttoscientistswhostudytherelationshipsbetweenpollutantsanddiseases.Whilelinkstomanydiseasesarealreadyknown,scientistsfacehugebarriersindeterminingwhatpollutantsanindividualorgrouphasbeenexposedto,when,andatwhatlevels.

“Ifwecandetectwhatapersonisexposedtointheirenvironmentandthenmeasurehowtheirbodyresponds,wecanbegintounderstandhowtopreventcertaindiseases,”Birnbaumsaid.

Thedevicebeingsoughtwouldimproveonexistingfixedsensors,suchasthosethatmonitoremissionsinafactory,bybeingportableandaddingafeaturethatmeasuresphysiologicalresponses.

Measuring the body’s response to pollutantsWhilethepossibilitiesformeasuringphysiologicalresponsestopollutantsareinfinite,theinitialdevicewillmostlikelymeasurecardiacandrespiratoryresponses,saidNIEHSSeniorAdvisorAllenDearry,Ph.D.,whomanagestheMyAir,MyHealthChallenge.Thehealthmeasurementsmayincludethingssuchasbloodpressure,heartrate,somemeasureoflungcapacity,andgalvanicskinresponse,ameasureoftheelectricalconductivityofskin,whichcanchangeinresponsetostress.

Devicesenteredinthecompetitionmustbeabletomeasureairbornepollutants,includingchemicalsand/orparticulates.

Engineers,peoplefamiliarwithfixedsensors,andmobilesoftwareexpertsareamongthoseexpectedtocompeteasindividualsorteams,butthecompetitionisopentoanyoneintheU.S.ThedeadlineforentriesisOctober5.

Asmanyasfourfinalistswillreceive$15,000eachinphaseoneofthecompetition.Thewinnerofphasetwo,whichrequiresasmall-scale,proof-of-conceptproject,willbeawarded$100,000.

Inadditiontohealthandenvironmentalresearchgroups,thedevicewillbeusefultocommunitygroupsconcernedaboutlocalizedpollutionandevenindividuals,saidNIEHSprogramadministratorDavidBalshaw,Ph.D.,whoisinchargeoftechnicalinputforthechallenge.“Thisislike‘FieldofDreams,’”headded.“Webelieveifwemakeit,theywillcome.”

TheMyAir,MyHealthChallengeissponsoredbyNIEHS,theEPA,andtheU.S.DepartmentofHealthandHumanServicesOfficeoftheNationalCoordinatorforHealthInformationTechnology.TheannouncementwasmadeataforuminWashington,D.C.,sponsoredbytheHealthDataInitiative,apublic-privatecollaborationthatencouragesinnovatorstodevelopandusehealthdataapplicationstoimprovemedicaloutcomes.

Sustainability and health“Sustainabledevelopmentisawidelysharedgoal,andourmessagehasbeenthathumanhealthisbothadriverofsustainabledevelopmentandanindicatorforjudgingsuccessorfailure,”saidNIEHSSeniorAdvisorforPublicHealthJohnBalbus,M.D.,whohasledNIEHSeffortstoensurethathealthwascentraltoconferencediscussionsanddecisions.

“Weknowthatmakingtheplanethealthiergoeshandinhandwithmakingpeoplehealthier,”Balbusadded.“Makingthatgoalarealityisourultimatemission.”

Ininitiallysettingthegoalofre-energizingglobaldevelopment,andinasustainableway,theU.N.createdaframeworkfordecisionsaroundseventopics—jobs,energy,cities,food,water,oceans,anddisasterreadiness.

Althoughhealthdidnotmakethelist,itisinextricablylinkedtoeveryissue,saidBalbus,whohelpedlaunchanumberofactivitiestomakethoselinksexplicitandprovideguidanceabouthowtoincorporatehealthintoactionablegoalsfortheworldcommunity.

Behind the scenes with WHOInpartnershipwiththeWorldHealthOrganization(WHO),NIEHSsupportedameetingofexpertsinGenevawhoproducedsixbriefingdocumentsthatmadethecaseforhumanhealthasacriticalelementoftheissuesbeingdiscussed.Thebriefingdocumentsalsodevelopedasetofindicators,basedonhealthoutcomes,formonitoringprogresstowardsthegoals.

Thebriefingpaperonenergy,forexample,notedthatnearlytwomilliondeaths,annually,areduetohouseholdairpollutionfromstovesfueledbycoalandrudimentarybiomass.About1.3milliondeaths,peryear,areattributedtoambientairpollutionincities.Additionally,insomedevelopingcountries,overone-halfofhealthcarefacilitieslackreliableelectricity,withsomehavingnoelectricityatall.

Thebriefingpaperthensetoutcorehealthindicatorstobeusedtomonitorprogressinsustainableenergyaccess.Forexample,reducingratesofdiseaseattributabletoindoorandoutdoorpollutionwouldprovideevidenceofachievementtowardsthegoalofmovingtowardssustainableenergy.Anincreaseintheproportionofhealthcarefacilitieswithreliableelectricitywouldbeanothermeasure.

CommunityoutreachandengagementveteranAndreaHricko,oftheUniversityofSouthernCalifornia(USC),deliveredapresentationMay24atNIEHS,focusingoneffectivecollaborationbetweenresearchersandcommunitymembers.

“ThecommunityisconcernedaboutenvironmentalhealthrisksrelatedtointernationaltradethroughtheportsofLosAngelesandLongBeach,”Hrickoexplained.“ThesearethelargestportsinAmericaandareoftendescribedastheeconomicengineofSouthernCalifornia,with40percentoftheentirenation’sgoodstransportedthroughtheirfacilities.”

Inhertalk,titled“RespondingtoCommunityConcernsonEnvironmentalPublicHealth:AdvancingtheScientificResearch,Translation,andPolicyAgenda,”Hrickodetailedtheprogressthathasresultedfromtheeffectiveengagementofresearcherswithcommunitymembers,butstressedthatmuchworkremainstobedone.LiamO’Fallon,programanalystintheNIEHSDivisionofExtramuralResearchandTraining(DERT)atNIEHS,hostedthepresentation,whichwaspartoftheDERTKeystoneScienceLectureSeminarSeries.

Positive outcomes of community involvementCommunity-engagedresearchhasledtoseveralnotablesuccesses,suchastheinclusionofresearchfindingsintoenvironmentalimpactstudiespreparedforport,railyard,andfreewayexpansionprojects;changesintheidlingrulesfortrucksgoinginandoutoftheport;andeducationofCalifornialawmakersonthehealthbenefitsofprohibitingnewschoolsfrombeingsituatedwithin500feetofamajortransportationcorridor,whichresultedinastatelaw.However,giventheprojectedgrowthoftheports,muchworkremainstobedone.Forexample,newschoolscannotbesituatedclosetoamajorhighway,yetthereisnorestrictiononneworexpandedhighwaysorrailyardsbeingbuiltnexttoexistingschools.

Hrickonotedthatsomeoftheadvantagesofcommunity-academicpartnershipsincludeincreasedunderstandingof,andconnectionto,thesciencebycommunitymembers;abetterawarenessofthesocialjusticeimplicationsbyacademicsintheirresearch;andmoreeffectiveadvocacycampaignsbycommunitygroupsthathaddevelopedthenecessaryskillstoexplainthescientificbasisoftheirconcernstopolicymakers.Hrickoconcludedhertalkbyobservingthatthesuccessfulmodelofcommunity-academicpartnershipandoutreachthatshehelpeddevelopatUSCisjustoneofmanyeffortsthatarepartoftheNIEHSPartnershipsinEnvironmentalPublicHealthnetworkthathavebeenwellpositionedtoaddressotherenvironmentalhealthconcerns,suchastheGulfoilspillandhydraulicfracturing.

NIEHS town meetings spur involvement

HrickodetailedhowtheCenterisanationalleaderinresearchonthehealtheffectsofairpollution,andshesaidthatitsoutreachprogram’sfocusongoodsmovement,ortransportofcontainerswithproductsinsidethem,grewoutofcommunityconcernsraisedata2001USC-NIEHStownhallmeetinginLosAngeles.Atthemeeting,residentsspokeabouttherapidgrowthattheports,andtheconcomitantpollutionfromthetrucksandlocomotivestransportinggoodsarrivingfromAsia.Asoneexampleoftheimpactofincreasedlevelsoftransportation,Hrickodescribedaschoolneararailyardwhere500trucksperhourpassbyandwhere28percentofthestudentsalreadyhaveasthma.Shenotedthattherailyardisproposedforexpansion,despitetheoppositionoflocalresidents.

HrickoexplainedthatoneapproachusedbytheCenter,toensurethatcommunitymembersareactivelyinvolvedinairpollutionresearch,isthecreationofneighborhoodassessmentteams,orA-Teams.TheseareteamsofUSC-trainedcommunitymemberswhotrackdata,suchasthenumberofbigrigtruckspassingaschoolinagiventimeperiodandtheconcentrationofultrafineparticlesmeasuredusingreal-timemonitoringdevices.A-Teammembersareoftenparentsofchildrenaffectedbyasthmaandarechosenbasedontheircommunityleadershipcapabilities.TalentedA-Teammembershavegoneontoserveaspermanentstaffmembersonprojects.

OldenservedasdirectorofNIEHSandNTPfrom1991to2005,whenhelefthisleadershiproleandbecamealeadresearcherintheLaboratoryofMolecularCarcinogenesisMetastasisGroup.In2008,heacceptedanofferfromtheCityUniversityofNewYork(CUNY)tobefoundingdeanofthenewSchoolofPublicHealthatHunterCollege,auniqueprogramwithanurbanpublichealthfocus.

Oldenledthedevelopmentofthecurriculumandrecruitmentof26tenure-trackfaculty.Inearly2011,Oldenmovedwiththeschoolfromitstemporaryhometoaneweight-story,147,000-square-footgreenbuildinginEastHarlem.Lastsummer,theCouncilonEducationforPublicHealth(CEPH)announceditsaccreditationoftheCUNYSchoolofPublicHealth(SPH)atHunterCollegeforafive-yeartermextendingtoJuly1,2016.

Fromtheoutset,OldensawhisroleatCUNYasathree-yearcommitmenttogetthenewschoolunderway.“Oncethetransitiontoournewlocationiscomplete,mytask[hereinNewYork]willbeover,”Oldensaidwhenaccreditationwasannounced.

AccordingtoastoryabouthisappointmentsintheJune8issueofInsideEPA,Oldenwilloverseeeffortstorevamptheagency’sIntegratedRiskInformationSystem(IRIS),whichproducesmanyofEPA’smostinfluentialandsometimescontroversialriskassessments.Industryhascomplainedthattheassessmentsareoverlyconservative,publichealthadvocateshavecomplainedaboutdelays,andlastyeartheNationalAcademyofSciencesissuedacriticalreportoutliningrecommendationsforimprovingIRIS.

OldensucceedsNCEAActingDirectorRebeccaClark,whoservedfortwoyearsfollowingthedepartureoftheprogram’slong-timeformerdirector,PeterPreuss.Olden’sprimaryassignmentwillbeatNCEAheadquartersinWashington,D.C.,althoughResearchTrianglePark,N.C.-basedregionaldirectorJohnVandenbergsaidheanticipatesOldenwillalsohaveofficespaceatthefacilityinN.C.

“Writinghasalwaysbeenmypassion,butIwasn’tsurewhichcareerpathwastherightoneforme,”saidEmilyZhou,Ph.D.,aformerNIEHSresearchfellowwhorecentlymovedontohernewpositionasamedicalwriteratPPD.“NIEHSprovidedthesupportIneededtofigurethatout.”

ZhouobtainedherPh.D.inpharmacologyfromtheUniversityofNorthCarolinaatChapelHill,andcametoNIEHSinApril2009asaresearchfellowworkingwithStephenShears,Ph.D.,intheLaboratoryofSignalTransduction.ShearswasverysupportiveofherinterestinwritingandofferedheropportunitiestocomposecommentariesonpublishedliteratureonFaculty1000.Moreover,herstintwiththeEnvironmentalFactorbroadenedherexperienceinwritingforawideraudience.“I’mveryappreciativeofopportunitiesavailablewritingfortheEnvironmentalFactor.Iwasabletobuildandshowcaseaportfolioofwritingsamples.”

Inhernewposition,Zhouprovidesfeasibilityservicestopharmaceuticalclientswhowishtoconductclinicaltrialsbyanalyzingexistingdataandinfrastructure,andsubsequentlymakingrecommendationsastowhetherthetrialsarefeasibleornot.“Iwasn’tsurewhatIwasgettingmyselfinto,”shelaughed,whenaskedhowhercurrentpositionisdifferentfromherresearchroleatNIEHS.Hercurrentjobisfastpaced,andhavingashortturnaroundtimeforherassignmentshasbeentestingherabilitytothinkonherfeet.“Ihaveaweektoanalyzedataandwriteanaverage40-pagereport,anditdoesgetveryintense.However,IenjoythefactthatIamworkingwithsomethingdifferenteveryday,”shecommented.

Career development and networkingApartfromherresearchresponsibilitiesatNIEHS,ZhouwasactivelyinvolvedwiththeNIEHSTraineesAssembly(NTA)andwasaco-chairforthe2011BiomedicalCareerFair.HerinvolvementwiththeNTAsubcommitteesgavehertheopportunitytomeetpeopleoutsidethelabandinotherorganizations,andexposedhertovariousmanagerialandorganizationaltasks,allofwhicharecrucialtohercurrentjob.Timemanagementskillsplayedabigroleaswell,asshehadtobalanceherresearchwithhereffortsinbuildingherprofessionalnetwork.Shealsoacknowledgedthattheanalyticalandcriticalthinkingskillsshelearnedasagraduatestudentandresearchfellowhelphertremendouslyinherroleasamedicalwriter,wheresheoftenhastoanalyzedataandinformationonvarioustopicsthatareoutsideherfieldoftraining.

Zhoufoundherjobonline,butwithatwist.“Andywasjob-huntingtoo,sowehelpedeachotherout,”shesaid,referringtoNIEHSpostdocAndySeipel,Ph.D.,whoworksinthelabdownthehallfromhers.Seipelcameacrossthisopeningonlineandsentittoher,andtherestishistory.Therefore,onepieceofadviceshehastoofferistoformasupportgroupandhelponeanotherout.

“Jobhuntingcanbeverystressful,anditdoeshelptohaveanotherpairofeyesouttheretomakesurethatnostoneisleftunturned,”Zhousaid.Shealsofeelsthatwhileitisimportanttomakeone’scareeraspirationsapriority,itisalsoessentialtobeattentivetochangesinthejobmarkettrendstoensurehighersuccessinfindingajobthatsuitsone’sinterests.

Atthispoint,Zhouisunsureaboutherplansbeyondhercurrentpositionorwhereitwilltakeher.“ButIamveryhappywhereIam,”shesmiled.

InspiredbyanewstudyfromaresearchteamledbyNIEHS/NTPDirectorLindaBirnbaum,Ph.D.,PublicRadioInternationalbroadcastaninterviewwithBirnbaumJune8aspartofitspopular“LivingonEarth”series.Duringthefive-minutediscussionwithhostSteveCurwood,Birnbaumaddressedconcernsaboutfireretardantchemicals,inamessagedesignedtoinform,withoutundulyalarming,ageneralaudience.

PublishedonlineMay31byEnvironmentalHealthPerspectives,thestudyattractedtheattentionof“LivingonEarth”editorswithitsfindingsofdetectablelevelsofthechemicalhexabromocyclododecane(HBCD),abrominatedflameretardantusedinpolystyrenefoamsinthermalinsulationandelectricalequipment,in42percentoffoodsamplesfromDallassupermarkets.Fattyfoods,suchasbeef,poultry,fish,andpeanutbutter,weremostlikelytocontaintracesofthechemical.Inmeats,itwaslikelytheresultofbioaccumulation.Inpeanutbutter,theculpritwasprobablycontaminatedpackaging.

Anexpertinthefieldwhohaspublishedanumberofstudiesaboutfireretardantchemicals,Birnbaumwascarefultostrikeabalancebetweencommunicatingherconcernsaboutthechemicalsinhumansandanimals,bothinfoodandfromexposuretoproductstreatedwithfireretardants,andreassuringtheaudience.

“Ithinkit’simportanttostressthatthelevelsthatwefoundarevery,verylowatthispoint,”shetoldCurwood,“and,infact,thatthelevelsarealmostathousandtimeslowerthanwhathasbeenconsideredtobeavirtuallysafedoseforthesechemicals.”

AsBirnbaumexplained,sheandothershaveconcernsaboutpotentialeffectsofcumulativeexposureontheimmuneandreproductivesystems,aswellasonthedevelopingnervoussystemandinendocrinedisruption.Whiletherearecertainlyappropriateapplicationsforfireretardants,suchasinairplanes,shesaidtheyareprobablynotasusefulinmostoftheproductswheretheycanbefound.

“Ithinkthequestionweshouldbeaskingaboutsomeoftheseflameretardantsis,‘Dowereallyneedtheminnursingpillowsandbabystrollersandbabycarseats?’”Birnbaumconcluded.“Ithinkthat’sreallyaveryimportantquestiontoask,whethertheyreallyareprovidingafiresafetybenefitinallsituations.”

Workshop challenges high school students with a call to action By Eddy Ball

StudentsattendingthesecondannualNIEHS/U.S.EnvironmentalProtectionAgency(EPA)ClimateChangeWorkshopJune11-15gotsomethingmorethantheymighthaveexpectedfromtheirlearningexperience.Alongwithpresentationsbyexperts,thehighschoolerswerechallengedtoreachouttostudentsandfacultyattheirschoolsthisfall,andgivendirectionindesigningeffectivestrategiesforraisingawareness,byveteranenvironmentalorganizerJohannadeGraffenreidoftheAllianceforClimateEducation(ACE).

Todrivehometheworkshop’scalltoaction,workshoporganizersBonoSen,Ph.D.,ofNIEHS,andKellyLeovicofEPAscheduleddeGraffenreidastheopeningandclosingspeakerattheworkshop.InbetweendeGraffenreid’sopeningsessionMondaymorningandhercourseinProjectPlanning101,thestudentsheardfrom,andinteractedwith,specialistsinclimatechangeandhealthfromNIEHS,EPA,andelsewhere,onarangeofclimatechange-relatedtopics(seetextbox).

“Welearnedalotfromlastyear,”Sensaidofthedebutworkshopin2011(seestory).“Thisyear,Ibetthewholecohort[ofstudents]willengageinfollow-upactivities.”Senpointedtotheadditionofmoremovementandtourstotheschedule,aswellastheimmediacyofmuchoftheworkshopcontent.

Accordingtostudentevaluations,SenandLeovic’splanningpaidoff.Asonestudentwrote,“Iwillpassonmyknowledgetomembersinourenvironmentalclub,andthisprogrampropelledmetoparticipateinmoregreenprograms.”

As timely as the evening newsReinforcingtheworkshop’scalltoactionwasthetimelinessofseveralofthepresentations.NIEHSSeniorAdvisorforPublicHealthJohnBalbus,M.D.,discussedeffortsbyNIEHSandotherfederalgroupstomakehealthafocusofsustainabledevelopmenteffortsunderwayattheRio+20UnitedNationsConferenceonSustainableDevelopment(seestory).“Healthisoneofthoseareaswherewecanreallydosomethingaboutthiscredibleimpassewehaveontheinternationalscene,”Balbustoldattendees,ashedescribedtheeconomicbenefitsofimprovedhealthfromclimatechangemitigation.

ThestudentswerealsotreatedtoalivelypresentationJune14on“TheBattleforNorthCarolina’sCoast,”byEastCarolinaUniversitygeologistStanRiggs,Ph.D.,arepresentativeoftheN.C.CoastalResourceCommission’sSciencePanelonCoastHazards.Riggsdiscussedhisgroup’spositiononapossibleone-meterincreasein

DeGraffenreid’sclosingsessionJune15combinedtimelinessandacalltoaction,withamoduleonhydraulicfracturingorfracking.LikeRio+20andsealevelriseonthecoast,frackingwasinthenewsthatweek,asN.C.legislatorsconferredoverdifferentversionsofabilltolegalizethepracticeinthestate.TheEPAbuildinginResearchTrianglePark(RTP),N.C.,wheretheworkshopwasheld,sitsontheedgeofaTriassicBasinshaleoildepositthatrunsfromDurham,N.C.,totheSouthCarolinaborder.

The impact on participantsStudentsevaluatedactivitieseachdayand,astheworkshopcametoaclose,theyhadanopportunitytocommentontheentireprogram.Someparticipantsofferedsuggestionsabouthowtotweaktheworkshopscheduleandsuggestedactivities,butthestudents’commentswereoverwhelminglysupportiveoftheexperience.Amajoritywrotethattheywouldattendtheworkshopagain,evenitdidn’tfeaturethemodeststipendtheprogramofferedthisyear,whichwasmadepossiblethroughfundingprimarilybyEnvironmentalHealthPerspectives(EHP).

“[Theworkshop]completelychangedmyviews,”wroteonestudent.“Ihadnoideaclimatechangewassourgent.”Anotherstudentagreed,writing,“[I]learnedalotabouttheactualscienceandpoliciesregardingclimatechange,andIfeelmoreawareandconsciousabouthowmyactionsaffectclimatechange.”

Justasorganizershoped,thestudentswereinspiredbytheworkshop’scalltoactionandemphasisonplanningandcommunication.“InowknowaprojectIcandoatmyschooltohelpourplanetgogreener,”oneparticipantconcluded.Otherslistedspecificprojects,includingcomposting,conservationofwaterandelectricity,andinstallationofsolarpanels.

Although Balbus gave one of the longer presentations, he kept the students’ attention with his outline of the cobenefits that would accrue from mitigating the health impact of climate change, and his ongoing involvement in the international discourse about climate. Resnik book tackles ethical issues in environmental health By Eddy Ball

NIEHSBioethicistDavidResnik,J.D.,Ph.D.,istheauthorofathought-provokingnewbookappropriatelytitled“EnvironmentalHealthEthics.”Thebook,whichisResnik’seighth,isnowavailablefrompublisherCambridgeUniversityPress,aswellasseveralonlinebooksellers.

Resnikoffersreadersamethodforweighingbenefitandharminavarietyofsituations,rangingfrompesticideuse,antibioticresistance,nutritionpolicy,vegetarianism,andurbandevelopmenttooccupationalsafety,disasterpreparedness,andglobalclimatechange.ThebookbeginsbydescribingthebirthofthemodernenvironmentalmovementinspiredbyRachaelCarson’s“SilentSpring”in1962.ThedebatethatensuedoverthepowerfulinsecticideDDT,Resnikexplained,continuestodayandinvolvesweighingimportanthumanbenefits,increasedcropyield,andthereductionofdiseasestransmittedthroughinsectswithharmtotheenvironmentfromdamagetowildlifeandhumansexposedthroughdietordirectcontact.

One outcome of student collaboration at the workshop was this brochure the students put together to bring home a message about the very real threat to health and wellbeing posed by rising sea levels.

Striving for a conceptual frameworkInthiskindofsituationaldecision-makingarena,Resnikargues,compromiseamongpeoplewithcompetingvaluesandinterestsisessentialandunavoidable.Absoluteethicalprinciplesgiveway,becauseideological,economic,orreligiousethicalsystemsaremorelikelytoalienateopponentsthanhelpthemtofindamiddleground,wherecompetitorswhocannotwinentirely,atleastcanendupfeelingtheyhaveachievedafairamountofwhattheyneedanddeserve.

“Anotherthemeofthebookisthathumanhealthandtheenvironmentputanewtwistonsomeofthetraditionalquestionsofethics,”Resnikwrote.Theseincludeissuesofindividualrightsandthecommongood,themoralbasisforgovernmentrestrictionsonpropertyrights,andjustice,allcomplicatedbytheirintersectionwithinthecontextoftheenvironmentalpolicyandenvironmentalhealth.Withinenvironmentalhealthethics,thereareusuallyfarmorethantwosidestoeveryissue.

Intothismultifacetedethicalbattleground,Resnikintroducesaconceptualframeworkthatheappliestopracticalproblemsrelatedtoenvironmentalhealth,whichwillbeveryfamiliartoreaderswhatevertheirbackgroundorinterests.Hisgoalisambitious—tomakepossibledecisionsandpolicies,thatareconsistentandbasedonevidenceandargument,opentopubliccriticism.

NICEATM seeks comment on draft plan for 2013–2017 By Debbie McCarley and Cathy Sprankle

TheNTPInteragencyCenterfortheEvaluationofAlternativeToxicologicalMethods(NICEATM)istakingpubliccommentsuntilAug.13onadraftfive-yearplandevelopedincollaborationwiththeInteragencyCoordinatingCommitteeontheValidationofAlternativeMethods(ICCVAM).ICCVAMisaninteragencycommitteeofthefederalgovernmentthatNICEATMadministers.TheplanisavailableontheNICEATM–ICCVAMwebsite.

ThedocumentwillprovidestrategicdirectionforNICEATMandICCVAMduring2013–2017.Itoutlineshow,consistentwithICCVAM’sstatutorydutiesandpurposes,NICEATMandICCVAMwillcontributetothetransformationofsafetytesting,byfosteringandpromotingtheincorporationofscientificadvancesandinnovativetechnologiesintonewimprovedtestmethods,andintegratedtestinganddecisionstrategies.

ICCVAMwasestablishedtopromotedevelopment,validation,andregulatoryacceptanceofnewtoxicologyandsafetytestingmethods.Newtestmethodsthattakeadvantageofadvancesintechnologycanbettercharacterizethesafetyandpotentialhazardsofchemicalsandchemicalproducts,whichinturncanprovidebetterprotectionofpeople,animals,andtheenvironment.Newtestmethodsalsoprovidetheopportunitytousefewerornoanimalsforchemicalsafetytesting,ortoenhanceanimalwell-being,andlessenoravoidpainanddistressinthosecaseswhereanimaltestingisstillnecessary.

In2008,NICEATMandICCVAMpublishedafive-yearplanfor2008-2012.TheplandescribedICCVAM’svisiontoplayaleadingroleinfosteringandpromotingthedevelopment,validation,andregulatoryacceptanceofscientificallysoundalternativetestmethods,bothwithinthefederalgovernmentandinternationally.

Respondingtothefactthatthetimeperiodcoveredbythecurrentfive-yearplanwascomingtoaclose,lastNovember,NICEATMpublishedarequestforcommentondevelopmentofafive-yearplanfor2013-2017.Commentsreceivedwereconsidered,asNICEATMworkedwithanICCVAMsubcommitteeinearly2012todevelopthedraftplanthatwasreleasedlastmonth.

“Recentadvancesinemergingscienceandtechnologyinnovationsaredrivingtransformativechangesintoxicologyandhowsafetytestingisperformed,”notedICCVAMvicechairJoannaMatheson,Ph.D.,oftheConsumerProductSafetyCommission,whoischairingtheICCVAMsubcommittee.“Forexample,datafromin vitrotestingbatteriesandintegrateddecisionstrategies,thatconsiderchemicalandtoxicologicaldatabaseinformation,arebecomingmoreimportanttoregulatorydecision-making.ThenextfiveyearswillbeanessentialtransitionperiodforNICEATMandICCVAMinthischangingregulatorytoxicologyenvironment.”

“Indevelopingthisplan,werecognizetheneedtobeabletotakeadvantageofadvancesinscienceandtechnologythatwilltakeplaceoverthenextfiveyears,”commentedRearAdm.WilliamStokes,D.V.M.,whoisdirectorofNICEATM.“WewanttobestpositionNICEATMandICCVAMtopromoteandhelptranslatetheincorporationofnewscienceintoimprovedandinnovativetestmethodsthatbettercharacterizethesafetyorhazardofnewchemicalsandproducts.Theseimprovedmethodswillbetterprotectpeople’shealth,andarealsoexpectedtoreduceandreplaceanimaluseintesting.”

SRP scientist Brian Jackson engages his community about arsenic By Heather King

OntheeveningofJune20,BrianJackson,Ph.D.,oftheNIEHSSuperfundResearchProgram(SRP)atDartmouthCollege,metupwithfellowscientistsJosephAyotteoftheU.S.GeologicalSurvey(USGS)andPaulSuscaoftheNewHampshireDepartmentofEnvironmentalServices(NHDES)foraninformalgatheringatapopularlocalrestaurant.TheywerejoinedbyapackedhouseofConcord,N.H.,residentswhocametotakepartinaScienceCaféNewHampshirediscussiontitled“ArsenicInOurEnvironment:AreLevelsUnsafe?”andtolearnabouttheimportanceofarsenictestingforprivatewells.

A toxic combination of rock, water, and wellsArsenic,adeadlypoisonthatkillsquicklyathighdoses,canalsocauseharmintheparts-per-billion(ppb)range.Longtermexposuretolowlevelsofarsenicindrinkingwaterhasbeenassociatedwitharangeofhealthproblemsfromcancertoimmunesystemproblemsandreproductivedefects.

Jackson,directoroftheTraceMetalAnalysisCorefacilityatDartmouth,specializesincharacterizinginorganicenvironmentalcontaminants,suchasarsenic,fromsamplesrangingfromorganicnutritionbarstobedrock.InNewHampshire,arsenicinbedrockisabigconcern,becauseoverhalfamillionofitscitizensdrinkfromprivatewellsdrilledintoandaroundthatrock,andthesewellsarenotregulatedormonitoredbyanystatelawsorotheragencies.

Ayotte,ahydrologistwhostudiesgroundwaterqualityinNewHampshire,canalsoattesttothetroublesomecombinationoflowpHandlowoxygeninthewaterflowingacrossthoserocksthatallowsittocarryarsenicintodrinkingwellsatlevelswellabovetheU.S.EnvironmentalProtectionAgencystandardof10ppb.

Susca,whosupervisestheNHDESDrinkingWaterSourceProtectionProgram,knowshowmucheffortgoesintokeepingtheNewHampshirewatersupplysafe.Healsounderstandsthatinformedandactivecitizensarethekeytopreventinghealthissuesrelatedtoarseniccontaminatedprivatewells.

High-stakes community outreachJacksonandfellowScienceCafépanelistswerepleasedtohavethisopportunitytocometogetheranddemonstrateacommonconcernforpublichealth,byexplaining,directlytocommunitymembers,theimportanceofhavingprivatewellstestedforarsenic.Participatinginlocal,communityeventsofthiskindisamajorfocusoftheDartmouthSRP,whichhasanactiveResearchTranslationprogramthatsponsorseventssuchasriskworkshopsandconferences,andhasevenproducedashortfilm,“InSmallDoses:Arsenic,”toinformthecommunityaboutwellwatertesting.

Bidirectional communicationBidirectionalcommunicationandcollaborationbetweentheSRPandfederalandstateagencies,suchastheUSGSandNHDES,wasclearintheteamworkshownbythepanelists,buttheirultimategoalwastofostercollaborationwiththepublic.Althoughthescienceandtechnologybehindthestoryofwellwaterarsenicinvolvestechnicalresearchandcomplexpublicpolicy,theultimatemessageisoneforthecommunity—“Protectyourfamily.Testyourwell.”

Effectiveoutreachdoesmorethanteach.Itgetspeopletocollectsamplesoftheirtapwaterandtakethemintobeanalyzed—thecriticalstepintranslatingresearchintoimprovedhumanhealth.Severalpeopleinattendancealsotookasurvey,providingimportantinformationabouttheirwellwaterusethatcanfeedbackintotheresearchbeingdone.

Althoughmanyscientificprogramsaremeasuredbyprestigiousjournalpublicationsandconferencelectures,andtheDartmouthSRPgroupcanboastplentyofboth,thisprogramalsohaskeeninsightintotheimportanceofcommunityengagement.Inparticipatinginthisevent,theyremindfellowscientiststhatsometimesavisittoTheBarleyHouserestaurantcanhaveahigherimpactthananypaper.

Science NotebookJohns Hopkins researcher gives Distinguished Lecture By Robin Arnette

Translation,orhowlivingsystemsassembleproteins,isacomplexmechanismthattakesplaceonacellularstructurecalledtheribosome.ForresearcherRachelGreen,Ph.D.,understandingitsintricaciesprovidestheultimateinscientificinvestigation.ShepresentedherworkduringherNIEHSDistinguishedLectureJune12.

Greenstudiestwospecificaspectsoftranslation,namelyhowtheribosomechooseswhichtransferribonucleicacids(tRNAs)toaddtothemessengerRNA(mRNA)templateandhownewly-formedproteinsarereleased.

Havingalwaysbeenfascinatedbyquestionsabouttheoriginoflife,GreensaidexamininghowRNAstitchesaminoacidstogetherallowshertoseehowtranslationbegan.

“Ifweopenuptheribosomeandtakealookinside,allyouseeisRNA,whichgivesusconfidencethattheprocessmusthaveevolvedearlyoninanRNAworld,”Greensaid.“Proteinscamelateinevolutionasasupplementtomakewhatweknowastranslationbetter,faster,andstronger.”

Making a proteinGreengaveabriefreviewoftranslation,whichisthewalkoftRNAsalonganmRNA.Itoccurswhenapeptidyl-tRNA,oratRNAthathasastringofaminoacidsattachedtoit,ispositionedoverthreenucleotides,orcodons,onthemRNA.Duringthisinitialstepofproteinsynthesis,thepeptidyl-tRNAispositionedinthePsiteoftheribosome.Witheveryroundofaminoacidaddition,theribosomeneedstoselectatRNAfromsolutionthatcorrespondstothecodonpresentedintheneighboringaminoacylorAsite.E. coliandmostorganismshaveapproximately40tRNAsthatcorrespondto61codonsthatcodeforaparticularaminoacid.Thefinalthreecodons,UAA,UAG,andUGA,arestopcodonsbut,insteadofbeingdecodedbytRNA,theyarerecognizedbyproteinscalledreleasefactors(RFs).

Inbacteria,RF1andRF2bindwithanenergy-producingGTPasemoleculecalledRF3toterminatetranslation.Inhigherorganisms,eukaryoticreleasefactor1(eRF1)anditsGTPaseeRF3dothejob.Greenstressedthatalthoughtheseproteinshavethesamenamesandfunctions,theyarequitedifferentproteinsthatevolvedindependently.

Green was elected to the National Academy of Sciences in May and is a professor in the molecular biology and genetics department at Johns Hopkins University School of Medicine, as well as a researcher at the Howard Hughes Medical Institute. Degradation machineryPreliminaryworkledGreentothinkthatterminationeventswerecloselyrelatedtothemonitoringofRNAquality,alsoknownasmRNAsurveillance.IftheRNAhaserrorsinit,thecellwantstogetridofthoseaberrantmessages,becausetheyaretoxic.Researchbyothergroupsfoundthattwoproteinstriggerthedegradationofabadmessage,Dom34andHbs1.Surprisingly,Dom34isaneRF1homologandHbs1isaneRF3homolog.

“Sinceithadjustbeenshown,genetically,thatDom34andHbs1areimportantfortargetedmRNAdecayincells,wewantedtoseewhattheywoulddoiftheyencounteredaribosome,”Greenadded.

OneofGreen’sgraduatestudentsreconstitutedtranslationin vitro,byusingpurifiedyeastribosomalsubunits,mRNAs,tRNAs,andminimalinitiation,elongation,andterminationfactors.Afterdoingkineticanalyses,herantheproductsonanativegelandfoundthatDom34andHbs1triggerthereleaseofpeptidyl-tRNA.

FurtherworkdeterminedthatDom34andHbs1aretermination-likefactorsthatdon’tcatalyzetermination,butareinvolvedinrecycling.Recyclingisavitalstepintranslationthatresultsinthedissociationofribosomalsubunitsafterafull-lengthproteinisreleased.Thisdissociationallowsfornormalinitiationandsubsequentroundsoftranslation.Scientistsknewthatbacteriausedribosomerecyclingfactorforthisaction,butsinceeukaryoteslackedanapparenthomolog,itwasn’tclearhowthisprocesstookplaceinhigherorganisms.Green’sinvestigationsshednewlightonthetopic.

LaboratoryofStructuralBiologyresearcherTraciHall,Ph.D.,hostedtheseminarandlaudedGreen’sworkinthefield.Hallsaid,“Herexquisitebiochemicalstudieshaveexposednovelaspectsofproteintranslationandgenomeregulation,includingqualitycontrolofproteinsynthesis.”

Understanding diseases with an environmental componentBirnbaumopenedhertalkbypresentinghercomprehensivedefinitionofenvironmentandplacingendocrinedisruptioninthelargercontextofdiseaseswithaknownorsuspectedenvironmentalcomponent.Sheexplainedthatenvironmentisanimportantpartofthematrixofdiseasecausation,andemphasizedtheimportanceofindirectordelayedeffectsofexposureinshapingtheendocrinesignalsthatgoverneveryorganandprocessinthebody,aswellasthefrankmanifestationofphysiologicalharm,whichclassictoxicologycallsapicalendpoints.

“Themainpointhereisthatdeathorcancerarenottheonlyendpointsthatareimportantwhenconsideringenvironmentalhealth,”sheexplained.“Reproductivefunction,braindevelopment,lungfunction,andotherphysiologicalpropertiesrelyonproperdevelopmentinordertomaintainhealth,andenvironmentalstressorscanaffecteachofthesefunctions.”

Thepresentationfeaturedseveraldetailedslides,includingaschematicoftheeffectsofinsulinintissue,underscoringthecomplexityandvulnerabilityoftheendocrinesystemtoEDCexposureinitsmanypointsofregulationandcontrol.BirnbaumexplainedthatEDCsareubiquitousintheenvironment,occurringinahostofwidelyusedpesticidesandindustrialchemicals.Theycanhaveawiderangeofeffectsonhealthatlowdoses,especiallywhenexposureoccursduringwindowsofsusceptibilityinthecourseofdevelopment,shesaid.

Lookingahead,BirnbaumdescribedthepotentialofTox21,andotherhigh-throughput,screening-basedpredictivetoxicologyefforts,inmorecomprehensivelyidentifyingpotentialEDCsanddiscoveringnew,relevantendocrineendpoints.Shecalledformoreresearchintomixtures,asharperfocusonpublichealth,andthedevelopmentofeffectiveregulatorypolicytohelppreventdisease.

Re-thinking dose response with EDCsWhileBirnbaum’stalkhadbeenasurveyofEDCs,Heindel’spresentationfocusedonthecharacteristicsandprinciplesthatconvincehimEDCsshouldbeassessedaccordingtodifferentcriteriathanareusedwithmosttoxicsubstances.Thebasisofhisargumentwasthattheendocrinesystemfunctionsaccordingtodistinctprinciples,andthat,byimplication,EDCsshouldbeassessedaccordingtosixendocrine-specificprinciples:

Moving forward with the EU community strategy for endocrine disruptorsPresentationsanddiscussionsattheconferenceexploredtheeffectsofendocrinedisruptorsonhumanhealthandtheenvironment,therisks,theidentificationofendocrinedisruptors,andpolicyobjectives.AlongwithareportcommissionedbytheDirectorate-GeneralfortheEnvironmentin2009andpublishedthisyear,“StateoftheArtoftheAssessmentofEndocrineDisrupters,”theoutcomeoftheconferencewillfeedintothereviewoftheEuropeanCommission’scurrentstrategyonendocrinedisruptors.ItwillalsoprovideinputtotheCommission’supcomingproposalforcriteriafortheidentificationofsubstanceswithendocrinedisruptingproperties.

TheEU“CommunityStrategyforEndocrineDisrupters,”firstpublishedin1999,focusedonshort,medium,andlong-termactionsthat,whenimplemented,wouldcontributetoensuringabetterenvironmentandhealthforpeoplewithintheEU.TheEuropeanCommissionhassupplementedtheoriginalstrategydocumentwithaseriesofimplementationdocumentsrefiningproposedactions.

Prenatal chlorpyrifos exposure alters brain structure, affects IQ in children By Carol Kelly

AnewNIEHS-fundedstudyreportsthatwhenapregnantwomanisexposedtochlorpyrifos,acommonlyusedpesticide,herchild’sdevelopingbrainmaybedamaged,leadingtoloweredintelligence.

“Theprenatalperiodisavulnerabletimeforthedevelopingchild,andthattoxicexposureduringthiscriticalperiodcanhavefar-reachingeffectsonbraindevelopmentandbehavioralfunctioning,”saidNIEHSgranteeVirginiaRauh,Sc.D.,aprofessorattheColumbiaUniversityMailmanSchoolofPublicHealth.“Wehavedemonstratedthatthereisarelativelymonotonicdose-responseeffect,suggestingthatsomesmalleffectsoccuratevenverylowexposures.”

Theevidencecomesfromresearcherswhoconductedarecentbrainimagingstudyofchildrenexposedprenatallytochlorpyrifos.InadditiontoColumbiaUniversityscientists,theteamincludedresearchersaffiliatedwithDukeUniversityMedicalCenter,EmoryUniversity,andtheNewYorkStatePsychiatricInstitute.

Thisstudyisthefirstofitskindtousemagneticresonanceimaging(MRI).DigitalimagesproducedfromMRIscansarequitedetailedandcandetectsmallchangeswithinthebrain.

Inthescans,structuralchanges—abnormalareasofenlargementandindentationacrossthesurfaceofthebrain—appearedamongthechildrenwhosemothershadbeenexposedtohigherlevelsofthepesticide.Inthreeoffourbrainregions,higherchlorpyrifosexposurewasconsistentwithlowerfull-scaleintelligencequotient(IQ)measuresintheexposedchildren.

ThebrainabnormalitiesappearedtooccuratexposurelevelsbelowthecurrentU.S.EnvironmentalProtectionAgencythresholdfortoxicity,accordingtotheresearchers.

Data implications Toconductthestudy,certainbrainsurfacefeaturesof40urbanchildrenwereexamined.Thesamplesize,althoughmodest,neverthelesspermitsgeneralizationoffindingstoalargerurbanpopulation,becauseitwasarepresentativecommunity-basedsample.

“Thegroupdoesnothaveexposuretopolycyclicaromatichydrocarbons[PAHs]orsecondhandsmoke,twootherexposuresthatmightalsohaveadverseeffectsonbraindevelopmentandneurocognitiveoutcomes,”explainedRauh.“So,thesmallsamplesize,commoninMRIstudies,isbalancedbyarelativelypureorunconfoundeddesign.”

Althoughresidentialapplicationofchlorpyrifoswasbannedin2001,theinsecticideisstillapprovedforotheruses.Chlorpyrifoscontinuestobepresentintheenvironmentthroughitsextensiveuseinwoodtreatmentsandpublicspaces,suchasgolfcourses,parks,andhighwaymedians.Peoplenearthesesourcescanbeexposedbyinhalingthechemical,whichdriftsonthewind.Additionally,chlorpyrifosisapprovedforfoodagriculture,andexposurecanalsooccurbyeatingfruitsandvegetablesthathavebeensprayed.

“Evenifdietarydosesfromresidueonfoodarelow,andwethinktheyareforanyonefood,whathappensifapersoniseatingfivedifferentfoodsonadailybasis,eachwithasmallamountofchlorpyrifosresidue?”positedRauh.“Thisissomethingtothinkabout.”

Future research to continue examining pesticide effects on children“Itwillbeimportanttocontinuetofollowthiscohortofchildren,toseeiftheproblemsresolveorwhethertheyareirreversible,whichisanoutcomesuggestedbyanimalresearchliterature,”saidRauh.“Itmightbepossibletodesignaninterventiontoaddressthemoderatecognitivedeficits.”

“Iftheanimalstudiessuggesttheremaybeaproblem,andwefollowupwithhumanworkthatisconfirmatory,thenwewouldbewisetolimittheuseofsuchchemicalswhentheevidencestartstobuildup,”addedRauh.“Thereareplentyofexamplesofwidespreadorcommonlyusedexposures,forexample,leadandtobacco,wherewewaitedtoolongtotrytoreduceexposure,withtheunfortunateresultofgreatharmtolargenumbersofindividuals,whichmighthavebeenpreventablehadwepaidattentiontothemountingevidence.”

Building on previous findingsPreviousworkconductedattheColumbiaCenterforChildren’sEnvironmentalHealth(CCCEH)demonstratedexposuretopesticidesduringpregnancyharmsthehealthygrowthanddevelopmentofbabiesinthewomb,aswellasadverselyaffectingdevelopmentinearlychildhood.Theirstudiesarethefirsttofollowexposedmothersandtheiroffspringovertimewhiledocumentingchangesinthechildren’scognitivedevelopment.

A2006CCCEHstudyshowedthatchildrenwhowereprenatallyexposedtochlorpyrifoshadsignificantlyworsecognitiveandmotordevelopmentatage3.Earlydelaysaffectchildren’sabilitytolearnandtheirlaterschoolperformance.Thesechildrenwerealsosignificantlymorelikelytoscoreintheclinicalrangeforbehavioralandattentionproblemssuchasattentiondeficithyperactivitydisorder,aconditionassociatedwithalteredbrainfunctioningandcharacterizedbyaninabilitytofocusontasks.

“Thesefindingsindicatethatprenatalexposuretotheinsecticidechlorpyrifosnotonlyincreasesthelikelihoodofdevelopmentaldelay,butmayhavelong-termconsequencesforsocialadjustmentandacademicachievement,”saidRauh,inaColumbiapressreleaseaboutthe2006study.

Ina2011CCCEHstudyinvolvingurbanminoritymothers,childrenborntowomenwhohadthehighestlevelsofchlorpyrifosintheircordbloodduringdeliveryscoredthreepointsloweronIQtestsatage7,comparedwithchildrenborntomotherswiththelowestlevelsofthepesticide.TherelationshippersistedevenafteraccountingforotherfactorsthatcouldexplaindifferencesinIQ,suchasmaternaleducation,thehomeenvironment,andpossibleunderlyingattentionordevelopmentdisorders.

Connecting the dots in Parkinson’s disease By Ernie Hood

ThefirstimagethatcomestomindaboutParkinson’sdisease(PD)islikelytobetheclassicmusculartremorsassociatedwithfamouspatientssuchasMichaelJ.FoxandMuhammadAli.Buttheconditionactuallybeginstomanifestitselfmuchearlierthantheonsetofthosemotorsymptoms,withavarietyofnonmotororpremotorsymptomsthatmayappeardecadespriortothemotorfeatures.Scientificunderstandingoftheroleofpremotorsymptomsintheetiology,development,andprogressionofPDisatanearlystage,buttheexistingresearchisintriguingandthepotentialforimportantnewknowledgetoemergeisenormous.

ThepotentialiswhatledapproximatelytwentyleadingexpertsinthefieldtogatherJune7-8attheParkinson’sDiseasePremotorSymptomSymposium.Themeetingwasco-organizedbyNIEHSepidemiologistHongleiChen,M.D.,Ph.D.,andprogramadministratorCindyLawler,Ph.D.“Wewereinterestedinorganizingthissymposiumbecause,first,bystudyingthepremotorsymptoms,wemaygetawindowtoidentifythediseaseearly,andequallyimportant,wemaylearnmoreaboutearlydiseaseetiology,”saidChen.

“WeneedtobelookingmorebroadlyatParkinson’sdiseaseasasystemicillness,”explainedLawler.“Therehasbeenafocusonthemotorsymptomsandthedopaminesysteminthesubstantianigra,effectsthatoccurlaterinthediseaseprocess.Ifwereallywanttohaveachancetoaltertheprogressionofthedisease,weneedtolookearlieratthisbroaderconstellationofsignsandsymptomsthatareveryimportantintermsofqualityoflifeand,atleastinsomecases,appeartoprecedetheonsetofmotorsymptoms.”

According to Chen, at least one million Americans currently have PD, with an annual cost of $25 billion. “If we identify a potential combination of premotor symptoms that is highly predictive of the disease, we can study the environmental factors that are related to either disease development or disease progression,” Chen told participants. Accordingtoresearchfindingspresentedatthesymposium,thatbroaderconstellationofsignsandsymptomsincludesconstipation,lossofsenseofsmell,excessivedaytimesleepiness,andmoodoranxietydisorders,aswellasREMsleepbehaviordisorder,characterizedbykickingorotherjerkymovementsduringdeepsleep.Someoftheconditions,particularlyincombination,canbequitedebilitatingandmayappearuptotwentyyearspriortothedevelopmentoftheclassicmotorsymptoms.

Three areas of researchThesymposiumdevotedthreemainsessionstothetopicsofclinicalresearchandepidemiology;animalmodelsandmechanisms;andneuroimaging,pathology,andbiomarkers.Asthemeetingprogressed,itwascleartheresearcherswereexcitedbywhattheywerelearningfromtheircolleaguesandeagertopursuenewtypesofcollaborationstoexploretheroleofPDpremotorsymptoms.

Althoughthefindingsaretantalizing,thespeakersagreedthatnoneofthesymptomsbythemselvesarespecificorsensitiveenoughtobedefinitivelyconsideredpredictiveprecursorstoPD.Identifyingcombinationsofpremotorsymptomsthatwouldbereliablypredictivewouldenableearlierdetectionandintervention,withagoalofhaltingoratleastslowingprogressionofthedisease.

“BecausethereissuchalongpreclinicalphaseofParkinson’s,whenpeoplehavesomenonmotorsymptoms,buthavenotdevelopedmotorsymptoms,thatwouldbethetimetointervenewithneuroprotectivestrategies,”saidLawler.“Ifyoucouldfindawaytoevenkeepthematthatleveloffunctioning,itwouldhaveatremendousimpactonqualityoflife.”

New directionsAddressingthegapsintheresearchwillbeverychallenging,buttheopportunitytoelucidatePDetiologyandenhancepreventionorprovideclinicaltreatment,willencourageresearcherstocontinuetoanalyzepremotorsymptoms,incorporatingsomeofthenewideastheyheardatthemeeting.TheresultsofthemeetingwillbeusedtohelpdefinehighpriorityareasforfutureextramuralinvestmentsinNIEHSParkinson’sdiseaseresearch,saidLawler.“Thisisgoingtotakealotofeffortinthinkingof,andplanningfor,thebeststrategytoworkonthepremotorsymptoms,”saidChen.“ThesymposiumgavemealotofideasaboutwhichpremotorsymptomsIshouldfocuson,whatkindofplanningworkIneedtothinkabout,andwhataretheimportantquestionsIneedtoaddress.”

The dual-hit hypothesisGrowinginterestinexploringhowpremotorresearchcanimproveourunderstandingonPDetiology,atleastpartially,arisesfromthehypothesisputforwardin2003byGermananatomistHeikoBraak,M.D.,andcolleagues.Braakandtheothersexpandedonthehypothesisina2007papertitled“Parkinson’sdisease:adual-hithypothesis,”inwhichtheresearcherspositaviralPDetiology,withthevirusenteringdifferentregionsofthebrainfromboththegutandtheolfactorybulb.Thepublication’sabstractoutlinestheconcept.

AccumulatingevidencesuggeststhatsporadicParkinson’sdiseasehasalongprodromalperiodduringwhichseveralnon-motorfeaturesdevelop,inparticular,impairmentofolfaction,vagaldysfunctionandsleepdisorder.EarlysitesofLewypathologyaretheolfactorybulbandentericplexusofthestomach.Weproposethataneurotropicpathogen,probablyviral,entersthebrainviatworoutes:(i)nasal,withanterogradeprogressionintothetemporallobe;and(ii)gastric,secondarytoswallowingofnasalsecretionsinsaliva.Thesesecretionsmightcontainaneurotropicpathogenthat,afterpenetrationoftheepitheliallining,couldenteraxonsoftheMeissner’splexusand,viatranssynaptictransmission,reachthepreganglionicparasympatheticmotorneuronesofthevagusnerve.Thiswouldallowretrogradetransportintothemedullaand,fromhere,intotheponsandmidbrainuntilthesubstantianigraisreachedandtypicalaspectsofdiseasecommence.Evidenceforthistheoryfromtheperspectiveofolfactoryandautonomicdysfunctionisreviewed,andthepossibleroutesofpathogenicinvasionareconsidered.ItisconcludedthatthemostparsimoniousexplanationfortheinitialeventsofsporadicParkinson’sdiseaseispathogenicaccesstothebrainthroughthestomachandnose—hencetheterm‘dual-hit.’

Transcription factor binding dominates talks at minisymposium By Jeffrey Stumpf

DukeUniversityresearcherGregCrawford,Ph.D.,summarizedthespeakers’interestsingenomesciencesbyremindingtheaudiencethat98percentofthegenomedoesnotcodeforaminoacidsequences.Themeetingemphasizedmanywaysthatthenoncodingregionsareinvolvedingeneregulation.

WhileDNAwasthefavorednucleotideatthesymposium,BrownUniversitycomputationalbiologist,CharlesLawrence,Ph.D.,proudlyprovidedarespite.“Ipromise,therewillbenoDNAinthistalk,”jokedLawrence,whoinsteadfocusedongeneregulationbymicroRNAs(miRNAs).Anaverageofonly22nucleotideslong,miRNAsareprevalentinthehumangenomeandrepressgeneexpressionbybindingtotranscripts.BecausefindingthetargetformiRNAsisdifficult,LawrencediscussedhowhismathematicalmodelsnotonlyimprovepredictionofmiRNAsecondarystructures,butalsodissectapossibletranscript-bindingmotif.

When the nucleosomes are away, the transcription factors playAlthoughDNAbindingmotifsareusefulindiscoveringtranscriptionalnetworks,therearedeterminantsforDNAbindingotherthansequence.InthecourseofasessionthatshowcasedhowresearcherstookadvantageoflocatingregionswherenucleosomesbindDNA,severalspeakersemphasizedthatthebindingofnucleosomestoDNAdeterstranscriptionfactoractivation.

“Wecanusenucleosomeevictionasamarkertopredictinterestingregulatoryactivity,”Liebexplained.

CliffMeyer,Ph.D.,aresearcherattheHarvardSchoolofPublicHealth,likenedthestudyoftranscriptionfactorbindingtoPlato’sfamousexampleofinferringtruthfromshadowsinhisAllegoryoftheCave.Toascendfromtheproverbialcave,MeyeridentifiedregionsthatarehypersensitivetoDNasetreatment,indicatingsectionsofthegenomewherenucleosomesbindlessfrequently.

“Theupregulationofgeneexpressioncorrelatedwiththelackofnucleosomeoccupancygloballyandviceversa,”Meyerexplained.

LiebusedasimilarapproachtodeterminenucleosomeoccupancybutappliedittoDrosophiladevelopment.Bysearchingregionswithdecreasednucleosomebinding,Liebidentifiedaregulatoryelementina27-kilobaseregionthatprogramscellstobecomehalteres,structuresthatimprovebalanceduringDrosophilaflight.Furthermore,Liebdescribedchromatinprofilesofvariouscellfatesatdifferenttimes,andwassurprisedbytheconclusion.

“Regardingthechromatinprofile,differenttissuesatthesamestageindevelopmentaremoresimilarthantheirowndaughtercells,”Liebsummarized.

Computer modeling aims for usefulnessInlightofthebiologicaldatapresentedbyLiebandMeyer,DukeUniversityprofessorAlexanderHartemink,Ph.D.,providedacomputer-generatedmodelcalledCOMPETEtovisualizethecompetitionbetweennucleosomesandtranscriptionfactorsinbindingDNA.Hismodeldemonstratedthatincreasingonetranscriptionfactormightdisplacenucleosomestootherregions,causingreducedbindingofnearbytranscriptionfactors.

Consideringhismodelwasdevelopedusingonlycomputationalmethods,Harteminkinterpretedhisresultscautiously,remindingtheaudienceofaquotefromthestatisticianGeorgeBox,Ph.D.,“Allmodelsarewrong,butsomemodelsareuseful.”

NIEHS researchers publish in scientific video journal By Robin Arnette

Forresearchers,beingabletoduplicatetheresultsthatappearinjournalarticlesisfundamentaltoscience,buteventhemostmeticulouslywrittenmaterialsandmethodssectiondoesn’texplainalloftheessentialdetails.That’swhyLaboratoryofMolecularCarcinogenesis(LMC)researcherGuangHu,Ph.D.,andvisitingfellowXiaofengZheng,Ph.D.,usedavideotodemonstratetheprocedurestheyusedintheirexperiments.

Symposium highlights cross talk between biologists and computer scientistsThesearchforDNAbindingmotifsinvolveshigh-throughputtechniquesthatonlyhavebeenpossiblewithinthelastdecade.NIEHSScientificDirectorDarrylZeldin,M.D.,introducedthesymposiumwiththemessagethatminingthroughthewealthofdataisournextgreatestchallenge.

“Asourabilitytogeneratelargeamountsofgeneticdatahasadvancedatanastronomicalrate,ourabilitytoanalyzethedataandunderstandfundamentalbiologicalquestionshasnot[advancedatthesamepace],”Zeldinremarked.

Lialsoacknowledgedtheimportanceofmixingthebestoftwoseparate,yetequally,importantworlds.

“Advancesintechnologiessuchasnext-gensequencingmakeitpossibletostudygenome-widegeneexpressionorproteinbindinginasingleexperiment,”Linoted.“However,makingsenseofthehugeamountofdataremainsachallenge,makingbioinformaticsanincreasinglyintegralpartofmodernbiology.”

Themarriageoftwodistinctdisciplinesnotonlyhasshapedtoday’sresearch,butalsohasprovidedthesciencecommunitywithmanycomputationaltools.Manyoftheseprogramsarepubliclyavailable,includingENCODE(theEncyclopediaofDNAElements,producedbyCrawford),BACH(Bayesian3DconstructorforHi-Cdata,describedbyspeakerJunLiu,Ph.D.,oftheHarvardSchoolofPublicHealth),ChromHMM(Chromatin-statediscoveryandcharacterization,writtenbyspeakerJasonErnst,Ph.D.,ofUCLA),andCOMPETE.

Althoughthesymposiumfeaturedspeakerswithdiverseexperienceandexpertise,Zeldinnotedtheircommonattributes.

“Theseareinternationallyrecognizedthoughtleaderswhohaveachievedbothsuccesses—notonlydealingwithsophisticatedbiologicalquestions,butalsorenderingthefindingsaccessibletoanswerimportantenvironmentalhealthproblems,”Zeldinconcluded.

ZhengandHupublishedtheirfindingsintheJournalofVisualizedExperiments(JoVE),thefirstscientificjournaldedicatedtocapturingtheintricaciesoflifescienceresearchusingprintedwordandvideo.Theirpaper,titled“Oct4GiPreporterassaytostudygenesthatregulatemouseembryonicstemcellmaintenanceandself-renewal,”appearedonlineMay30inJoVE.

Lights, camera, actionAJoVEvideographercametoNIEHSandfilmedZhengdoingatechniqueheandHudevelopedcalledtheOct4GiPreporterassay.Theassayallowsresearcherstoquicklyidentifyandstudyimportantgenesinmouseembryonicstemcellself-renewalanddifferentiation.

“Comparedtoothermethods,itismoreconvenient,sensitive,quantitative,andhasalowercost,”Zhengsaidoftheprocedure.“Itisespeciallysuitedforlarge-scalestudiessuchashigh-throughputscreensorgeneticepistasisanalysis.”

OneofthestepsoftheOct4GiPreporterassayrequiresintroducingsmallinterferingRNAs(siRNAs)artificiallyintocells,alsoknownastransfection.Zhenghadtocarefullycalculate,count,pipet,andmixthetransfectionreagentsandcellstoensurethatthecellswereevenlyplatedandthatamajorityofthemsuccessfullyreceivedthesiRNAs.ZhengandHubelievehavingthestep-by-stepprocessonvideowillhelpotherscientistsbypassmanyofthepotentialpitfallsthatcouldleadtoexperimentalfailure.

Seeing is believingJoVEstartedin2006and,sinceJune2012,haspublished64issuescontainingmorethan1,700videoprotocols.Thevideosaredividedintoinsixcategories:general,neuroscience,immunologyandinfection,clinicalandtranslationalmedicine,bioengineering,andappliedphysics.

Pollution and obesity impact asthma control in seniors By Eddy Ball

Anewstudy,fundedinpartbyNIEHS,explorestheeffectsoftrafficpollutionandobesityonthegrowingnumberofasthmaticsintheU.S.whoare65orolder.ThepaperappearsintheJuneissueoftheAnnalsofAllergy,Asthma,andImmunology.

Despitethepublichealthimplicationsofthehighratesofmorbidityandnearlyfive-timesgreaterasthma-relatedmortalityinthispopulation,theresearcherswrote,predictorsofasthmacontrolinolderpeopleremainpoorlyunderstood.Totheresearchers’knowledge,theirsisthefirststudytodemonstrateasignificantassociationbetweenestimateddailyresidentialexposuretodieselparticulatematterandasthmaseverityinpeople65andolder,whomakeuptwo-thirdsofasthma-relateddeathseachyear.

“Poorasthmacontrolcanleadtoadecreasedqualityoflifeandanincreasedriskforemergencydepartmentvisits,hospitalizations,anddeath,”leadauthorTollyEpstein,M.D.,wasquotedassayinginaUniversityofCincinnati(UC)pressrelease.Epsteinisanassistantprofessorinthedivisionofimmunology,allergy,andrheumatologyinUC’sDepartmentofInternalMedicineandaUCHealthallergistwhoheadedtheteamofeightcliniciansandenvironmentalscientists.

A growing public health issue“Thehealtheffectofoutdoorairpollutantsonasthmainbabyboomers,aswellasyoungchildren,issubstantialandunderappreciated,”saidstudyco-authorDavidBernstein,M.D.,aprofessorintheUCimmunology,allergy,andrheumatologydivisionandalsoaUCHealthallergist.

Accordingtothestudy’sauthors,thispopulation,withanasthmaprevalencerateashighastenpercent,isestimatedcurrentlytoinclude3.1millionpeopleandisexpectedtodoublewithinthenext25years.Seniorstypicallyexperiencehigherratesofobesityaswell,whichthestudyfoundwasalsoassociatedwithpoorerasthmacontrol.Inaddition,obesitymayhaveexacerbatedhostresponsestotrafficpollutionforpeoplewithasthma.Reasonsforthisareunclear,butcouldpossiblybeduetoweight-relatedinflammation.Theresearchersspeculatedthatanage-relateddeclineinantioxidantdefensesmayexplainwhyolderasthmaticsareespeciallysusceptibletotheeffectsofsuchairpollutantsasnitrogendioxide,ozone,andfineparticulatematter.

Asthmatic seniors in CincinnatiTheresearchers,allofwhomhaveexperiencewithsimilarstudiesinvolvingchildren,carefullycontrolledtheselectionandstudyoftheirpopulationof104patients,65yearsorolder,whowererecruitedfromallergyandclinicalprogramsinthegreaterCincinnatiarea.Enrollmentrequiredaphysician’sdiagnosisofasthmaaswellasindependentclinicalconfirmationbasedonobjectivetesting.Theteamexcludedpatientswithchronicobstructivepulmonarydiseaseorcongestiveheartfailure.

Theparticipantscompletedastandardized,close-endeddemographicsandmedicalquestionnaire,whichwasverifiedwithmedicalrecords,andtheAsthmaControlQuestionnaire(ACQ),whichwasusedtoquantifysymptomseverity.Theteamestimateddailyexposuretoelementalcarbonattributabletotraffic(ECAT)usingaland-useregressionmodelbasedontheparticipant’scurrentresidentialaddress.

AnalysisshowedthatparticipantsexposedtohigherlevelsofECATwerenearlythreetimesmorelikelytohavehigherACQscores,indicatingmoreseveresymptoms.Obeseparticipants,definedasthosewithabodymassindexgreaterthan30,weremorethanfivetimesaslikelytohavehigherACQscores.

endocrinedisruptingeffectsandmayhaveharmfuleffectswhenexposuresoccurduringcriticalperiodsofneurodevelopment.PBDEshavechemicalstructuressimilartothyroidhormones.

AccordingtoStapleton,indoordustlevelsofPBDEsarehigherthanmostpeoplewouldexpect,andaresurprisinglysimilartothelevelssheandhercolleaguesfoundinarelatedstudythatmeasuredPBDEsinmunicipalsewagesludge.

From house dust to hands Oneoftheprimaryobjectivesofthisstudywastobettercharacterizechildren’sexposuretoPBDEsfromhousedust,whilealsoexaminingtherelationshipbetweenPBDEexposureandothervariablesthatmayaffectexposure.Thestudyenrolled83childreninNorthCarolinaandmeasuredPBDElevelsinbloodandhandwipesamplestakenfromeachchild,aswellasinhousedustsamples.TheresearchersfoundastrongcorrelationbetweenthePBDElevelsfoundonthehandwipesandthelevelsmeasuredintheblood.

ThefindingssuggestthattoddlershaveasignificantamountofexposuretoPBDEsbytransferringhousedustparticlesfromtheirhandstotheirmouths.Thissuggestsexposuretootherhouseholddustcontaminants,suchaspesticides,polycyclicaromatichydrocarbons(PAHs),perfluorinatedcompounds(PFCs),andothertypesofflameretardants,mayoccurthesameway.PAHsareproducedasbyproductsofburningfuel,whilePFCsarefoundinproductsthatresiststains,oil,andwater.Allofthesecontaminantsarebelievedtoposeahumanhealthrisk.

Themethodsusedinthisstudyalsosuggestthevalueofusinghandwipesasatooltomeasurechildren’sexposuretodifferentchemicalsfoundindust.“Handwipesmightbeabetterroutetomeasureexposure,”Stapletonsaid.

Other risk variablesThestudyalsofoundotherfactorsthatmaycontributetoexposurelevels,includingage,socioeconomicstatus,anddurationofbreastfeeding.Whilethesefactorsmaybesignificantpredictorsofexposurelevels,thestudypointsoutthatfurtherresearchisneededtoexplorewhythesefactorsareinfluencingPBDEexposureinchildren.Thisinformationmayhelpinmediatingthepotentialhealthrisksfromexposure,byincreasingpublicawarenessaboutwhatotherfactorscontributetoexposure.

Whenaskedabouttheimportanceofthislineofresearch,Stapletonsaidthestudyisonlyoneofmanythatshefeelsarenecessary,inordertogatherasmuchinformationaspossibleaboutchildren’sexposuretoPBDEsandotherchemicalsfoundindust,tounderstandthepotentialhealthrisksthatmaycomefromthistypeofexposure.

Public health concerns AlthoughtwoformsofPBDEs,pentaBDEandoctaBDE,werephasedoutofuseintheU.S.in2004,andathird,decaBDE,isscheduledtobevoluntarilyphasedoutstartingin2013,itisimpossibletodetermine

Stapletonisalsointerestedinconductingfuturestudiestoinvestigateexposurefromnewflameretardantsthatarenowonthemarketandarebeingusedtoreplacethephased-outPBDEs.Children,aswellasadults,aremorelikelytobeexposedtoacomplexmixtureofchemicals,insteadofjustone,becausetherearesomanychemicalsfoundinhousedust.“Weknowverylittleaboutthehealtheffectsfromexposuretocomplexmixturesofchemicals,”acknowledgedStapleton.

Statistical approachesInhiskeynoteaddress,UniversityofWashingtonbiostatisticianandgeneticistBruceWeir,Ph.D.,discussedclassicstudiesintheheritabilityofhumanheight,beginningwiththedataandfindingspublishedin1886byFrancisGalton.Whilefamilystudieshavedemonstratedthat80percentofheightisheritable,searchforgeneticvariantsassociatedwithheightcouldnotaccountformorethan10percent.Weirpresentedrecentadvancesinstatisticalmethodologythatincreasedthatfigureto45percentanddiscussedwaystoaccountfortheremaining35percent.Weir’sfellowpresenterDanielStram,Ph.D.,oftheUniversityofSouthernCalifornia,aswellasotherspeakersontheprogram,arestrivingtoaccountforthecomplexityofheritability,bybettercapturingthestrongpolygenicsignalcreatedbyadditiveheritabilityofmanycommonvariants.

Acommonthemeamongthestatisticiansatthemeetingwastheneedforevenlargersamplesizes,forutilizinghiddenheritabilityassociatedwithcommonSNPs,bybuildingstatisticalmodelsthataremoresensitivetohiddenstratification,tounravelthegeneticarchitectureinvolved.Speakersproposedseveralstatisticalrefinements,suchasvariancecomponentsmethods,ensembleorsettestingthataggregatesindividualfeatures,andaprototypesimilaritycollapsingapproachformoreeffectivelycapturingadditiveandnon-additiveeffectsamongmarkers.

Biological approachesAspartofasymposiumthatwasfilledwithstatisticaldiscussion,UniversityofChicagogeneticistNancyCox,Ph.D.,spokeontyingbiologicalfunctiontoanalyses,andNorthCarolinaStateUniversitygeneticistTrudyMackay,Ph.D.,addressedthegeneticandenvironmentalfactorsaffectingvariationinquantitativetraits,usingDrosophilaasamodelsystem.

AsthesymposiumatNIEHSdemonstrated,therearetwomajorapproachestoteasingmoretranslatableinformationfromthevolumesofdataavailablefromlarge-scaleassociationstudies.Onedeductiveapproachistorefineanalysismethodologytogiveassociationsbetweendataandendpointsmorestatisticalpower.Thesecond,moreinductiveinnature,involvesgroundinglarge-scaleassociationstudystatisticalanalysisinbiologyitself,throughanalysisoffunctionandorthologicalpatternswithmodelspecies.

Bothapproachesseektoilluminatewhathasbeendescribedasthedarkmatterofthegenome—missingheritability.Currentstatisticalapproachesarelimitedinthattheyaccountforonlytheadditivepartofheritability—thevariationtransmittablefromparentstooffspring.However,jointeffectsofallelicvariants,whilegenetic,arenottransmittableinthesameway,becausespecificcombinationsofvariantsarebrokendownbyrecombination.

AsZaykinexplained,thenon-additivepartcanbesubstantial.Thereisalsoaproblemofpredictingindividualrisk.Itisonequestiontoexplaingeneticvariationusingasample.Itisanother,andstillunsolved,questionofhowtopredictriskforanindividualgivenhisorhersequencedata.Thetechnologicallimitationisthestillincompletecoverageofallvariantsofindividualgenomes.

Workshop on informed risk assessment ponders new directions By Cindy Loose

Emergingscientificadvancescouldtransformthewayscientistsanalyzetheriskoftoxicsubstancestohumans,allowingbothmoreefficientandmoreexactriskassessments.

“Makingthatleap,however,isharderthanitsounds,anditsoundsprettyhard,”saidJohnBalbus,M.D.,NIEHSsenioradvisorforpublichealth.Hisremarkscameduringtheopeningofatwo-dayworkshopJune14-15sponsoredbyNIEHS.Theworkshopwasthe11thworkshopintheEmergingScienceforEnvironmentalHealthDecisionsseriesbegunin2009.

Scientistsfromaroundtheworldgatheredforthemeetinginacontinuingattempttobridgethegapbetweentraditionaltoxicologyassessment—testingofanimalstoexamineonechemicalatatime—andnewtechniquesthatresearchthecomplexactionsofchemicalsinthehumanorganismusingmolecularandsystemsbiology,toxicogenomics,computationaltoxicology,andotheremergingsciences.

Confronting the backlog of untested chemicals“Thisisthenextstepinalong,deliberatemarch,”saidBalbus,describingthepurposeoftheSystemsBiology—InformedRiskAssessmentworkshopheldattheNationalAcademyofSciencesinWashington,D.C.“Therearealotofchemicalsouttherethatstillneedtobetested,andwe’retryingtocreateabettersystemforpublichealthprotection.”

Thevastmajorityofthemorethan100,000chemicalsestimatedtobeinusehaveneverbeentestedfortoxicityand,eachyear,thousandsofnewman-madechemicalcompoundsarecreated.

KeynotespeakerKimBoekelheide,M.D.,Ph.D.,aprofessoratBrownUniversityandaveteranNIEHSgrantee,warnedthatchangingriskassessmentbyusingnewbiologyatthemolecularlevelwillrequirebreakingthetraditionalframeworkthathasbeeninplacefor50years.Thecurrentsystem,hesaid,issimplisticandlinear,withthegoalofsettingonenumberasanexposurethreshold.

A dynamic, interactive model for toxicologySystemsbiology,Boekelheidesaid,isactiveandinteractive,withlotsofmovingpartsandaresultingcomplexity.Eventually,however,furtherscientificadvanceswillallowtheprocesstobecomesimpler.“Butwewillhavethesimplicityofknowledge,ratherthanthesimplicityofignorance,”hesaid.

Systemsbiologyisaninterdisciplinaryfieldthatnotonlyidentifiesbiologicalcomponentsandtheirinteractions,butalsooffersexplanationsforhowtheseactionstakeplace.Systemsbiology,therefore,holdsthepromiseofpredictingthetoxicityofanentiregroupofchemicalsthatsharesimilarmode-of-actionpathwaysandmechanismsofcausingtoxicity.

Onehopeforsystemsbiologyisthatitwillalsoallowscientiststomakegreaterinroadsintounderstandingtheeffectofmixturesofchemicals—amorerealisticviewoftheriskstohumanslivinginachemicalsoup.

PresentationsrangedfromanhistoricalviewoftheU.S.EnvironmentalProtectionAgency’sstruggletoestablishtraditionalriskassessment,tocasestudiesofnewriskassessmentapproaches.

Manypresentseemedtoagreethatemergingsciencesrequireagreater-than-everneedforscientistsfromvarieddisciplinestoworktogether.Asecondconsensuswasthatnewriskassessmentapproachesrequireaparadigmshiftthatwillbepainfulandcontroversial,butthereisnogoingback.

Theliterature-basedanalysiscapabilitiesoftheNationalToxicologyProgram(NTP)tookcenterstageduringitsBoardofScientificCounselorsmeetingJune21-22atNIEHS.

RuthLunn,Dr.P.H.,directoroftheOfficeoftheReportonCarcinogens(RoC),andherstaff,werewellpreparedtopresenttheboardwithconceptsoutliningtheplannedreviewsforfivesubstancesproposedforpotentialinclusionintheRoC,whileKrisThayer,Ph.D.,andherOfficeofHealthAssessmentandTranslation(OHAT)teamwowedtheboardwiththeirplanstobringsystematicreviewmethodologyandnewinformationmanagementtoolsintotheirliterature-basedevaluations.

Systematic review will enhance transparency“TheboardisveryenthusiasticandsupportiveoftheNTPtakingaleadershiproleinsystematicreview,”saidBSCchairDavidEastmond,Ph.D.,oftheUniversityofCalifornia,Berkeley,ashesummarizedthesentimentsoftheboard,afterhearinganengagingpresentationbyThayer.

“Systematicreviewisascientificinvestigationthatfocusesonaspecificquestion,andusesexplicit,prespecifiedmethodstoidentify,select,summarize,andassessthefindingsofsimilarbutseparatestudies,”Thayerexplained.“It’straditionallybeenusedforevaluatinghealthcareinterventionsbut,fromwhatwe’veseenoverthepastyear,it’sgoingtobeareallypowerfultoolforconductingourliterature-basedevaluationsandhelpingNTPdevelopevidence-basedconclusions.”

Thayerstressedthatasystematicreviewdoesnoteliminatetheneedforexpertjudgment,nordoesitguaranteereproducibilityintheoverallevidence-basedconclusions,butsheemphasizedhowitwillenhancetransparencyandallowformoreconsistentdatacollectionandevaluation.

Acriticalinitialstepofconductingasystematicreview,shesaid,istodevelopaprotocol,orpredefinedapproach,thatoutlineshowtheevaluationwillbeconducted.ThayeraddedthattheNTPwillengagetechnicalexperts,interagencypartners,andmembersofthepublic,torefinethescopeofanevaluationandcreatethesystematicreviewprotocol.Thayerfinishedbywalkingtheboardthroughaseriesofdemonstrationsofhowaprotocolmightworkfromstarttofinish.

King-HerbertsaidtheapproachesforenvironmentalenrichmentproposedbyNTPareinaccordancewiththe2011GuidefortheCareandUseofLaboratoryAnimals.ShediscussedhowtheNTPwillphaseintwodifferentenrichmentitems,includingcrinklednaturalkraftpaperandplasticrectangularshelters,andhowtheNTPwillcloselymonitortheimpactoftheenrichment.AnNTPanimalwelfarecommitteehasbeenestablishedtooverseetheseactivities.TheBoarddiscussedthecomplexitiesofunderstandingthepotentialimpactsofenvironmentalenrichmentontoxicitystudies,butalsoexpresseditsstrongsupportformovingforward.

Strategic plan update, green light on genetic toxicity testing OthermeetingitemsincludedanupdatefromNIEHS/NTPDirectorLindaBirnbaum,Ph.D.,onthenearlyfinalizedNIEHSstrategicplan.“Thisplanwillbeablueprintfortheentirefieldofenvironmentalhealthsciences,”Birnbaumsaid.

NTPgenetictoxicologistKristineWittreceivedthegreenlightfromtheboard,whichunanimouslyvotedtoapproveaconceptthatwouldexpandtheNTP’sgenetictoxicitytestingcapabilitiestoincludehumancells.And,OHAT’sAndrewRooney,Ph.D.,receivedpraiseforcompletionoftheNTPMonographonHealthEffectsofLow-levelLead.

Report on Carcinogens process in actionAspartofthenewprocessforpreparingtheRoC,LunnandherstaffpresentedtheboardwithfivedraftconceptsonsubstancesbeingconsideredforreviewforpossiblelistinginfutureeditionsoftheRoC.Thefivesubstances—1-bromopropane,cumene,ortho-toluidine,pentachlorophenolandtrichloroethylene—camefromalongerlistof15nominatedsubstancesthatreceivedinputfromthepublicinJanuary.EachdraftconceptoutlinedtherationaleforthenominationandtheNTP’sapproachforconductingthecancerevaluation.Awebpagewillbedevelopedandcontinuouslyupdatedforeachsubstancereviewed.

TheboardreviewedandcommentedoneachdraftthatoutlinedtheNTP’sproposedevaluationstrategythatmayvarydependingonthecomplexityofthescientificinformationonthesubstance.

BirnbaumandNTPAssociateDirectorJohnBucher,Ph.D.,thankedtheboardmembersfortheirvaluableinput.BirnbaumwillmakethefinaldecisionthissummeronthesubstancesthatwillbedevelopedintomonographsandproceedthroughtheRoCevaluationprocess.

BucheremphasizedtotheboardhowtheNTPhasimprovedtheRoCprocess.“TheNTPmonographdevelopedforeachsubstancethatundergoesstudywillclearlyshowhowwereachedourconclusions.Itwillbeamoretransparentprocess.”

Gene variants linked with faster Parkinson’s disease progressionNIEHSgranteesreportthatParkinson’sdiseasepatientswithtwospecificvariantsofthealpha-synuclein(SNCA)geneprogressedtowardmotordeclinesignificantlyfasterthanpatientswithoutthesevariants.Thisworkcouldleadtonewtherapiesandhelpidentifythosewhowouldbenefitmostfromearlyintervention.

Theresearchersfollowed233well-characterizedParkinson’sdiseasepatientsincentralCalifornia,foranaverageof5.1years,andfoundthatcarriersoftheRep1263bpvariantoftheSNCAgenehadafour-foldhigherriskoffastermotordecline.PatientswithboththeRep1263bppromoterandrs356165variantshadanevenstrongertrendinprogressiontowardmotordecline.

Althoughthefindingsneedreplicationinotherwell-characterizedParkinson’sdiseasepopulations,theresearcherssaythattheirworkshowsthatthesegenevariantscouldbeusedtoidentifypatientswhowilllikelyexperiencefasterdiseaseprogression.Theworkalsopointstothealpha-synucleinpathwayasapromisingpotentialtherapeutictarget.

Theresearchersrecruited125maleandfemaleresidentdoctors,whoworkedatacentralBeijinghospital,toparticipateinthestudy.Participantshadneversmoked,werefreeofdisease,andhadanaverageageof24.Theyvisitedtheclinicbeforetheairpollutioncontrolswereinplace,whilethepollutioncontrolswereused,andafterthegameshadended.

DuringtheOlympics,thestudyparticipantsshowedsignificantreductionsinvonWillebrandfactorandsolubleCD62Plevels,whicharebothassociatedwithbloodcoagulation.SolubleCD62PandsystolicbloodpressurelevelsincreasedsignificantlyoncethepollutioncontrolswereliftedaftertheOlympics.

Pol II pausing modulates basal gene expression in signal transduction cascadesNIEHSscientistshaverevealedthatRNApolymeraseII(PolII)pausingdoesnotnecessarilyleadtohighergeneexpressionuponinductionofstimulus-responsivenetworks.Rather,itisimportantinmodulatingbasalgeneexpression.TheresearchoffersanewmodelforunderstandinghowpausedPolIIimpactsgeneexpressioninrestingcells.

UsingthesystemsbiologyapproachinDrosophilaS2cellsandfocusingprimarilyontheimmuneresponsepathways,theyfoundthatmanydownstreamtargetgeneswererapidlyinduced,despitenotharboringpausedPolIIattheirpromoterspriortopathogenicchallenge.Instead,theyobservedahigherprevalenceofPolIIpausingatpromotersofgeneswhoseexpressionlevelsweremoremodest.Genesinthelattercategoryencoderegulatorycomponentsandreceptorligandsthateitherinitiateordampenthesignalingcascade.

ThescientistsalsoknockeddowntheexpressionofNELF,acomponentrequiredforPolIIpausing,andfoundthatthemagnitudeoftheoverallimmuneresponsewasdiminished,regardlessofwhetherthegenesexperiencedPolIIpausingornot.Theirfindingsindicatethat,althoughtheregulatorycomponentsaremoremodestlyexpressedinresponsetoimmunechallenge,theirbasalexpressioninrestingcellsismoretightlyregulatedsothatthesecellsarebetterpoisedtorapidlyinitiatetheimmuneresponsecascadeasneeded.(SY)

intherestofthegenome.Thedatachallengedthetraditionallyheldviewthatmutationsoccurrandomly.Furtheranalysissuggestedthatthesemutationsoccurredatthesametimeinstretchesofabnormallylongsingle-strandedDNA.

Theresearchteamdevelopedbioinformaticstoolstodetermineifhumancancerscontainedsimilarmutationclustersand,surprisingly,nearlyhalfofthemdid.Thesemutations,however,werenotcausedbyenvironmentaldamage,butbyaspecificsetofproteinsknownasAPOBECcytosine-deaminases.APOBECproteins,whicharepartofthehumanimmunesystem,attackvirusesthatenterthebody.

Theresultsofthisstudysuggestthatseveralantiviraldrugs,capableofstimulatingAPOBECgenes,shouldbeconsideredpotentialmutagensastheyrelatetomutationclusters.(SP)

STAT6 and LRP1 polymorphisms are associated with food allergen sensitization in Mexican childrenAnewstudy,conductedbyNIEHSscientists,providesevidencethatthepolymorphismsinSTAT6,signaltransducerandactivatoroftranscription-6,andLRP1,low-densitylipoproteinreceptor–relatedprotein-1,genesareassociatedwithsensitizationtofoodallergensinasthmaticpatients.Asthmaticpatientsareatincreasedriskforsensitizationtofoodallergens,sotheresultsmaynotbecompletelygeneralizabletotheentirepopulation.

Priortothepublicationofthispaper,epidemiologistshadidentifiedfamilyhistoryasariskfactorforfoodallergies,butnogeneticvariantshadbeenconclusivelyidentifiedforfoodsensitizationorclinicalfoodallergies.Therefore,theresearchteamexaminedtheassociationsbetweenfoodallergensensitizationandsinglenucleotidepolymorphisms(SNPs)infiveautosomalcandidategenes:CD14,IL10,IL13,SPINK5,andSTAT6.

Thestudyincluded162asthmaticchildrenfromtheMexicoCityChildhoodAsthmaStudy,whotestedpositivetoatleastonefoodallergenusingskinpricktests,andtheirparents,usingthecase-parenttriaddesign.TheresearchteamfoundthatseveralSNPsinornearSTAT6,andtwomoreinthenearbyLRP1gene,wereassociatedwithsensitizationtofoodallergens.(AP)

Socioeconomic adversity in early life impacts the future risk of rheumatoid arthritisEpidemiologistsfromNIEHSreportthatwomenwithlowerchildhoodsocioeconomicstatusaremorelikelytodeveloprheumatoidarthritis(RA)inadulthood.Thestudyisthefirsttodescribeanassociationofmultiplechildhoodsocioeconomicfactors,suchashouseholdeducation,income,andmaternalage,withadultonsetRA.

Theresearchersanalyzedinformationfrommorethan50,000womenaged35-74fromtheNIEHSSisterStudycohort.Participantswereaskedaboutperinatalfactors,demographics,residentialhistory,lifestyle,medicalhistory,andmedicationuse.Thestudyshowedthatwomenwhowereraisedinahouseholdwithlowereducation,lowerincome,foodinsecurity,andyoungmaternalagewereathigherriskofdevelopingRAasadults.TheimpactofchildhoodsocioeconomicadversityonRAwasthemostapparentinwomenwithloweradulteducationalattainment.WomenwithfatherswhosmokedthreemonthspriortoconceptionalsohadahigherriskofRA,regardlessofsocioeconomicfactors.

Inside the InstituteZeldin welcomes 2012 summer interns to NIEHS By Ian Thomas

HighschoolandcollegestudentsfromacrossthecountrygatheredinRodbellAuditoriumfortheJune14kickoffofthe2012NIHSummerInternshipProgramatNIEHS,whichrunsfromMayuntilAugust.LedbyScientificDirectorDarrylZeldin,M.D.,themeetingprovidedparticipantswithasnapshotofwhattoexpectfromtheirtimeattheInstitute,followedbyalengthyquestionandanswersessionabouttheprogram.

“Overthenextseveralweeks,you’regoingtoexperiencealot,sodon’tbeshyaboutaskingquestions,”saidZeldin,whoopenedhistalkwithahandfulofentertainingphotographsfromhishighschoolandcollegedays.“Ultimately,yourtimeherewillbewhatyoumakeofit,sobeproactivewithyourmentorsandgetinvolvedineverythingthatyoucan.”

A plethora of experiencesThroughoutthesummer,studentswillworkalongsidemanyofthetopresearchersandstaffatNIEHS,contributingtoprojectsandconductinglabworkaspartofacomprehensive,hands-onlearningcurriculum.DuringtheirtimeatNIEHS,internshavetheoptionoffocusingonanynumberofenvironmentalhealthspecialties,suchasDNArepair,cancerbiology,bioinformatics,orclinicalresearch.

“Ican’twaittogetstarted,”saidHannahLeker,apublichealthmajorattheUniversityofNorthCarolinaatChapelHill.“Atthispointinmycareer,Ihaven’tcompletelydecidedonwhichdirectionIwanttogo,sothechancetogetinvolvedinalittleofthisandalittleofthatisreallyexciting.”

Thisnotionofbranchingoutintoavarietyofdifferentareasprovedtobeamajorthemeofthemorning,aspointedoutbyZeldin.

“Betweennowandthefall,you’llbeexposedtoeverythingfrombenchworkinalabtoclinicalresearch,someofityou’lllikeandsomeofityouwon’t,”saidZeldin,whoelaboratedonsomeoftheprosandconsofanM.D.versusaPh.D.,aswellasthetypesofcareerpathsthatareassociatedwitheach.“Thehopeisthat,bythetimeyouleavehere,you’llhaveamuchclearerideaofwhereyouwanttofocusyourstudiesgoingforward.”

Career exploration panel gives advice to summer interns By Ian Thomas

Participantsinthisyear’sNIHSummerInternshipProgramatNIEHSattendedaquestionandanswerstylecareerpanelmeetingcomprisedoffiveofenvironmentalhealth’sbrightestmindsJune19.FeaturingrepresentativesfromNIEHS,theUniversityofNorthCarolinaatChapelHill(UNC),andtheUniversityofMaryland(UMD),thepaneltreatedattendeestostoriesofwhatitmeanstobuildacareerinpublichealth,whileofferingthemthechancetoaskquestionsoneverythingfromdegreeprogramstomentorships.

“Regardlessofthecareer,there’snosubstituteforexperiencetohelpyoudecideonafield,”saidPerryBlackshear,M.D.,Ph.D.,headoftheNIEHSPolypeptideHormoneActionGroup,whoencouragedstudentstobeproactiveduringtheirtimeattheInstituteandbeyond.“Ifyouthinkyouwanttodoresearch,getinvolvedinalab.Ifyouwanttoworkinmedicine,volunteerwithaclinicandseeifyoulikeworkingwithpatients.Whatevertheinterest,takeadvantageofthechancetolearnaboutitfirsthand.”

Many roads, same destinationOnemajortopicofdiscussionduringthesessionwasthenotionthatscienceisadiversefield,comprisedofcountlesscareerpaths,specialties,anddegrees,notallofwhichareforeveryone.Thispointwasreflectedinthediversebackgroundsofthepaneliststhemselves.

“Iactuallybeganmycareerinbiology,thinkingIwantedtogotovetschool,”saidErinHopper,Ph.D.,directoroftraininginitiativesinbiomedicalandbiologicalsciencesatUNCandaformerpostdocatNIEHS(seestory).“EvenafterIchangedmymajortochemistryandearnedmyPh.D.,Iwasstillinterestedinacareerawayfromthebench.However,itwasn’tuntilIgottoNIEHSandstartedgettinginvolvedwithpeoplethatIfoundmywayintowhatIdonowincareerdevelopment.”

Remembering the basicsWhiledegreesandprogramsofstudywillbeamongthemajordecisionsstudentswillmakeintheyearstocome,thepanelistsagreedthattheyshouldn’tlosesightofthelittlethingswhenpreparingforacareerinscience.

“Nomatterwhatyouchoosetodoforaliving,takethetimetolearntoread,write,andspeakeffectively,”saidWilliamHiggins,Ph.D.,oftheUMDbiologydepartment.“Somanyofourstudentstodaydon’tspendenoughtimedevelopingthesesimplestofskills,andthey’reabsolutelyvitaltosuccessinanyfield,publichealthincluded.”

The value of role modelsAsmanyofthesestudentscontinuetoexploretheiroptionsforpotentialcareers,thepaneliststoldtheinternsthatsurroundingthemselveswiththerightpeopleiscrucialtotheprocess.Nowhereisthatmoreimportantthanintheselectionofamentor.

“Beasselectiveaboutchoosingyourmentorastheyareaboutchoosingyou,”notedBlackshear.“Differentpeoplementorindifferentways,justasdifferentpeoplelearnindifferentways.Findonethatbestmatcheswhoyouareandgettoknowthem.”

“Whetherit’syourmentors,yourteachers,oryourfriends,surroundyourselveswithpeoplewhoexcelatwhattheydoandtakenotes,”addedHiggins.“Learningwhatmakesthemsuccessfulandintegratingthatintohowyoubuildyourowncareercouldonedaytakeyoutosimilarheights.”
