Spent grain wax - the key ingredient of STIMU-TEX® AS
For generations, beer brewers have been aware of the ability of spent grain wax to soothe skin and relieve itching.
Dermatological studies1 performed on skin with atopic conditions have confirmed this traditional knowledge. Equally outstanding results were achieved when a spent grain extract was applied to skin through bath-therapy or in a cosmetic cream. These treatments not only alleviated itching significantly, they also stopped it recurring.
But what exactly is spent grain wax? It is what remains once pure barley (Hordeum vulgare) has been progressively transformed into malt through several stages of physical and fermentative processing. DSM obtains spent grain wax through extraction from malt using supercritical CO2. This high-tech extraction process is particularly gentle and produces golden, 100%-pure spent grain wax which is free from organic solvents and preservatives. It contains large amounts of unsaturated fatty acids, which are essential for the regeneration of the epidermal barrier and for maintaining homeostasis in the skin.
Argan oil has been valued for its numerous health and beauty benefits for centuries.
This oil is obtained from the seeds of Argania spinosa, a tree endemic to southwest Morocco. It is known to the Berbers living there as the “Tree of Life”. Moroccan women have traditionally regarded argan oil as a beauty elixir, using it daily in topical applications for skin, hair and nail care, and also integrating it into their diet.
Our gentle cold-pressing process provides a perfectly balanced oil which is highly concentrated in active compounds. Up to 80% unsaturated fatty acids, combined with polyphenols, tocopherols, phenolic acids, and squalene give it a unique profile and make it highly valuable for skin care. Its nourishing, regenerating, and protecting qualities help maintain skin’s youthful assets.
Shea butter, the “African wonder,” has been used in Egypt for at least 2,000 years for its soothing properties.It is believed that Queen Nefertiti owed the magnificence of her beauty to the daily use of shea butter, which contains extraordinary skin healing, regenerating, and emollient components.
The shea tree itself can live for as long as 300 years. The extraction of the fruit’s kernels provides the butter, which consists mostly of fatty acids – linoleic acid in particular – and unsaponifiables such as phytosterols called sheasterols, which stimulate healthy cellular processes and cell regeneration.
Histamine effect blocking assay
Result:STIMU-TEX® AS nearly completely blocks E-selectin expression on endothelial cells, a downstream target of histamine.
Efficacy in vitro
Histamine is a marker for allergic reactions. Histamine-blocking products inhibit the symptoms of allergy such as itching, swelling, and red skin.
An in vitro assay with human endothelial cells was developed to quantify histamine effects: Histamine combined with TNF (Tissue Necrosis Factor) enhances the induction of E-selectin on endothelial cells. E-selectin induction is the starting point of an inflammatory reaction.

Result:STIMU-TEX® AS significantly decreases the expression of IL-1α by 58%, a marker for inflammatory reactions.
Determination of the inflammation marker interleukin-1α, released in the supernatants of stripped human skin disks.
A human skin fragment was taken after abdominal plastic surgery and stripped 10 times. STIMU-TEX® AS was tested on the skin surface at a concentration of 1 mg/cm2. Dexamethasone is a glucocorticoid, used as positive control.

Efficacy in vivo
Histamine alleviating efficacy
The release of histamine induces a vasodilatation, an increase of vascular permeability, inflammation, and skin redness. Laser Doppler flow measurements enable this increased blood flow to be quantified.
Histamine was applied on the inner forearm of 10 healthy female volunteers between the ages of 18 and 39. Penetration of histamine into the skin was stimulated non-invasively by iontophoresis in order to produce a mild reaction. Results for distilled water (negative control), histamine alone (positive control), and 5% STIMU-TEX® AS with histamine were compared.
The reactions were determined non-invasively as perfusion modification by means of a laser Doppler imager and as wheal formation by computerized surface measurement.
