Measuring SO2: A Perennial Issue

SO2 in aqueous solutions

• When SO2 gas is added to water, some of the SO2 is hydrolyzed to form the acid H2SO3hydrolyzed to form the acid H2SO3.

SO2 in wine or juice

• H2SO3 is a relatively strong acid, so at wine pH, most of the H2SO3 is ionized to form HSO3

• Similarly, as the conjugate base of a strong acid, some of th HSO - f d f t ill i k t tthe HSO3 formed from meta will pick up a proton to become H2SO3

• At pH 3.0, about 7% of the SO2 will be in the “molecular” form, the remainder will be primarily HSO3

• At pH 4.0, less than 1% of the SO2 will be in the “molecular” form

SO2 Adducts

• HSO3- will readily combine with a variety of compounds

Adduct Stability

• Anthocyanin adduct is not stable—it will hydrolyze readily with acid addition or to re-establish equilibrium ifreadily with acid addition or to re establish equilibrium if free SO2 is consumed

• Sugars, pyruvate & alpha-keto-glutarate are moderately stable, but will hydrolyze with acid and to re-establish equilibrium.

Free SO2 analysis schemes

• Most commonly used analyses start with an acidification step to convert the free SO2 to the molecular formstep to convert the free SO2 to the molecular form– Direct titration or other reaction of the molecular form

Release of bound SO2

• Both Ripper and A/O start with an acidification step

• As many SO2 adducts are acid labile, some bound SO2is released during these analyses

• “Free” SO2 can better be characterized as “free” plus some dissociated bound SO22

• The amount of dissociated bound SO2 depends on the types of adducts present in the wines and the speed oftypes of adducts present in the wines and the speed of the analysis

Flow Injection Analysis for SO2

• Again usually based on reacting SO2 with a reagent to form a colored productform a colored product– Pararosaniline is common but there are others

• The first step is often to acidify the sample, forcing the SO2 to the molecular form, which is then separated from the wine matrix via a gas permeable membranethe wine matrix via a gas permeable membrane

• The color-forming reagent is then added, followed by a g g , ymixing/incubating step. The colored product is then measured by the instrument’s spectrophotometer
