Services Metabolomics and proteomics in the LIFE research program

The coupling of liquid chromatography and mass spectrometry enables rapid and highly sensitive analysis of biologically active metabolites, peptides, and proteins in body fluids. Within the LIFE research program a mass spectrometric platform for metabolom and proteom analysis has been established, allowing targeted and simultaneous quantitation of metabolites and proteins. By means of ultra-high per-formance liquid chromatography (UHPLC) in combination with quadrupol linear ion trap mass spectrometry, it is possible to detect and quantitate metabolites in con-centration ranges of ng/L.

In the past years method development focused on metabolic pathways that are related to vascular and cardiovascular di-seases such as arteriosclerosis or cardiac insufficiency, as well as diabetes and de-mentia. Standardized preanalytical proto-cols covering sample drawing, handling, and sample preparation were developed for ensuring reliable quantitation of partly very low abundant metabolites and pro-teins in body fluids. Methods developed in the mass spectrometry workgroup were validated for the analysis in body fluids and were partly accredited according DIN EN ISO 15189:2014 and 17025:2005.

In the LIFE research program, samples from patient popula-tions are used to create biomarker profiles paving the way for identifying novel candidate genes for civilization diseases in the line of genome-wide association studies (GWAS). Aim of method development is their potential future application in the clinical routine laboratory. Since the beginning of the year 2016, the simultaneous quantitation of nine steroid hormo-nes via liquid chromatography tandem mass spectrometry is pioneeringly and invariably applied to routine diagnostic ana-lysis at the Institute of Laboratory Medicine, Clinical Chemistry and Molecular Diagnostics of University Hospital Leipzig AöR.
