Fluka’s LC-MS and LC-MS Ultra CHROMASOLV® solvents offer high purity and low levels of alkali metal impurities, making them ideal for classical MALDI-MS sample deposition techniques and in LC-MALDI-MS coupled methods.

LC-MS Ultra grade solvents are tested specifically for UHPLC, offering extremely low baselines, high lot-to-lot reproducibility and minimal adduct formation due to high quality leaching-resistant packaging materials.

Additionally, ready-to-use blends with 0.1% of trifluoroacetic acid (TFA), ammonium actetate or formic acids are also available

To improve sensitivity and reduce interferences in the mass spectrum, additives or co-matrices are often added to the sample-matrix-solvent mixture. Trifluoroacetic acid, formic acid and various ammonium salts are most commonly used in this respect.

Derivatization of sample analytes, for instance permethylation or peracetylation, can have advanta-geous effects in MALDI-MS, such as increasing analyte stability or improving the ionization efficiency.

Developed in collaboration with the Protein Quantitation Consortium, Sigma’s AQUA Peptides enable accurate, efficient mass spectrometric quantitation of protein biomarkers.

• Focus your analysis on significant protein biomarkers

• Accurately quantitate low abundance proteins

• Eliminate costly, time consuming stable isotope labeling steps

• Measure site-specific phosphorylation states

• Validate gene silencing at the protein level

Stable isotope labeling with amino acids in cell culture (SILAC) is a straightforward approach for the incorporation of a stable isotope label into cellular proteins for use with mass spectrometry-based quantitative proteomics. This method works by comparing cell cultures treated with light isotope (natural abundance) amino acids and heavy isotope (13C, 15N and/or D) labeled amino acids which produces two almost identical proteomes that only differ by their masses.

Although not as commonly connected with MS as other separation methods such as LC, thin layer chromatography is a facile and fast separation method that can quite easily be combined with subsequent MALDI-MS analysis. TLC-MALDI-MS has been successfully applied to a large variety of different sample and analyte types, including proteins, oligonucleotides and carbohydrates.
