Glucose may be oxidized completely if sufficient oxygen is available and is summarized by the following reaction: " $% " + 6 % 6 % + 6 % + All organisms, including plants and animals, oxidize glucose for energy. Often, this energy is used to convert ADP and phosphate into ATP. It is known that peas undergo cell respiration during germination. Do peas undergo cell respiration before germination? Using your collected data, you will be able to answer this question concerning respiration and non-germinated peas. Using the CO2 Gas Sensor, you will monitor the carbon dioxide produced by peas during cell respiration. Both germinated and non-germinated peas will be tested. After your preliminary investigation, you will investigate how various factors may influence the rate of cellular respiration. OBJECTIVES In this experiment you will Use a CO2 Gas Sensor to measure concentrations of carbon dioxide during cell respiration Determine whether germinating peas and non-germinating peas respire Compare the rates of cell respiration in germinating and non-germinating peas Design your own experiments to measure rates of respiration of different seeds and organisms as well as the effect of several variables including salinity, germination time, and temperature MATERIALS LabQuest 2 Germinated and Non-germinated Peas Vernier CO 2 Gas Sensor Various Seeds (both Germinated and Non-germinated) 250mL Respiration Chamber Ice Thermometer Hot Water Bath Crickets Mealworms

PROCEDURE

1. EnsureyourCO2GasSensoryisproperlyconnectedtotheLabQuest2andchoose“New”fromtheFileMenutosetupanewexperiment.

2. RecordtheroomtemperatureinTable1.Askyourinstructorforthecurrenttemperature3. Preparefordatacollectionbychangingthecollectionmodeto“TimeBased”.Settheintervalto4

seconds/sampleandthedurationto300secondsasinthepicturebelow:

4. Obtain25germinatedpeasandblotthemdrybetweentwopiecesofpapertowel.Placethe

germinatedpeasintotherespirationchamberasshowninthefigureonthepreviouspage5. PlacetheshaftoftheCO2GasSensorintheopeningoftherespirationchamberasinFigure16. Waitonetotwominutes,thenstartdatacollection.Datawillbecollectedfor5minutes.7. Whendatacollectionhasfinished,agraphofcarbondioxidegasvs.timewillbedisplayed.8.
