Bei der 1. Reaktion des Pentosephosphatwegs wird Glucose-6-P zu Gluconsäure-6-P, dehydriert.Hier werden also 2H frei, die auf das freie NADP übertragen Werden.( Im Gegensatz zu dem NAD wird NADP dann verwendet, wenn es etwas zu synthetisieren gibt – in diesem Fall z.B. Steroidbiosynthese.)
Das Enzym: Glucse-6-P-Dehydrogenase, ein regulatorisches Enzym (welches z.B. durch Acyl-CoA-Überschuß gehemmt wird!!)

Der 3. Schritt, Gluconsäure-6-P zu 3-Ketogluconat-6-P, bei dem das 2. NADPH+H entsteht, wird katalysiert von dem Enzym Gluconsäure-6-P-Dehydrogenase.Dieser Vorgang ist deshalb nennenswert, da der Pentosephosphatweg auch auf dieser Stufe durch dieses Enzym reguliert werden kann.Also auch die Gluconsäure-6-P-Dehydrogenase ist ein regulatorisches Enzym!!!
