Sensitive Enzyme-Immunoassay for Progesterone inHuman Plasma and Saliva

High sensitivity enzyme-immunoassays (EIA) forsteroid hormones are achieved by the use ofheterologous systems, sophisticated purification ofthe enzyme-coupled hormone by affinity chromato-graphy (1) and a new type of methodology, thefluoro-enzyme-immunoassay (2). A /?£^rologousEIA for progesterone covering the range of0.1-10 pmol/test tube (0.03-3 ng) (3) was improvedto measure high sensitivities of 10-1000 fmol (3—300 pg) by simply changing the test conditions butstill using a normal photometer. Results with thisEIA are shown for the measurement of physiologicallevels or in follow-up therapeutic treatment:
a) Sensitive range (10—1000 fmol) for determinationin human plasma and in saliva. Control of treatmentof infertility. The level in saliva is only about 1/100compared to plasma.
b) normal range (0.1 —lOpmol) for determinationof progesterone in pregnancy. This also seems to be

possible by direct determination in serum withoutextraction of the progesterone.
The values of direct assay agree well with reportedhormonal levels, for example:
7. week 76 nmol/1 (24 jig/1);10. week 105 nmol/1 (33 fig/1);17. week 150 nmol/1 (47 fig/1);29. week 225 nmol/1 (70 ng/1);38. week 550 nmol/1 (170 ̂ ig/1).
