4. Explanation of procedure:
New Zealand White rabbits are being utilized as animal models for these infections. This project characterizes the genes required for the systemic disease caused by rabbitpox. Rabbitpox is lethal to rabbits, while vaccinia, which is over 95% identical at the gene level to rabbitpox, causes only a local lesion and is used as a vaccine. The goal of this experiment is to identify the genes required for the systemic infection of rabbitpox in order to have potential targets for antiviral therapies in the treatment of other human pathogenic poxviruses such as smallpox and monkeypox. Since the differences in pathogenesis cannot be seen in cell culture the use of animal models is necessary.
The first part of this work (experiment #1) examines the dosage and verifies the differences in disease caused by vaccinia and rabbitpox virus in rabbits. Specifically, the project re-determines the dose of rabbitpox virus that causes systemic disease and verifies that comparable or higher doses of vaccinia virus do not cause systemic disease. The critical part of experiment #1 will be to determine whether rabbits infected with mixtures of vaccinia and rabbituox viruses develop systemic disease, with accompanying secondary lesions typical of rabbitpox. This last experiment mimics the clinical situation in which humans are vaccinated after exposure to smallpox. These results help determine effective vaccine regimens. The experiments require minimum manual restraint of the rabbits, shaving of both hind flanks and the intradermal inoculation of lOOul buffer containing the corresponding plaque forming units of virus, either alone or as a mixture. Following infection, animals are routinely monitored two times a day (early morning and late afternoon), until or unless symptoms of disease develop. Upon onset of symptoms, the animals are closely observed four times a day (early morning, mid-day, late afternoon and late evening) for signs of severe disease (as indicated by decreased respiratory rates, dyspnea, open-mouth breathing, severe lung sounds, temperature equal to or exceeding 106"F, anorexia (not to exceed four days), dehydration, andlor morbidity). At this point the animals are euthanized according to the AVMA Panel on Euthanasia. A set of criteria for scoring the level of disease in the animals has been created. Upon euthanasia, the animals undergo necropsy to examine the spread of the virus to secondary sites and for the isolation of viruses for further characterization.
The dominance of rabbitpox virulence upon infections with mixtures of vaccinia and rabbitpox viruses provides a system for the identification of the rabbitpox virus genes, which particularly dispose the virus to cause systemic disease in rabbits. Identification of such genes provides the possibility of developing antiviral drugs and immunological reagents against these specific virulence determinants.
Experiment #2 is designed to infect rabbits with mixtures of recombinant vaccinia and rabbitpox viruses in order to identify rabbitpox virus genes associated with
systemic disease. Recombinant viruses from cell culture are isolated and then used to infect rabbits. The recombinants capable of causing systemic disease can then be isolated from the secondary lesions typical of systemic disease. In the event virulence of rabbitpox virus in mixed infections of vaccinia and rabbitpox viruses is not dominant, and is attenuated during mixed infections, then specifically engineered vaccinia virus containing segments of rabbitpox virus will be constructed, purified and assayed in rabbits for increased systemic disease. The experiment requires minimum manual restraint of the rabbits, shaving of both hind flanks, and the intradermal inoculation at up to five sites on the flanks of lOOul buffer containing the recombinant viruses. Following infection, animals are routinely monitored two times a day (early morning and late afternoon), until or unless symptoms of disease develop. Upon onset of symptoms, the animals are closely observed four times a day (early morning, mid-day, late afternoon and late evening) for signs of severe disease (as indicated by decreased respiratory rates, dyspnea, open-mouth breathing, severe lung sounds, temperature equal to or exceeding 106OF, anorexia (not to exceed four days), dehydration, and/or morbidity). At this point the animals are euthanized according to the AVMA Panel on Euthanasia. Upon euthanasia the animals undergo necropsy to examine the spread of the virus to secondary sites and for the isolation of viruses for further characterization.
Experiment #3 represents an alternative method for identification of rabbitpox virus, which predisposes the virus to virulence. The third set of experiments is to replace the identified gene segments of rabbitpox responsible for virulence with the corresponding genes in vaccinia and assay for attenuation after the injection of the recombinant viruses into rabbits. Attenuation of this engineered rabbitoox virus - strain is recognized by the appearance of a local lesion with no accompanying systemic disease. Following infection, animals are routinely monitored two times a day (early morning and late afternoon), until or unless symptoms of disease develop. Upon onset of symptoms, the animals are closely observed four times a day (early morning, mid-day, late afternoon and late evening) for signs of severe disease (as indicated by decreased respiratory rates, dyspnea, open-mouth breathing, severe lung sounds, temperature equal to or exceeding 106"F, anorexia (not to exceed four days), dehydration, andor morbidity). At this point, the animals are euthanized according to the AVMA Panel on Euthanasia. 5. Justification:
Analgesics are not indicated in these studies because their effects on the inflammatory and immune responses hinder the distinctions observed in the two viral diseases. It is critical that the rabbitpox and vaccinia viral diseases be distinguishable. For the screening of recombinant viruses it is also critical for the disease pathways to be uninhibited for the detection of virulent viruses.

Proposed experiments in rabbits: 1. Determining the lethal dose of rabbitpox in rabbits.

Experiment 1 determines the dosage of rabbitpox required to lethally infect rabbits. This requires manually restraining the rabbits and infecting them with rabbitpox by intraderma1 injection.-i he site ofinoculation will be a shaved region of each hind flank. Following infection, animals will be routinely monitored two times a day (early morning and late afternoon) until symptoms of disease develop. Upon onset of symptoms (see criteria below), the animals are closely monitored four times per day for signs of severe disease (as indicated by decreased respiratory rates of 25 bpm or less, difficulty breathing, open-mouth breathing, severe lung sounds, temperature equal to or exceeding 106'F, loss of appetite [not to exceed four days], and dehydration) at which point the animals are humanely euthanized.
A set of criteria for scoring the level of disease in the animals has been created to determine the time when the project shifts monitoring from 2 to 4 times a day and to determine when euthanasia is performed.
Experiment 2 determines the efficacy of LC16m8 against lethal rabbitpox challenge in rabbits. Dryvax and placebo are also being tested as positive and negative controls, respectively. This requires vaccinating the rabbits and then subjecting them to challenge by infecting them with rabbitpox. Briefly, the rabbits are vaccinated by scarification and are challenged with rabbitpox intradermally at day 28. Challenge is being done at day 28 since it is the average time at which - - - neutralizing antibodies are at peak titers in response to a vaccine.
Considering that this is the first time that vaccinia-based vaccines are being tested for their abilities to neutralize rabbitpox, it is necessary that a range of concentrations of challenge rabbitpox be used in the study. This circumvents the possibility that could occur if we only used one concentration of challenge rabbitpox, where the chosen concentration simply overwhelms even an efficacious vaccine. Therefore, the vaccine-challenge study is done using the two lowest lethal concentrations of rabbitpox (referred to as "lowest lethal pfu" and "second lowest lethal pfu," respectively) as determined in Experiment 1.
As detailed above, the animals are closely observed, up to four times a day, using the guidelines outlined in the first experiment above, for signs of severe disease (as indicated by decreased respiratory rates of 25 bpm or less, difficulty breathing, open-mouth breathing, severe lung sounds, temperature equal to or exceeding 106OF, loss of appetite (not to exceed four days), and dehydration, at which point the animals are humanely euthanized.
For endpoint analyses, blood is drawn from vaccinated rabbits at baseline, day 14 and 28 (pre-challenge) to measure neutralizing antibodies against rabbitpox in vitro. Also, internal organs are excised from a specific number of rabbits from each vaccine arm at day 38 (i.e., 10 days post-challenge, last day of the experiment) to determine the influence of vaccination on challenge rabbitpox titer in these organs.
5. Justification:
Rabbitpox is a lethal infection in rabbits. The rabbits will be vaccinated with LC16m8, Dryvax, or placebo. They are then injectedscarified with rabbitpox to determine whether vaccination protects the rabbits against a lethal rabbitpox infection. Therefore, providing these animals with any other form of anesthesia, analgesia or treatment will not be appropriate. It is necessary for the project to determine whether the animals develop severe symptoms, as that is the only way to tell whether the vaccine treatment is working. Presumably, if they "don't" develop severe symptoms, the vaccine is working as expected.

IACUC reviewed, discussed and approved the following exemptions from USDA Policy.
Protocol #01-078 - Granted an exemption for not using analgesia in Hamsters and Rabbits for antibody production with Freund's adjuvant. The IACUC reviewed, discussed and approved the exemption fiom USDA Policy. Protocol #01-173 - Exemption granted for Non Human Primates (NHPs) for IV injection and subsequent hydration for 2-12 hours. Animals are observed continuously and provided additional environmental enrichment while restrained. Protocol #02-241 - Exemption granted for pig restraint by sling for a total of 5 hours. Animals are observed continuously while restrained. Protocols #02-257, 03-130 and 03-131 - Exemption granted for restraining board restraint for NHPs for 1 hour for IVGTT and 2-12 hours for STZ injections or critical care as needed. Animals are observed continuously and provided additional environmental enrichment while restrained. Protocol #03-069 - Granted an exemption for not using analgesia in Rabbits for antibody production with Freund's adjuvant. The IACUC reviewed, discussed and approved the exemption from USDA Policy. Protocol #04-171 - Exemption granted for streptozotocin injection: the animal will be placed on the restraining board, injected with ST2 and dehydration for 2 hours post injection. The animal will be placed in a cage, and 8-14 hours after the injection - when hypoglycemia occurs-the monkey will be placed on the restraining board for an additional 2-12 hours to administer glucose and maintain normoglycemia. Metabolic challenges: (glucose, arginine, glucagons) the monkey will be sedated with ketamine, placed on restraining board and maintained there for up to 1-2 hours to collect samples. Thyrnoglobulin fludarabine administration: The first dose of thymoglobulin is administered over a 4 hour period, the 2nd over a 3 hour period and subsequent doses over a 2 hour period. Each antibody is considered in the context of known side effects of administration (or lack thereof). Mabs are given over a time period up to 1 hour that prevents documented side effects in NHP or humans. Animals are observed continuously and provided additional environmental enrichment while restrained. IACUC Committee approved an exemption to individually house a rhesus monkey with individually housed cynomologus monkeys. Common infectious agents have been ruled out and compatibility has been established.

These animals are pre-diabetic models wich predisposes :hem to a high likelihood of the development of cardiovascular conditions, such as atherosclerosis, and other co-morbidities, such as neuropathy, blindness, dermatitis, and urinary tract infections, which have the potential to produce pain, discomfort, or distress.
5. Provide scientific justification why pain and/or distress could not be relieved. State methods or means used to determine that pain andlor distress relief would interfere with test results. (For Federally mandated testing, see Item 6 below)
The natural aging process and development of diabetic-related conditions and associated pathologies can potentially produce pain, discomfort, or distress. Veterinarians are always consulted when pain, discomfort, or distress is recognized. All pathological processes associated with thb agin and pre-diabetic condition are evaluated and those that are amenable to current veterinary medical care standards are promptly placed on appropriate pharmacological therapies.
