The study on hand aimed at assessing the practical value of the pdymerase chain reactkm (PCR) fw both the diagnosis of w-tine lekhmaniosis (CL) end the followup after the pentavalent antimonisl n-methyigl-ine puinee&Jgueel~ndaswell.

A specific PCR ampliing a lo0 bp fragment of Lek+unenie infenfum (Pianwx et al., 1993) ws cwried out. DNA isolation of the blood (PBL) or bme m- (BM) vas performed using a cummercielly available kit (CtlAmp Blood Kite). Out of 18 dinically confirmed cases of CL, 13/15 PBL- ad 14l14 BM-samples prwed to be posittve priw to treatment.

Fulhermwe, e total of 16 cases of CL could be monitored efter treatment wing the PCR. The treatment gmups consisted of: (1) Six dogs treated ti n-methytglucamine (day 1-2: 50 rnoncg tnv end day 510: 100 rng/lQ bw up to 3 bwtment cycles). (2) Three dcgs treeted tith allopuiinoi (2x10 mgkg kfdey for 5 weks). (3) Fw dogs received ellopuinol (3 to 5 weeks, instXficient impwement) and subsequently n-methytglucamine. (4) Three dogs received aliop$rml fcf 5 weks up to 20 months after en initial course of Glucmtime

One dog receiving a Glucantlmeetherapy and 2 dogs each with en allopwtnol or a combination therapy, have so far been clinically healthy end PCR-negative in both the PBL end BM. Although the clinical state of the remaining 11 dcgs improved. relapse may be expected since blood cf bone marrow wre still PCR-positive after treatment.
