ACADEMIC PROJECTS
Title: Biophysical studies on human prion proteins (90-231) through IASC. Banglore At IISER mohali. Period: 2 monthsDescription: human prion is a long length protein ranging from 90-231 .
The recombinant protein corresponding to human prion protein domain encompassing residues (huPrP90-231) was expressed in E.coli as inclusion body and purified to homogeneity. Spectroscopic data indicate that the conformational properties and the folding pathway of humanPrP(90-231)are strongly pH dependent. Acidic pH induces a dramatic increase in the exposure of the hydrophobic patches on the surface of the protein The secondary structure of the human PrP(90-231)is largely alpha-helical and after transition the stable structure rich in beta-sheet is observed. These findings have important implications for understanding the initial events on the pathway towards the conversion of the normal into pathological form of prion protein.
