Nu ht phn giai on pht trin thch hp th cc bao phncn li ca n hoa c dng trong nui cy Cc bao phn c th c nui cy theo 2 cch: Trn mi trng MS c (c 0,6-1,0%(W/v) agar) Nui cy ni trn mi trng lng gm: KNO3-950(mg/l); NH4NO3- 825(mg/l);MgSO4.7H2O- 185(mg/l); CaCl2- 220 (mg/l);KH2PO4-85 (mg/l); FeSO4.7H2O-27,8 (mg/l); Na2EDTA-37,3 (mg/l);saccarozo-20000 (mg/l)6. Qu trnh nui cy c duy tr nhit 25oC trong ti hoc nh sng yu.Cng nh sng thch hp khong 300lux. Sau 4-6 tun nui cy, cc cy con s xuthin t mt s bao phn.

    Khi m bao phn t 3mm th tch cy con ra khi m v chuyn sang mi trngto r (c th dng mi trng MS vi thnh phn khong gim 1 na). Cng nhsng cao hn thi gian chiu sng khong 10-12h/ngy

    3.2. Bin php ngng sinh trng tm thi

    y l phng php bo qun di hn cc vt liu trong hoc trn b mtnit lng (- 1760C n - 1960C ).

    Bo qun ng lnh trong, trn nit lng c th c chia ra cc cng onsau:+ Tin sinh trng, x l vi 5 - 10 % proline, 3 - 6 % manitol.+ X l bo v chng ng bng.

    3.2. Bin php ngng sinh trng tm thi a vo bo qun trong hoc trn mt nit lng (- 1960C 1580C).

    Lm tan tuyt, x l nhit 35 400C trong iu kin phng th nghimbnh thng.

    Mi trng nui cy:Trong th nghim s dng mi trng vo mu l MS + 30g

    saccarose/l + 6.5g agar/l, 1ml NAA, pH l 5.7

    Tin hnh:Mu sau khi thu thp c a v phng th nghim, tin hnh kh

B sung thm 2 ml dung dch ra ra proptoplast ht khi li lc l. Chuynhn hp protoplast-enzyme (tng cng 15 ml) vo tube ly tm loi 15 ml v ly tm 50gtrong 10 pht.

    Loi b phn dch ni pha trn, ti huyn ph tiu th bng 10ml dung dchtch (PI + 20% sucrose), thm 1ml dung dch ra ln trn b mt dung dch tch vprotoplast sao cho khng lm ha ln hai dung dch, tt nht l to thnh hai pha (nh tt dung dch ra bn thnh tube trnh bn tung to ln dch ra bn di), ly tm 50g

    trong 10 pht. Cc protoplast sng st s nm trn b mt dung dch .

    Dng micropipette ht lp protplast mu xanh lc ni trn tube ly tm v chuynn sang mt tube ly tm sch, thm 10 ml dung dch ra v ly tm li. Protoplast s lngxung y tube v c dng vin.

    Ra protoplast thm mt ln na trong 10 ml dung dch ra. Loi b phn ni trnmt v ti huyn ph protoplast trong khong chng 1 ml mi trng nui cy protoplast(PC).

Cy c bo qun nhit 28oC, cng nh sng1000 lux trong 16 gi hng ngy bng n hunh quang.- Sau 1 tun dng knh lp kim tra xem chi ghp c sng khng. Nu chighp sng, ch 1 thng sau c thm 2 l mi, t tiu chun ghp ln th 2.
