Tissue engineering scaffoldsplay avital role in regenerative medicine.

It not only provides a temporary3 dimensional (3D) support during tissuerepair, but also regulates the cellbehaviour, such as:-Cell adhesion-Cell proliferationand-Cell differentiation Scaffold should have the key

characteristics:

- highly interconnected porous structure(which allowscell penetrationandnutrient& waste transportation)

- Biocompatibility and biodegradability(which are the basic requirement for the biomaterials used forscaffolds)

- Suitable mechanical propertiesto meet the specific applications- Appropriate surface modification and topographyto support cell adhesion and growth

Why Calcium Carbonate filled Hydrogel as Scaffold?

Guided bone regeneration (GBR) method is a well establishedtherapy to repair mandible and alveolar bone defects infected byperiodontal diseases.

The principle of GBR method is to prevent an invasion of non-functional scar tissues.

GBR membranes also have an important function whichencourages bone growth.

GBR membranes are occasionally utilized with dental implant orbone grafting materials.

The basic principle of Guided Bone Regeneration (GBR) involves the placement of mechanical barriers to protect blood clots and to isolate the bone defect from the surrounding connective tissue, thus providing bone-forming cells with access to a secluded space intended for bone regeneration.

GOAL

The development ofresponsive biomaterials capable of demonstratingmodulated function in response to dynamic physiological and mechanicalchanges in vivo remains an important challenge inbone tissue engineering.

Many natural and synthetic hydrogels are already reported as havingpotential biological applications and considered as promising material for tissueengineering, asscaffoldsfor bone-tissue and dental transplants.

Further, inspired bynatural biogenic minerals, many researchers haveattempted tomimic natural biomineralization through organic-inorganichybridization in order to developa new material for medical applications.

Actually, biomimetic chemical compositionsand structures in the bone-regenerative scaffoldshave shown enhanced ability for cell biomineralizationand osteogenesis.

Thus, attempted has been taken for the preparation of biomimetic compositionbased biomaterial scaffold (i.e.Biomineralized PVP-CMC Hydrogel) forbiomedical applications

The morphological image confirms the deposition of calcium ions in thePVP-CMC matrix. The crystals obtained are granular in shape which increaseswith mineralization time. After filling up the pores of hydrogel matrix, theCaCO3 are deposited on the surface.

Swelling study of MPC (varying thickness) was performed in presence ofphysiological solution at 37oC to explore the potential biomedical applicationslike drug delivery and /or for the treatment of a dynamic bone disorder etc.

Further, the rheological properties of the MPC explained the elastic andviscous nature of the developed material at low and high strain. The linearnature clearly indicates the solidity of the polymeric material. Also, when thestrain increases from 1% to 10% the elasticity of the material reduces andslowly viscous nature is observed.
