Restriction enzymes and their boschizomers

INTRODUCTIONSince the last compilation of restriction enzymes (1), 300 new entries have been addedincluding 12 new specificities. With the growing size of this database and the recognitionthat the most widespread use of the information is as a database for computer programspredicting restriction enzyme cleavage patterns, the new format has been continued. Thisformat is intended to contain the minimal amount of information required by a computerprogram. It should be noted that only enzymes for which the recognition sequence is knownare included. This new list is shown"in the first Table, while an alphabetical listing ofall Type II enzymes is presented in the second Table. A copy of the restriction enzymedata base in its previous format (2), including enzymes of unknown recognition sequence,will be available upon request. It should also be noted that an alternative compilation ofthese enzymes has recently been produced (3).
The database shown in these Tables is available online through the BIONET computerresource. A version corresponding to the printed text is located in the file< ROBERTS > RESTRICT.NAR Several alternative versions are available and aredocumented in < ROBERTS > RESTRICT.DOC. It is also possible to get regular monthlyupdates by electronic mail. Enquiries should be directed to roberts@cshlab.bitnet.
In forming this list, all endonucleases cleaving DNA at a specific sequence have beenconsidered to be restriction enzymes, although in most cases there is no direct geneticevidence for the presence of a restriction-modification system. The endonucleases are namedin accordance with the proposal of Smith and Nathans (4).
