The VariantPro™ system is powered by a

Recent advances in next-generation sequencing
(NGS) technologies are providing new
opportunities to characterize individual
genomes and identify causal mutations relevant
for diagnosis and therapy. Though NGS is
capable of very high levels of coverage even on
complex genomes, it is still advantageous to
reduce the complexity of samples and sequence
smaller targeted regions – in particular, when
sample numbers are very high and the goal is
detection of less prevalent or extremely rare
mutations. Targeted sequencing has become
the method of choice for variant analysis due to
more moderate costs, manageable data
amounts and straightforward interpretation of
analysis results.

perform quantitative prediction of PCR primer

VariantPro™ Targeted Sequencing
Technology
In a single tube, a pair of common primers are added into a mixture of multiple pairs of specific
primers and genomic DNA to form a single step, two stage multiplex PCR reaction.
A single PCR run accomplishes two functionally separated reaction phases, namely target replication
and library amplification, without primer removal step. This results in a significantly simplified work flow.
The addition of the common primers induces a profound change to multiplexing PCR by limiting the role of specific primers to only
the first two thermo cycles for target replication and enabling an automatic switch to common primers in the remaining thermo
cycles for library amplification. This arrangement eliminates a significant cause for amplicon to amplicon variations in end product
due to priming efficiency variations among specific primers which would be exponentially amplified if used as amplification primers.
An Omega shaped primer is composed of three functional sections: a 5’ arm designed
to ensure stable binding to a corresponding template, a 3’ arm designed to double
check sequence specificity and initiate polymerase extension, and a separation
segment between the two arms. This segmented primer design provides desirable features and/or properties for targeted
sequencing applications. The use of two separate binding segments enables:
flexible primer design freedoms such as the balance of priming specificity and binding strength,
reduction in probability of forming amplifiable primer-primer dimmers in highly multiplexed reactions,
primer sections of captured probe sequences remain as short as possible so as to maximize reading length of native sequences.
