AgraStrip® Gluten G12 Test Kit includes:

Shelf life: 12 months
This test kit’s performance was reviewed by AOAC Research Institute and was found to perform to the manufacturer’s specifications. The following matrices have been validated according to AOAC Performance Tested Methods (PTM) protocols: rice flour, bread, cookies, ice cream and dark chocolate. Stainless steel has been validated per AOAC PTM protocol for environmental surface swab testing.

Transfer Dilution Buffer (red label) into the dilution tube already filled with extract to one of the lines demarcating the test cut off levels (5/10/20 ppm Gluten). Close the dilution tube with the cap and shake vigorously by hand for 15 seconds.
Transfer 3 drops (100 μl) of the sample supernatant into the dilution tube by squeezing the extraction tube.
After the liquid has soaked up to the “flow” level remove the test strip from the dilution tube and place it upright (arrows pointing down) into a slot of the Tube Holder and allow to develop for 10 minutes and then read off the result immediately.
Take off cap and place a test strip vertically (arrows pointing down) into the dilution tube and allow liquid to flow up the strip to the „flow“ level line. (this takes about 45 seconds).

Result Interpretation
Negative Result (N):
One single blue line in the control zone (c) of the strip
Positive Result (P):
One red line in the test zone (t) and one blue line in the control zone (c). Even a faint line in the test zone implies a positive result(see strip at bottom). The sample contains the specific allergen higher than the cutoff level (which needs to be validated for individual matrices).

Homogenise the sample and weigh 0.2 g of sam-ple and add to the ex-traction tube. (If balance is unavailable 0.2 g can be estimated by filling the extraction tube cap). Fill tube with extraction solution to level shown in picture.
Close tube using the ex-traction tube cap, and shake vigorously by hand for 1 minute. Remove extraction tube cap and replace with dropper tip.

Fill extraction tube with Extraction Buffer up to where the tube gets thicker, wet the swab by dipping into the buffer. Wipe an area of 5 cm x 5 cm using side to side movements, rotating the swab tip as you go.
Place the swab into the extraction tube and care-fully break off the end at the pre-scored point. Close the tube with a cap and shake vigorously for 1 minute. Remove cap and replace with dropper tip.
