    Storage Conditions: Store at room temperature.

    Quality Control: Ambions 7.5 M Lithium Chloride solution is tested in the following quality control assays

    before and after final packaging. The product is filtered through a 0.2 m filter into the final

    package and autoclaved.

    Non-Specific Endonuclease/Nickase Activity: None detected after a 14-16 hour incubation

    of 0.75 M LiCl with 300 ng of supercoiled plasmid DNA, analyzed on an agarose gel.

    Exonuclease Activity: None detected after a 14-16 hour incubation of 0.75 M LiCl with 40

    ng of32

    P-labeled Sau3A fragments of pUC19, analyzed by PAGE.

    RNase Activity: None detected after a 14-16 hour incubation of 0.75 M LiCl with 25 ng of32

    P-labeled RNA, analyzed by PAGE.

    USER INFORMATION:

    This solution is ready for use and requires no extra preparation. Though not necessary, this solution can be re-

    autoclaved. If precipitants form, warm solution to 37C to resuspend.

    LiCl precipitation is very useful following RNA isolation or in vitro transcription because RNA is efficiently precipitated,

    while free NTPs, RNA molecules smaller than 200 bases, protein, and DNA are very inefficiently precipitated or are not

    precipitated at all.

    To precipitate RNA in a solution with 400 ng/l or greater RNA concentration, bring the final concentration of LiCl in the

    RNA solution to 2.5 M, chill the solution at -20C for 30 minutes, and centrifuge at top speed in a microcentrifuge for 15

    minutes. Discard the supernatant and wash the pellet with ice-cold 70% EtOH to remove residual salt, then resuspend

    in nuclease-free H2O or TE.
