1.0 PENGENALAN
Pada umumnya cuka boleh dihasilkan melalui proses biologikal dan kimikal
Cuka yang dihasikan secara biologikal adalah produk yang dihasilkan setelah sesuatu bahan asas (seperti anggur) melalui dua peringkat proses fementasi.
Peringkat pertama fementasi menghasilkan minuman atau produk terfementasi yang berkaitan yang mengandungi alkohol.
Peringkat kedua fementasi adalah mengubah produk beralkohol tersebut kepada cuka
Manakala cuka yang dihasikan secara kimikal adalah produk yang dihasilkan melalui percampuran bahan kimia

• Mikroorganisma yang mengubah gula dalam jus anggur kepada alkohol, dan pada masa yang sama menghasilkan rasa yang dikehendaki, adalah yis (Saccharomyces ellipsoideus atau Sachharomyces cerevisiae; Saccharomyces dalam bahasa Latin –fungi gula ; “sugar fungus”)

2.3.1 Fermentasi Wain (wine)
1. Semasa anggur matang, yis wain semulajadi iaitu Saccharomyces ellipsoideus berhimpun pada kulit anggur
2. Apabila hancuran anggur atau jus yang telah ditapis (filter) ditambah bahan tambah sulfite seperti potassium metabisulfite untuk menghalang pembiakan bakteria asid asetik, wild yeast and kulat (mould)
3. Jus diletakkan pada suhu sekitar 27°C maka proses fermentasi akan bermula dan menghasilkan pada dasarnya kuantiti molar sekata etil alkohol (ethyl alcohol) dan karbon dioksida (CO2) dan sedikit/surih (trace) juzuk-juzuk perisa (flavour compounds)
4. Dalam operasi komersial, strain khas (special strains) bagi yis Saccharomyces ellipsoideus digunakan sebagai tambahan kepada inokulum semulajadi (natural inoculum) dan kawalan proses fermentasi lebih terkawal akan dilakukan
5. Sulfur dioksida (Sulfur dioxide - SO2)i. Yis wain agak berdaya tahan (relatively resistant) kepada
SO2
ii. Biasanya ditambah kepada anggur dalam bentuk garamnya iaitu potassium metabisulfite
iii. Membantu mengawal agar mikroorganisma yang tidak dikehendaki (terutamanya bakteria) dapat hidup
iv. Ianya berkesan dalam menghalang enzim pemerangan yang terdapat dalam anggur dari bertindak
v. Memberi keadaan tindakbalas pengurangan melalui tindakbalas dengan gas oksigen
vi. SO2 terawat mungkin difermentasi terus atau selepas pumace diasingkan
5. Fermentasi menyebabkan suhu naik maka proses penyejukan diperlukan untuk menghalang yis dari tidak aktif (inactivation)
6. Fermentasi dari pendedahan air yang terhad diperlukan sehingga gula digunakan sepenuhnya, atau berhenti secara semulajadi atau fermentasi terganggu sebelum titik penamat
7. Sekitar suhu 27°C, fermentasi akan tamat sekitar 4-10 hari bergantung kepada jenis wain.
8. Fermentasi sempurna boleh berlaku secara semulajadi atau dihentikan dengan menambahkan spirit tersuling
9. Strain yis yang baik boleh menghasilkan 14-18% alkohol
2.3.2 First “racking” Wain (wine)
1. Wain dibiarkan sehingga hampir keseluruhan sel-sel yis dan bahan halus terampai (fine suspended materials) mendap (settle out)
2. Wain akan dialirkan keluar (drawn off) tanpa mengganggu mendepan (sediment or ‘lees’), yang mengandungi antaranya potassium bitartrate (cream of tartar)
3. Jika lees tidak dipindahkan segera yis akan autolis (autolyze) dan menyumbang perisa terubah (off-flavour) kepada wain
2.3.3 Penuaan Wain (Wine ageing)
1. Wain akan dipenuaankan didalam tong kayu (casks) atau tangki (tanks) yang tidak masuk udara untuk jangkamasa beberapa bulan ke beberapa tahun dimana kandungan sirih (trace) terakhir gula difementasikan dan penghasilan terakhir perisa berlaku
2. Semasa peringkat ini tambahan ‘racking’ mungkin dilakukan
2.3.4 Penjernihan Wain (Wine clarification)
1. Wain akan dijernihkan (clarification) melalui proses penurasan (filtration) atau pengemparan (centrifugation)
2. Mengasingkan/memisahkan sirih terakhir bahan-bahan koloid (last trace of colloid materials)
3. Mengasingkan/memisahkan garam asid tartarik (tartaric acid) Tartarik (tartrates) ini berada dalam jus anggur, akan menghablur didalam tong wain dan jika tidak sempurna diasingkan dari wain sebelum dibotolkan,
ianya perlahan-lahan muncul kembali sebagai hablur seumpama kaca (glasslike crystals) didalam botol terakhir semasaa penyimpanan
Garam asid tartarik boleh dilakukan dengan menggunakan rawatan pertukaran ion (ion exchange treatment)
2.3.5 Penstabilan Wain (Wine stabilisation)
• Ianya proses melibatkan penyejukan untuk menggalakkan penghabluran untuk meningkatkan kecekapan pemisahan/pengasingan
2.3.6 Rawatan haba wain (wine heat treatment)
1. Jika wain < 17% perlu diberi rawatan haba
2. Heat-pasteurized atau cold-pasteurised melalui turas selaput liangmikro (microporous membrane filters) sebelum sahaja dibotolkan
3. Wain berkilau samada fementasi sekunder (secondary fermentation) dilakukan dalam botol atau di tong adalah tidak di pasteur walaupun kandungan alkoholnya tidak lebih dari 14% tetapi kadang-kadang SO2 ditambah sebelum botol ditutup untuk menahan mikroorganisma yang tidak dikehendaki dari membiak
2.4 Wain (wine)
Jenis wain berdasarkan kepada 1. Warna i. kulit jenis anggur adalah dari ungu pekat
(deep purple), melintasi merah hinggalah ke warna hijau pucat (pale green)
ii. Warna terakhir wain akan ditentukan berdasarkan kestabilan pigmen semasa tempoh penyimpanan yang semuanya bergantung kepada tahap jenis anggur
2. Kemanisan & Kandungan alkoholi. Kemanisan dan kandungan alkohol ada hubungan dengan
fementasi menukarkan gula dalam anggur kepada etanol (ethanol)
ii. Semakin banyak alkohol dihasil semakin mengurang kemanisan iii. Bila semua gula difementasi maka terhasillah wain tanpa
kemanisan disebut “dry” (12-14% alkohol)iv. “Dry” wain boleh jadi manis setelah yis dikeluarkan dengan
menambah sedikit jus atau gula yang tidak difementasi v. Wain manis tidak bermakna rendah kandungan alkohol kerana
alkohol boleh ditambah dalam bentuk spirit suling (distilled spirits) sehingga kandungan alkohol; 17-21% dan dipanggil “fortified wine” (wain diperkuat)
vi. Wain diperkuat kurang cepat rosak dan mungkin boleh stabil tanpa pempasteuran
vii. “light wine” menunjukkan kandungan alkohol dari ± 5% ke 10%viii. Fementasi semulajadi amnya menghasilkan alkohol kurang dari
16% mengikut isipadu walaupun gula ditambah lagi.16% adalah toksik kepada yis dan fementasi akan terhenti

2.5.1 Wain merah1. Hasil daripada hancuran kulit, pulpa dan biji jenis anggur ungu
atau merah dan dibiarkan dalam keadaan jus semasa proses fermentasi
2. Penghasilan alkohol menyumbang kepada warna sedutan (extraction) jus dimana semakin lama kulit, pulpa dan biji bersama sedutan jus semakin pekatlah warna yang akan dihasilkan
3. Wain merah mengandungi resveratrol lebih tinggi berbanding wain putih
4. Resveratrol mungkin boleh mengurangkan kolesterol darah, halang sakit jantung and anti barah (Murano, 2003)
2.5.2 Wain merah jambu (Pink/ rose` wines)
1. Boleh dihasilkan dengan mengasingkan bukan jus “pumace” daripada cecair atau “must” (Latin; young wine, is freshly pressed fruit juice that contains the skins, seeds and stems of the fruit) diawal proses fermentasi iaitu hanya sedikit sahaja kuantiti pewarna (pigment) diekstrak.
2. Boleh juga dihasilkan dengan mencampurkan (blending) wain putih dengan sedikit kuantiti wain merah
2.5.3 Wain putih
1. Boleh dihasilkan dari buah anggur yang berpigmen dengan mengasingkan kulit, palpa dan biji sebelum proses fermentasi dengan menggunakan dengan kaedah
Rawatan Ion exchange dan activated charcoal untuk mengeluarkan pigmen
Menggunakan enzim anthocyanase yang menyahkan warna pigmen
2. Boleh dihasilkan dari buah anggur jenis putih yang dihancurkan dan mengasingkan pepejal bukan jus sebelum proses fermentasi bermula
3.0 Cuka Wain/Wine vinegar
3.1 Cuka (Vinegar)
Wine and other (designated) alcoholic beverages (cider – e.g from apple) aerobically converted by Acetobacter into condiments rich in acetic acid and ethyl acetate for salad dressing and similar usage. Formerly now seldom, a spoiled wine. An alternative product for wineries.

• Secara umumnya kepekatan asid asetik membayangkan jumlah penghasilan etil asetat (juzuk utama bau cuka- main aroma compound vinegar)
• Biasanya asid asetik yang ditentukan kandungan kerana ianya lebih mudah dianalisa berbanding etil asetat
• Umumnya kekuatan bau wain dalam cuka
ditunjukkan dengan kepekatan asid asetik
3.2 Asas penghasilan cuka wain (wine vinegar)
Alkohol dari fementasi yis – wain dengan kehadiran oksigen, dan bakteria s.p Acetobacteria aceti akan meneruskan fementasi kepada penghasilan asid asetik

3.2.1 Cuka wain (wine vinegar)
1. Bakteria cuka dikhuatiri oleh pakar wain iaitu bakteria cuka memerlukan oksigen untuk hidup maka dimana dalam pemprosesan wain alat yang bersesuian diperlukan (well-equipped wineries)
2. Bakteria cuka terbahagi kepada
i. Bakteria asid asetik (acetic acid bacteria)
Contoh ;
a) Acetobacteria aceti digunakan dengan meluas lebih bertahan terhadap etanol (highly ethanol tolerant) mudah dikawal dengan menyelenggara keadaan tak berudara
(anaerobic) dan jumlah SO2 yang berpatutan (0.8 mg/L molekular
SO2)
b) Acetobacter pasteurianus
c) Acetobacteria peroxydans
ii. Bakteria asid laktik (lactic acid bacteria) –sesuai untuk beberapa
jenis wain tetapi boleh merosak jenis-jenis wain yang tertentu• Contoh ;
a) Leuconostoc oenos
b) Lactobacillus brevis
Asid malik bakteria asid laktik asid laktik + CO2
Asid malik adalah asid organik yang memiliki perisa epal (apple-like flavour)
Leuconostoc oenos digunakan untuk merendahkan pH produk dan digunakan tambah kepada wain merah dan wain putih
Lactobacillus brevis akan mengurangkan pH produk dengan hebat (severely reduce total acidity) menyebabkan wain tawar/tidak punya rasa (insipid) dan tidak bersemangat (lifeless)
iii. Pediococcus cerevisiae bakteria boleh menghasilkan histamine dalam beberapa wain terutama wain merah meja
Histamine adalah sebatian (compound) organik yang merangsang peredaran darah dan mungkin membawa peranan dalam masalah pernafasan hidung (nasal stuffiness), pening dan beberapa tindakbalas alahan pada peminum wain

3.4 Penghasilan cuka wain (wine vinegar)
1. Dalam penghasilan cuka (vinegar) terdapat dua langkah fementasi
2. Langkah pertama melibatkan pertukaran (conversion) gula dalam jus (contoh jus anggur, jus epal) kepada alkohol yang dimulakan dalam keadan berudara (aerobic) untuk merangsangkan pembiakan yis dan menambahkan sel mas (cell mass) tetapi ditukar segera kepada keadaan tidak berudara (anaerobic) untuk memudahkan (favour) fementasi gula kepada alkohol
3. Langkah kedua melibatkan pertukaran alkohol kepada asid asetik dan biasanya dilakukan dalam janakuasa cuka (vinegar generator)
3.4.1 Janakuasa cuka wain (Vinegar generator)
1. Janakuasa cuka terdiri dari tangki besar (tank) atau tong (vat) dihimpun dengan susunan kayu (packed with wood shavings) untuk mengujudkan keluasan permukaan berudara yang besar (to provide a large aerobic surface area)
2. Janakuasa ini memerlukan pengawalan teliti
3. Keadaan berudara (aerobic condition) membantu kulapuk (mould)
membiak dan meyebabkan asid asetik terurai seterusnya
4. Pengudaraan berlebihan boleh menyebabkan asid asetik teroksida
kepada CO2 dan H2O
5. Alkohol sider (cider) atau wine (wain) setelah diinokolum (inoculation) dengan baketria cuka, di titis/dilelehkan (trickled) melalui susunan kayu (wood shavings) sementara udara ditiub keatas melalui shaving (blown up through the shavings)
6. Cuka akan dikeluarkan dari janakuasa bila kepekatan asid asetik mencecah 4% (atau tinggi sedikit) – tahap minimum kebenaran undang-undang bagi asid asetik dalam cuka
7. Memanjangkan tempuh penuaan cuka akan secara keseluruhannya meningkatkan juzuk-juzuk lain seperti bahan antioksidan (fenolik), bahan teruap (3-furfural)
Raw material
1. Vinegar is made from a variety of diluted alcohol products, the most common being wine, beer, and rice. Balsamic vinegar is made from the Trebbiano and Lambrusco grapes of Italy’s Emilia-Romagna region. Some distilled vinegars are made from wood product such as beech.
2. Acetobacters are microscopic bacteria that live on oxygen bubble. Whereas the fermentation of grapes or hops to make wine or beer occurs in the absence of oxygen, the process of making vinegars relies on its presence. In the natural processes, the acetobacters are allowed to grow over time. In the vinegar factory, this process is induced by feeding acetozym nutrients into the tanks of alcohol.
3. Mother of vinegar is the gooey film that appears on the surface of the alcohol products as it is converted to vinegar. It is natural carbohydrate called cellulose. This film holds the highest concentration of acetobacters. It is skimmed off the top and added to subsequent batches of alcohol to speed the formation of vinegar. Acetozym nutrients are man-made mother of vinegar in powdered form.
4. Herbs and fruits are often used to flavour vinegar. Commonly used herbs include tarragon, garlic, and basil. Popular fruits include raspberries, cherries and lemons.
Design
• The design step of making vinegar is essentially a recipe. Depending on the type of vinegar to be bottled at the production plant – wine vinegar, cider vinegar, or distilled vinegar – food scientists in the kitchen and laboratories create recipes from the various vinegars. Specifications include the amount of mother of vinegar and/or acetozym nutrients added per gallon of alcohol product. For flavored vinegars, ingredients such as herbs and fruits are macerated in vinegar for varying periods to determine the best taste results.
The Manufacturing Process
I The Orleans method1. Wooden barrels are laid on their sides. Bungholes are drilled into
the top side and plugged with stoppers. Holes are also drilled into the ends of the barrels.
2. The alcohol is poured into the barrel via long-necked funnels inserted into the bungholes. Mother of vinegar is added at this point. The barrel is filled to a level just below the holes on the ends. Netting or screens are placed over the holes to prevent insects from getting into the barrels.
3. The filled barrels are allowed to sit for several months. The room temperature is kept approximately 85°F (29°C). Samples are taken periodically by inserting a spigot into the side holes and drawing liquid off. When the alcohol has converted to vinegar, it is drawn to the spigot. About 15% of the liquid is left in the barrel to blend with the next batch.
II. The submerged fermentation method
1. The submerged fermentation is commonly used in the production of wine vinegars. Production plants are filled with large stainless steel tanks called acetators. The acetators are fitted with centrifugal pumps in the bottom that pump air bubbles into the tank in much the same way that an aquarium pump does.
2. As the pump stirs the alcohol, acetozym nutrients are piped into the tank. The nutrients spur the growth of acetobacters on the oxygen bubbles. A heater in the tank keeps the temperature between 80 and 100°F (26 - 38°C).
3. Within a matter of hours, the alcohol product has been converted into vinegar. The vinegar is piped from the acetators to a plate-and-frame filtering machine. The stainless steel plates press the alcohol through paper filters to remove any sediment, usually about 3% of the total product. The sediment is flushed into a drain while the filtered vinegar moves to the dilution station.
III. The generator method1. Distilled and industrial vinegars are often produces
via the generator method. Tall oak vats are filled with vinegar-moistened beechwood shavings, charcoal, or grape pulp. The alcohol product is poured into the top of the vat and slowly drips down through the fillings.
2. Oxygen is allowed into the vats in two ways. One is through bungholes that have been punched into the sides of the vats. The second is through the perforated bottoms of the vats. An air compressor blows air through the holes.
3. When the alcohol product reaches the bottom of the vat, usually within a span of several days to several weeks, it has converted to vinegar. It is poured off from the bottom of the vat into storage tanks. The vinegar produced in this method has a very high acetic acid content, often as high as 14% and must be diluted with water to bring its acetic acid content to a range of 5 – 6%.
4. To produce distilled vinegar, the diluted liquid is poured into a boiler and brought to its boiling point. A vapor rises from the liquid and is collected in a condenser. It then cools and becomes liquid again. This liquid is then bottled as distilled vinegar.
The production of vinegar
Bascsamic vinegar1. The production of balsamic vinegar most closely
resembles the production of fine wine. In order to bear the name balsamic, the vinegar must be made from the juices of the Trebbiano and Lambrusco grapes. The juice is blended and boiled over a fire. It is then poured into barrels of oaks, chestnut, cherry, mulberry and ash.
2. The juice is allowed to age, ferment, and condense for five years. At the beginning of each year, the aging liquid is mixed with younger vinegars and placed in a series of smaller barrels. The finished product absorbs aroma from the oak and color from the chestnut.
3.5 Cuka semulajadi dan cuka buatan
i. "Natural vinegar " is acetic acid that is made in a biological process using the Acetobacter aceti bacteria.
ii. "Artificial vinegar" is acetic acid that is made by a chemical process by the action of methanol and carbon monoxide.
i. “Cuka semulajadi” pula adalah asid asetik yang diperbuat secara proses biologikal menggunakan bakteria Acetobacter aceti.

6.1 Produk makanan menggunakan cuka wain
6.2 Fungsi Cuka
1. Pengawet – memanjangkan tempoh hayat sesuatu makanan
2. Pengsekata asid (Acid regulator) – mengubahsuai pH makanan
3. Perisa (flavour) – meningkatkan nilai rasa makanan
4. Melambatkan ketengikan atau kerosakan lain disebabkan proses pengoksidaan disebabkan cuka mengandungi antioksida (polifenol/polyphenol)
5. Memperbaiki tekstur makanan seperti menghaluskan pes kanji (starch paste –sebagai pemekat) dalam salad dressing melalui proses asid hidrolisis (acid hydolysis) – (Potter and Hotchkiss, 1998)
6. Mengurangkan kandungan kolesterol kepada pengguna
6.2.1 Possible cholesterol and triacylglycerol effects
• Tikus yang diberi makan cuka atau asid asetik mampu menurunkan tekanan darah namun kesannya belum diuji pada manusia.
• Rats fed vinegar or acetic acid have lower blood pressure than controls, although the effect has not been tested in humans.
• "Dietary acetic acid reduces serum cholesterol and triacylglycerols in rats fed a cholesterol-rich diet“

Possible cholesterol and triacylglycerol effects
• Sebuah kajian 2006 menyimpulkan bahawa sekumpulan tikus yang diberi ujian dengan asid asetik (komponen utama dari cuka) mempunyai "nilai signifikan lebih rendah untuk total serum kolesterol dan “triacylgleycerol”
(Johnston and Gaas, 2006).
Possible cholesterol and triacylglycerol effects
• Mengurangkan risiko penyakit jantung iskemik fatal diamati dalam peserta dalam satu mahkamah yang makan cuka dan minyak salad dressing secara kerap.
• Reduced risk of fatal ischemic heart disease was observed among participants in a trial who ate vinegar and oil salad dressings frequently.
