EXECUTIVE SUMMARY

This Annual Environmental Report (AER) for Takeda Ireland Limited (Grange Castle)-TILGC

presents Environmental Management for the period of the 01st January to the 31st of December

2012.

The format of this report follows guidance issued by the Environmental Protection Agency and

presents information on environmental performance at TILGC.

In 2012 TILGC‟s main focus continued to be compliance with the licence and making

improvements on our existing environmental management system. There have been a number of

key achievements over this period which include:

Production of three API‟s SYR-322, TAK-375 and TAK-700;

Finalized the design and commencement of work on a new Wastewater Treatment Plant;

Installation of chemically resistant liners on 3 bunds.

Completion of a combined Environmental, Health and Safety Awareness Week by the

Environmental and Health and Safety Committees.

Introduction of Carbon Footprinting and Carpooling for the site.

Validation of environmental laboratory procedures.

Preparation for increased production levels and shift work in 2013.

TILGC is committed to continuing this high level of compliance and to continually improving on

environmental performance in the future. TILGC‟s good environmental record is testament to the

commitment of all employees in achieving environmental goals.

1. INTRODUCTION

Takeda Ireland Limited (Grange Castle)-TILGC was established in 2002. In that year the facility

was designed, an Environmental Impact Statement (EIS) was completed and planning permission

for the site was given. Construction on the site began in 2003 and in 2004 TILGC‟s Integrated

Pollution and Prevention Licence (IPPCL P0693-01) was granted. The commissioning and validation

of the plant was completed by the beginning of 2007. Commercial production began in April 2007

and to coincide with this the IPPCL was fully commenced.

TILGC is Takeda‟s first Active Pharmaceutical Ingredient (API) plant constructed outside Japan.

This is the fourth Annual Environmental Report (AER) for TILGC. It covers the 12 month period

from the 01st January until the 31st of December 2012.

The content of this AER is based on Schedule 5(i) Recording and Reporting to the Agency of the

IPPCL and follows guidelines set in the “Guidance Note for: Annual Environmental Report” issued

by the Agency.

2. SITE DESCRIPTION

2.1. Site Location and Layout

The site is located in Grange Castle Business Park, Clondalkin, Dublin 22. This is a new industrial

estate which was previously used as farmland. TILGC‟s facility was constructed on a green field

site. At 38 acres it holds five buildings, the Administration Building, the Main Production Building,

the Utilities Building, the Warehouse and the Drum Store.

2.2. Site Activities

TILGC is a multi-purpose API (Active Pharmaceutical Ingredient) Plant. It is primarily engaged in

the manufacture of pharmaceutical products from clinical trial stages through to launch and full

commercialisation. Due to the nature of the plant TILGC is continuously introducing new products

each year.

Over the period of this AER TILGC has produced three products which include TAK-700, TAK-375

and SYR-322. TAK-700 is a drug which is used to treat prostate cancer, TAK-375 is a drug used to

treat insomnia and SYR-322 is primarily used for the treatment of Type II diabetes.

2.3. Environmental Management Policy

Takeda is a global company which researches, develops, manufactures and markets a wide variety

of pharmaceuticals. Takeda strives towards better health for individuals and progress in medicine

by developing superior pharmaceutical products. TILGC regards environmental and energy

management as an integral and essential part of good business practice.

TILGC is committed to achieving and maintaining a high standard of environmental quality in all

our operations.

To achieve our goal and to set the standard, TILGC will:

At a minimum meet, and where appropriate exceed, the requirements of all applicable

legislation, corporate standards and industry standards to which TILGC subscribes.

Promote the concepts of continuous improvement and pollution prevention through the

setting and reviewing annually of environmental and energy efficiency objectives and

targets.

Provide all of the necessary information and resources to achieve objectives and targets.

Provide necessary training and support to employees so they can carry out their

environmental and energy management responsibilities effectively and professionally.

Implement a documentation system which will ensure that all documents required by the

IPPCL, environment and energy management system are generated and adequate records

are kept.

Keep up to date with all applicable legal requirements, industry standards and best

practices.

Make our environmental and energy management policy and records available to the

public and interested parties.

Promote the adoption of environmentally sound practices by contractors and suppliers.

Communicate our policy to all TILGC employees.

Ensure the Environmental and Energy Policy is reviewed regularly and updated as

necessary.

3. SUMMARY INFORMATION

3.1. Emissions to Atmosphere Summary

There are two main emission points specified in the IPPCL for TILGC – 1. EP-P1-02 (VOC Recovery

Unit) and 2. EP-P1-03 (Scrubber Hydrogenator). The Scrubber Hydrogenator has not been

operational since March 2010 and hence a summary of emissions has not been included for this

emission point for 2012. A summary of mass emissions to atmosphere from EP-P1-02, the VOC

Recovery Unit, for the period of 2010-2012 are presented in Table 3.1.1 and Figure 3.1.1. The

VOC recovery unit is used on a continuous basis when the plant is operating. The data presented is

based on monthly emissions monitoring of the parameters specified in the IPPCL, as analysed by

our external contract laboratory. All results were in compliance with the IPPCL.

3.2. Emissions to Sewer Summary

Emissions to sewer from TILGC are controlled under Schedule 2(i) of the IPPCL. Wastewater from

the laboratories, production building, boiler blow down sump, bunds and containment structures is

collected into our wastewater tanks. When the tanks are full they are pH adjusted and then the

wastewater is tested for pH and COD. If these parameters are within licence limits the wastewater

is then released to sewer. This is a batch operation with no continuous discharge to sewer.

A sample of wastewater discharged to sewer is taken and analysed on a monthly basis for the

parameters specified in Schedule 2(iii) of the IPPCL. These results are used with our continuous

monitoring results to calculate the mass emissions. The mass emission (kg) figure was determined

by multiplying the monthly data by the effluent flow mass emission for that month and adding the

sum for the year. A summary of our mass emissions is presented in table 3.2.1.

In addition to the parameters detailed in table 3.2.1, both pH and temperature were continuously

monitored on our DCS (Distributed Control System) during discharges. All emissions as detailed in

table 3.2.1 were 100% compliant with the IPPCL.

3.3. Surface Water Discharge Summary

Surface water from the car park, roofs and non-bunded areas are collected in the attenuation

ponds. This water is discharged to the river Griffeen. The discharge point (EP-WS-01) from the

attenuation pond is monitored in compliance with schedule 4(i) of the IPPCL.

A visual inspection of the ponds is carried out on a daily basis and the discharge point is monitored

continuously for pH and TOC during discharges.

In accordance with condition 9.1.2 of our IPPCL warning and action limits of 20 and 30ppm

respectively were set in relation to TOC. TILGC have also set internal limits for pH of 7-9.

Any value outside of the warning limits will cause an alarm on our DCS. Values outside the action

range will cause the discharge valve to shut automatically and the surface water will be retained in

the attenuation ponds.

One samples was taken by the agency during the period of this AER, the results from this analysis

are presented in Table 3.3.1. The results were within TILGC‟s warning and action limits.

3.4. Groundwater Monitoring Summary

Bi-annual monitoring of groundwater is required in accordance with Condition 9.3 of the IPPCL.

This monitoring was carried on two occasions during the period of this AER.

There are two groundwater monitoring points at TILGC, these are MP-WG-01 and MP-WG-02.

These monitoring locations are illustrated in figure 3.4.1.

Summaries of the monitoring results for the period of this AER and monitoring which was carried

out in 2010, 2011 and 2012 are detailed in tables 3.4.1 and 3.4.2. The results demonstrate that

the quality of groundwater under TILGC‟s facility has not been affected by activities on site.

3.5. Resource Consumption Summary

This section details figures for the total usage of energy and water at TILGC for 2010, 2011 and

2012. Production on site commenced in 2007; however levels of activity have been very low with

long periods of plant set up and cleaning. Due to this intermittent production and the multipurpose

nature of the plant there can be no meaningful comparison between resource consumption and

production figures. There is a two week shutdown in July of each year, as can be seen in figure

3.5.1 there is a noticeable dip in energy consumption during this period.

3.5.1 Energy

The two types of energy consumed at TILGC are electricity and gas. A breakdown of energy

consumption for 2012 is illustrated in figure 3.5.3.

TILGC utilises electricity from the mains supplied by ESBIE. The users of electricity on site include

ventilation and air conditioning, cooling, compressed air, vacuum pumps, motors and drives, trace

heating and lighting. Table 3.5.1 presents the electricity consumption at TILGC for 2010, 2011

and 2012. Figure 3.5.1 illustrates the electricity consumption for each month of 2010, 2011 and

2012. As illustrated in this figure, there was a small decrease in electricity consumption in 2012

relative to 2011. While this decrease might appear small we have considered it to be a significant

achievement considering production levels increased by approximately 75% in 2012. Again this

reduction is due to a number of energy saving initiatives which have been carried out on the site.

The source of this total electricity consumption data is from consumption reports provided by the

ESBIE.

TILGC utilises gas from the mains supplied by ESBIE. Natural gas is used to fuel the two main

boilers for the site (in a duty/stand by configuration) and two small gas burners in the admin

building which supply space and water heating for the whole site. Table 3.5.1 presents the gas

consumption at TILGC for 2010, 2011 and 2012. Figure 3.5.2 illustrates the gas consumption for

each month of 2010, 2011 and 2012. As illustrated in this figure, gas consumption is reduced over

the summer months; this is largely due to the reduced necessity for space heating. The source of

this total gas consumption data is from consumption reports provided by the ESBIE. There was

almost a 13% decrease in gas consumed on site since 2010. This reduction is mainly due to the

ramp down of most of the production building HVAC systems out of manned hours.

A programme for improvements in energy efficiency at TILGC has been included in the

Environmental Management Programme (Sections 4.1 and 4.2).

3.5.2 Water

TILGC‟s single source of water is South Dublin County Council Mains. Water is used in the

manufacturing process, utilities (generation of steam) and for cleaning equipment. Water is

purified before it is used in the process.

Table 3.5.2 details the water consumption at TILGC for 2010, 2011 and 2012. Figure 3.5.4 shows

the water consumption for each quarter of 2010, 2011 and 2012. The source of this total water

3.6. Waste Management Report

Waste generated at TILGC is classified into two categories, hazardous and non-hazardous.

The total quantities of hazardous waste generated on site in 2010, 2011 and 2012 are presented

in table 3.6.1. Table 3.6.2 presents all non-hazardous waste generated during the same time

period.

Figure 3.6.1 illustrates the composition of waste for 2012.

3.6.1 Hazardous Waste

During 2012, a total of approximately 1,300 Tonnes of hazardous waste was generated onsite. The

majority of this was aqueous solvent, which arose from production processes. Other hazardous

waste streams generated on site included solid hazardous waste, waste oils, spent fluorescent

tubes, batteries and waste electrical equipment.

In accordance with condition 7.7.4 of the IPPCL, documentation confirming acceptance and

disposal/recovery of all hazardous waste streams transferred from TILGC, are retained on-site for

inspection.

No waste was consigned from the site for recovery abroad as a „Green List‟ waste. There were no

rejected waste consignments during 2012.

The total quantities of hazardous waste generated on site in 2010, 2011 and 2012 are presented

in Table 3.6.1.

As shown in this table the volume of hazardous waste generated in 2012 has decreased from 2010

figures, and is primarily dependant on production activities.

3.6.2 Non-Hazardous Waste

During 2012, approximately 80 Tonnes of non-hazardous waste was generated onsite. Bund

washings resulting from the project to install chemically resistant liners in 3 bunds accounted for

the majority, approx 50%, of non-hazardous waste generated on site.

The remainder of the non-hazardous waste was generated from general waste and cleanout of

attenuation ponds. The total quantities of non-hazardous generated on site in 2010, 2011 and

2012 is presented in table 3.6.2.

3.7. Complaints Summary

In accordance with condition 12.3 of the IPPCL, TILGC is required to record and notify the Agency

of any environmental complaints received on a monthly basis. There were no complaints received

by TILGC in 2012.

3.8. Environmental Incidents

There were three environmental incidents reported to the Agency during the period of this AER.

These incidents are detailed in table 3.8.1. All incidents were reported in compliance with condition

4.1 of our IPPCL.

4. MANAGEMENT OF THE ACTIVITY

4.1. Environmental Management Programme Report

The following is a description of the Environmental Management Programme (EMP) completed

throughout 2012. Sections 4.1 and 4.2 present the EMP report for 2012 and the EMP Proposal for

2013, respectively.

The success and achievements associated with each of the proposed environmental objectives are

detailed under each of the applicable sections below.

4.1.1. Energy Usage

The 2012 EMP target in relation to this objective was to further progress the implementation of the

EN 16001 Energy Standard (now ISO 50001). This involved the following key actions;

Gain accreditation to ISO 50001

Implement phase 2 of the automatic monitoring system

Projects to improve efficiency of compressed air system

Provision of Training to engineering technicians on improvements to compressed air

system

Provision of awareness training as part of Environmental Week

Conduct Carbon Footprint Analysis of TILGC

A considerable amount of progress was made in relation to gaining ISO 50001 accreditation, a

mock audit was conducted in November 2012 which resulted in only minor observations. A full

accreditation audit has been scheduled for Q1 2013. Full accreditation was delayed due to

unrelated issues in the plant.

Phase 2 of the automatic monitoring system was implemented, this involved integration

of existing boiler gas and steam meters into our energy management software.

A review of the compressed air system was completed which identified isolation of the

dryer to be a potential energy saver. This was completed and an energy saving of

80,000 kwh was identified. Additional projects on the compressed air system were

identified for implementation in 2013.

Other projects implemented in 2012 under the energy management system included

optimisation of boiler controls, reduced HVAC operational hours and improved heat

tracing controls on external pipe work. Completion of these projects resulted in significant

energy savings which comprised of 6.2% electricity and 7.46% gas consumption on 2011 usage

levels. This equated to a total saving of 1,502,142 kWh. This saving is considered to be a

significant achievement taking increased production levels, of approximately 75%, into account.

Training was provided to all relevant engineering technicians in the form of tool box talks.

An energy awareness programme was incorporated into the environmental, health and safety

awareness week which was held in November 2013. Carbon Footprinting and Carpooling for the

site was also launched during this week.

A summary of this objective is presented in table 4.1.1.

4.1.2. Waste

This objective aimed to reduce the volume of bulk liquid waste leaving the site for disposal. This

involved the following key actions;

Finalize the design of the Wastewater Treatment Plant and

Commence Construction

The WWTP design was finalized in October 2012 with the completion of a HAZOP. Construction of

the WWTP also commenced in October and is scheduled for completion at the end of Q1 2013. As

there will be no change to the discharge point or discharge parameters construction of the WWTP

is being accommodated under condition 1.2 of the IPPCL subject to conditions outlined by the

Agency.

A summary of this is presented in table 4.1.2.

4.1.3. Emissions to Atmosphere

The objective in relation to air emissions for 2012 aimed at compliance with F-Gas regulations.

This involved the following key action;

Review Maintenance work instructions/records to ensure full compliance with the F-Gas Regulations.

A review of contractors carrying out maintenance work on chiller systems was conducted. This

involved a review of method statements, work orders and certification of personnel conducting the

work. Copies of all relevant certification have been retained.

A summary of this is presented in Table 4.1.3.

4.1.4. Water

This objective aimed to increase the chemical resistance and integrity of bunds on site by the

following key action:

Install chemically resistant liners in 3 bunds (Solvent, HCl and NaOH).

This was completed in August 2012. In order to do this scaffolding was erected over the entire

tank farm to make the area watertight. Specialist silicate based mortar was then applied to the

bund floors and walls and allowed to cure. The bunds were tested afterwards in accordance with

BS8007 and passed.

4.1.5. In House Analysis

This objective aimed to validate in house analysis in accordance with EPA Guidance. This involved

the following key actions;

4.1.6. Environmental Awareness

This objective aimed to increase Environmental Awareness on site through the following key

A joint environmental health and safety week was held on the 05th-09th November 2012. The

theme of the week was “shared responsibility” and the aim was to make employees aware that we

are all responsible for protecting the environment and for the health & safety of ourselves as well

as others, not only at work but also in the home.

The event consisted of two environmental days. The first day focused on general environmental

awareness, a booklet on environmental systems and how Takeda protects the environmental was

produced. The booklet was simple and aimed at children so that employees could share the

information with their families. In conjunction with this an environment themed colouring

competition was held. The second day focused on Energy Conservation, Carbon Footprinting and

carpooling for the site were both launched on this day.

Events each day included a presentation on the relevant topic (presented on powerpoint, at a stall

and on TV in the canteen), distribution of Did You Information relating to the topic and a spot prize

question relating to the topic. In addition, an EHS Quiz was held on the final day, the questions

were taken from the material presented to employees during the week.

4.1.7. List I and II Substance Reduction and Elimination of Carcinogens in use

This objective aimed to manage and control List I and II Substances and Carcinogens through

Chemical Management System. This involved the following key actions;

Retrospective evaluation of all chemicals on site.

A Hazardous Material Inventory Audit was completed in December 2012. This audit consisted of a

physical inspection of all storage areas including laboratory, workshop, drum store and warehouse

areas followed by desk top review of all documents including approval and inventory management

forms and associated databases. This resulted in an audit report detailing all actions carried out to

correct findings including updated databases for new SDSs, assigned list I and II substance and

carcinogenic classifications as well as F Gas status.

To identify our significant environmental aspects TILGC‟s register of environmental aspects and

impacts for the site was reviewed. This register is reviewed on an annual basis with a view to

including the most significant into the EMP. The aspects which were found to be significant have

been addressed in the EMP proposal for 2013.

Table 4.2.1 presents the Environmental Management Programme Proposal for 2013. It defines the

responsibilities, resources and timetables for delivery of TILGC‟s environmental objectives.

5. LICENCE SPECIFIC REPORTS

5.1. Bund Integrity Testing

In compliance with condition 5.2.1 of the IPPCL TILGC submitted a bund integrity assessment

report to the Agency on the 18th of July 2007.

This assessment report identified the bunds present on site, the design philosophy behind the

sizing and construction of these bunds and demonstrated how they were compliant with the EPA‟s

guidance note on “Storage and Transfer of Materials for Scheduled Activities”.

Details of all integrity tests previously carried out on the bunds and a schedule (table 5.1.1) for

future testing was also included in the report.

In compliance with the schedule presented in table 5.1.1, a bund integrity test was conducted on

the Solvent Tanks, HCL Tank, Caustic Tank and HTF Tank in 2012. These tests were conducted in

accordance with reference to BS 8007:1987 “Design of concrete structures for retaining aqueous

liquids” and deemed to have passed in accordance with the criteria set out in this standard.

During 2011, a structural assessment was conducted by external contractors on the Solvent Tank,

the HCl Tank and the Caustic Tank. This report concluded that chemically resistant liner‟s are

required in all 3 bunds. This work will be performed in August 2012. All bunds tests were

conducted after this work was complete.

5.2. Tank and Pipeline Testing and Inspection

In accordance with condition 9.4.5 of the IPPCL, TILGC is required to undertake a programme of

testing and inspection of active underground pipes at least every three years.

In 2009 a programme for the testing and inspection of underground tanks and pipes was

established. In accordance with this schedule testing and inspection of underground tanks and pipe

was conducted in November 2012.

CCTV surveying of the surface, foul and process drains was carried out as well as pressure testing

of the process drains. Some minor maintenance issues were identified which do not have a

negative impact on soil or groundwater protection; remedial work will be addressed and completed

as part of the maintenance programme for 2013. Repeat testing will be carried out on all repaired

drains. All test and survey reports are maintained on file for inspection. A programme for the

testing and inspection of underground tanks and pipe is due again in 2015.

5.3. Noise Monitoring

In accordance with condition 8.1 of the IPPCL a noise monitoring survey was carried out on the

27th of October 2012. This survey was carried out by an independent consultant and was

conducted in accordance with the Agency‟s Environmental Noise Survey Document. The results

obtained are presented in Table 5.3.1.

an average of 8HGV‟s and 17 cars passing during each monitoring period

Note 3: The main source of noise at these locations was identified to be a radio which had been left on overnight in

the adjacent scaffolding yard.

The report indicates there were no audible noises from the site at most of the noise locations.

Almost all of the sources of noise were due to external construction, passing traffic and aircraft.

In summary, both traffic and the contribution from a neighbouring facility play an important role in

determining the noise climate in the area around the facility and therefore it is not possible to

identify the single contribution of Takeda Ireland Limited to noise levels. The noise report

concluded that the noise activities at Takeda Ireland Ltd. can be considered compliant with the

licence requirements.

5.4. Residuals Management Plan/Environmental Liabilities Risk Assessment

IPPCL Condition 14.2.1 requires that a fully detailed costed plan for the decommissioning or

closure of the site shall be prepared. Accordingly TILGC submitted a closure plan to the Agency on

the 23rd Oct 2007 based on the EPA‟s „Guidance on Environmental Liabilities, Risk Assessment,

Residual Management Plans and Financial Provision (2006)‟. The total cost of the plan is estimated

at €225,000, with Takeda Ireland Limited (TILGC) acting as company guarantor. The addition of

an administration building, including workshop, may result in some additional cleaning, waste

disposal and maintenance costs however this will not be significant and hence amendments to the

plan are not proposed at this point.

TILGC is a category 2 facility and as such is required to complete an Environmental Liabilities Risk

Assessment (ELRA). This was conducted in 2010 and an updated version was conducted by an

independent consultant in 2011. There have been no significant changes to the plant since the

completion of this report.
