Massoud University of Louisiana at Lafayette, USA DNA Cleavage by Efficient Artificial Nucleases and Role of Steric Effect Understanding the factors that might affect the DNA cleavage is considered to be the key role in synthesizing “efficient artificial nucleases” for DNA cleavage. A number of cobalt(II) complexes based-tripod pyridyl tetraamines were synthesized and structurally characterized. The complexes, which provided different steric environment around the central metal ion, were used to study the DNA cleavage under physiological condition. All the complexes, [Co(L)Cl]ClO /PF cleaved the DNA at micromolar concentrations within 2 h at 37 °C and pH 7.0 4 6 2+ under hydrolytic conditions where the reactive species is [Co(L)(H O)] ion. 2 Compared to the un-catalyzed cleavage of DNA, rate enhancements up to 100-170 million were achieved in some cases. The reactivity of the complexes in promoting DNA cleavage, their associated rate enhancements over the un-catalyzed DNA and the mechanistic pathway of the cleavage process as well as the catalytic efficiencies of the 1,2 complexes will be addressed. 1.
