REBATEN – natural high intense sweeteningWith a forthcoming European approval of stevia, Seppic is ready with an inter-esting range of this natural high intense sweetener, which is well suited for e.g. food supplements, drinks and pharmaceuticals

By Thierry Liot, Stevia Development Manager, Seppic

In September 2009, the French gov-ernment approved stevia extracts with 97 % purity Rebaudioside A (Reb A) as food and beverage sweet-eners by inter-ministerial decree, tak-ing advantage of a two year window that allows approval ahead of the rest of the EU. This has given the French company Seppic a great opportunity to achieve useful experience with the use of this natural high intense sweetener.

Legal status in EUStevia is not approved in EU yet, but just a few months ago the expert panel of the EFSA (European Food Safety Association) expressed a posi-tive recommendation on the use of steviol glycosides in foods and beve-rages, emphasizing quality standards as expressed by JECFA (Joint FAO/WHO Experts Committee on Food Additives). This document is paving the way to a European approval, ex-pected by end 2010 or early 2011.

The REBATEN rangeBased on the recommendations ex-pressed in this document, it is SEPPIC’s intention to propose 4 JECFA-com-pliant grades for industrial appli-

cations (food, beverages and phar-maceuticals) and one specific grade for a ready-to-use powder table top sweetener – see table below. With these 5 grades, Seppic covers the en-tire panel of applications that Euro-pean producers are looking for (taste profile, corresponding to application, manufacturing constraints, cost/sweetness ratio, …).

REBATEN 97% in food supplements and drinksDue to its great stability and solubil-ity, REBATEN 97%, Stevia extract, can easily be used in low-calorie or no sugar added food supplements. The maximum usage dosage (expressed in Rebaudioside A) authorised by French regulation is as follows:

• Liquid food supplements - 600 mg/kg

• Solid food supplements - 2000 mg/kg

• Vitamin- and/or mineral-based food supplements in syrup or chewable form - 5500 mg/kg

Likewise because of its great stabi-lity and solubility, REBATEN 97% Ste-via extract can be used as a natural sweetener in a wide range of drinks like slimming drinks, energy drinks,

vitamin-enriched drinks, etc. This also makes it ideal for formulating low calorie or no added sugar drinks.

REBATEN 97% advantages• Improved flavour • No sugar, fewer calories! Reduced

calorie content • Low glycaemic index • 100% natural origin (Stevia plant

extract) • Ideal for formulating sugar-free

food supplements

Industrial benefits• Intense sweetness• High stability in terms of pH, light

Health benefits of AroliaAnthocyanins and proanthocyanidins are Arolia’s active principles. They be-long to a specific group of polyphe-nols called flavonoids and have a high antioxidant activity. ORAC (Oxygen Radical Absorbance Capacity) assay is the most commonly used method to evaluate the antioxidant activity of a product. This assay was performed to compare the antioxidant capacities of Arolia, its raw material (acai pulp) and several fruit pulps whose ORAC values are indicated in the Antioxi-dant U.S. Department of Agriculture database published in 2007.

Acai pulp has the highest ORAC value among other black/purple pig-mented fruits. Moreover, Arolia anti-oxidant capacity is respectively 2 and 27 times higher compared to freeze dried acai and acai pulp, respectively. These data show the importance of the extraction process designed to concentrate acai specific components responsible of the high antioxidant effects in Arolia.

In conclusion, one gram of Arolia is equivalent to 2,700 ORAC units and covers 65% of the recommended daily antioxidant intake.

Arolia – exclusive concentrated Acai extractOne gram of Arolia, which is equivalent to 2,700 ORAC units, covers 65% of the recommended daily antioxidant intake. Arolia can therefore easily complete the other antioxidant sources to reach the recommended level

Arolia is a natural exclusive extract of acai berries containing 2% proan-thocyanidins. The extraction process, developed by BIOPHYTIS, is specifi-cally designed to preserve and con-centrate the most interesting com-ponents of the acai berry. Arolia’s antioxidant capacity is triggered and its content in lipids is reduced in com-parison with acai berry and freeze-dried acai pulp.

Traceability and sustainability is to-tally implemented from the plant to the extract. Plants are strictly selected and the acai is harvested and pro-duced in Brazil. BIOPHYTIS extraction plant is located in Atibaïa, Brazil.

Being an alimentary extract of the ed-ible part of a food plant, Arolia has been approved as a food ingredient by French regulatory authorities, and can therefore be used for dietary sup-plements and functional foods.

About acaiAcai is the fruit of the palm tree acaizeiro which is typical from Ama-zonian lands. Acai berries have been traditionally consumed by native Americans from this region and have been recognised as healthy food due to their number of active principles with health benefits: fibres, mono- and poly-unsaturated fatty acids, phytosterols, phenolic compounds, vitamins and minerals.

Quality Commitment To ensure the confidence and satis-faction of its customers in accordance with regulatory requirements, SEPPIC has a sustainable progress policy fo-cused on the control of risks and a Quality Hygiene Safety and Environ-mental management system.

As a partner of the pharmaceutical, nutrition, and cosmetics industry, SEPPIC integrates and implements all relevant reference systems in its man-agement system.

About steviaBetter known simply as stevia, Stevia rebaudiana is a wild herb which originates from the sub-tropical forest located on the border of Paraguay and Brazil. However, today, stevia is also cultivated in East Asia, including China, Korea, Taiwan, Thailand, and Malaysia.

Steviol glycosides are what give Stevia leaves their sweet flavour. These compounds are 40 to 300 times sweeter than sucrose (table sugar). Among all steviol glycosides contained in the Stevia leaves, the following can be mentioned:

• Stevioside represents 5-10% of the dry matter (250 – 300 times sweeter than sugar). This substance has a bitter, mentholated taste.

• Rebaudioside A represents 2- 4% of the dry matter. This is the sweetest substance (200 to 400 times sweeter than sugar), and it has no bitter taste.

• Rebaudioside C represents 1- 2% of the dry matter.• Dulcoside A represents 0.5-1% of the dry matter.

4 effective trendy extracts from Afriplex Afriplex offers a wide range of plants extracts made from plants indigenous to the African continent. Among these are Buchu, Pelargonium sidoides and Irv-ingia, which - together with Rose hip - are of great interest at the moment for health supplements due to recently published studies

By Annette Strarup, Area Sales Ma-nager, Alsiano

Pelargonium sidoides for immune health/coldsThe Pelargonium is successfully em-ployed in the modern phyto-therapy in Europe to cure infection diseases in the respiratory tract. Until recently, a German company have had mo-nopoly on Pelargonium supple-ments, but their patent expired last year and Pelargonium extract is thus now available for use for everybody.

Pelargonium is a South African indigenous plant and has been used for centuries by the Zulu as a traditional remedy in the treat-ment of upper respiratory tract ir-ritations such as cough, fever and bronchitis. In the 19th century, the South African drug ‘Umckal abo’ became known in Europe where the pink-flowered Pelargo-nium renifine and the very similar but dark maroon-red flowers of Pel-argonium sidoides are ingredients in a German medicinal remedy called ‘Umckaloabo’.

Modern human studies comprise two relatively new ones: a study on com-mon colds from 20071) and a study from 20032) on the efficacy on acute bronchitis.

Besides extract in powder form, Pel-argonium sidoides from Afriplex is also available as a liquid solution. Ap-plications: tablets, capsules, mixtures.

Irvingia extract for weight loss for the US and Asian marketIrvingia extract is now available from Afriplex, due to high demand from

emptying and allow for a more gra-dual absorption of dietary sugar. Ini-tial studies have suggested that the high fibre content of Irvingia account-ed for its beneficial metabolic effects, but recent animal studies suggest other factors as well. Researchers are studying Irvingia for its potential use in treating diabetes, metabolic syn-

drome and obesity.

In a study of 40 obese Cameroonian patients done in 2005, signifi-cant decreases of body weight, total cholesterol, LDL or ‘bad cholesterol’ and triglycerides were ob-served by the end of the 10 weeks’ study. A 2008 study based on 102 obese patients3) indicated similar results with lower dosages of Irvingia extract. 150 mg seed extract twice a day for 10 weeks reduces

weight by 13 kg compared to 0.7 kg for placebo. More evidence is needed about long term safety and efficacy, but already now Irvingia is a popular natural ingredient for weight loss es-pecially in the US.

for treatment of diseases of the respi-ratory tract, thanks to its expectorant, bechic and antispasmodic properties.

JostEuropean regulations are getting more and more precise and restrictive regarding the levels of contaminants in food and nutritional products. As far as heavy metals are concerned,

there are a number of existing regula-tions that concern mineral salts.

Jost Chemical has been producing guaranteed high-purity minerals for nearly 25 years and is now also well-known for having developed a new technology for the manufacturing of ultra-pure calcium salts for infant for-mulations and drink applications.

ageing. During World War II rose hip syrup was an important source of vi-tamin C for children.

Over time, several studies have been made. A human study of a rose hip remedy called LitoZin4) showed pos-sible benefits to rheumatoid arthritis sufferers. Seventy-four people, most-ly females, took part in the six-month trial. Just under half took the rose hip remedy LitoZin while the others took a placebo. Both groups conti-nued to take their usual medication. According to results, activity among the group taking the rose hip remedy improved by 20-25%. The number of joints causing pain or discomfort fell by 40%, but did not change for those treated with placebo.

Rose hip from Afriplex is available in powder form and as essential oil. Applications: tablets, capsules, bever-ages.

No scientific evidence is available to justify Buchu´s herbal uses, but the diuretic and anti-inflammatoric ef-fect may be attributed to the vola-tile oil and flavonoid’s irritant nature. Despite the lack of evidence, buchu is still used today in western herbal medicine for urinary tract ailments. It is also used in combination with other herbs such as cornsilk, juniper, cranberry and uva-ursi.

Buchu from Afriplex is available as water extract spray-dried to malto-dextrin, essential oil and ethanolic ex-tract. Applications: tablets, capsules, cosmetics.

Rose hips for joint health applicationsRose hips are the fruit of the rose spe-cies Rosa canina. The fruit is ready to be harvested when it changes colour from green to bright red during early winter in South Africa.

The fruit contains mono- and digly-cerol (also known as G.O.P.O.) exhibit-ing anti-inflammatory properties and is therefore often used in joint health applications. Rose hip also contains an excellent source of vitamin C and antioxidant bioflavonoids. The carto-noids present in rose hips are beta-carotene, lycopene, betacryptoxan-thin, zeaxanthin, and lutein.

The benefits of rose hips have been known for centuries. Historians say the Vikings used rose hips to stave off scurvy on their long sea voyages and in the past, rose hips have been used across the world to treat a number of conditions, from arthritis, colds and flu to bladder stones and premature

Buchu for urinary tract infectionBuchu (Agathosma betulina) is a low growing shrub native to Cape Town, South Africa. It is part of the cultural heritage of the San and Khoi people as a stomach tonic and to treat uri-nary tract infections. Today, Buchu is widely used as a household medicine in South Africa, with a great repu-tation for treating kidney and uri-nary tract diseases. Buchu was first exported to Britain in 1790, and in 1821 it was listed in the British phar-macopoeia as a medicine for ‘cystitis, urethritis, nephritis and catarrh of the bladder’. Buchu is also listed in German Commission E Monographs and has moreover been consumed as herbal tea in Germany for decades.

The main compounds in the essential oils of Buchu are isomenthone and diosphenol (Buchu camphor). The fla-vour oil of Buchu has a characteristic blackcurrant smell and flavour. The essential oils are generally used be-cause of their antiseptic and diuretic properties. Buchu leaves also contain the flavonoids rutin, diosmin, hespe-ridin, and quecetin. The dry leaves are used in herbal preparations and contain around 3% essential oil.

Afriplex – the essence of AfricaAfriplex is a strong player in the international plant extract mar-ket, specialising in extracting the essence of Africa. The company specialises in providing high quality plant extracts for beverage, phar-maceutical, cosmetic and other applications. The extracts are pro-duced in client specific formats, thereby ensuring satisfactory prac-tical application to the client.

New innovative ingredient for ODTPEARLITOL® Flash is a new innovative compound excipient from Roquette for successful development of fast disintegration formulations such as orodisper-sible tablets

Directly compressibleThanks to its excellent compactibility, PEARLITOL® Flash makes possible the manufacture of good mechanically resistant tablets by direct compres-sion.

Under studied conditions, Korsch XP1 press fitted with 13 mm diame-ter flat bevelled-edge punches, 15kN compression force was sufficient to obtain 500 mg orodispersible tablets with acceptable hardness, friability and disintegration time characteris-tics.

Regulatory statusThe use of PEARLITOL® Flash in fast disintegrating tablets or orodispers-ible formulation tablets is suitable for both pharmaceutical and nutraceuti-cal applications. PEARLITOL® Flash is a compound of mannitol and extra white maize starch. Both ingredients comply with pharmaceutical com-pendial standards and food regula-tions. A US DMF type IV (excipient) is registered at the FDA.

PEARLITOL® Flash – ideal for ODT To sum up, the requirements for the ideal excipient in ODTs are good compatibility, fast disintegration and a nice mouth-feel (sweetener). PEARLITOL® Flash is an excipient that possesses all these abilities.

By Anders Hager, Sales Manager Healthcare, Alsiano, & Elham Blouet, Pharmaceutical Project and Develop-ment Manager, Roquette.

What kind of technology can achieve a fast delivery of active ingredients inside the mouth in 2 ml of water? The orodispersible dosage form has the characteristic feature of requiring neither water nor chewing during its administration - orodispersible tab-lets (ODT) disintegrates in the pres-ence of saliva.

ODT are uncoated tablets intended to be placed in the mouth where they disperse rapidly before being swallowed. Besides the fact that the ODT dosage form is one among oth-er means of extending the product lifecycle, this modern medication de-livery system offers various benefits:

• Easy to swallow – e.g. geriatric and paediatric populations

• On-the-go use – does not require drinking water as with a conven-tional tablet

• Better patient compliance - e.g. at-tractive mouth-feel

PEARLITOL® Flash: An innovative compound excipientBased on their wide experience with mannitol (PEARLITOL®), Roquette has developed an innovative binary com-pound excipient, PEARLITOL® Flash, which is composed of mannitol and maize starch.

PEARLITOL® Flash offers a technol-ogy that enables successful develop-ment of fast disintegration formula-tions such as ODT by providing:

• A uniquely pleasing taste• Fast melting in the mouth• Smooth texture• An easy DC tabletting process• A robust DC excipient• A surprisingly simple formulation

PEARLITOL® Flash provides a uniquely pleasing taste experience. The oro-dispersible tablets melt in the mouth with a very creamy and smooth tex-ture.

Contrary to usual orodispersible for-mulation tablets, no superdisinte-grant is needed, since PEARLITOL® Flash mannitol compound exhibits self-disintegration properties. In ad-dition, a low lubricant level (0.4%) is sufficient to achieve good compres-sion performance. This low level is beneficial to maintain the pleasant taste of PEARLITOL® Flash, whereas a high level of lubricant could compro-mise it.

Fast melting in the mouthOrodispersible tablets formulated with PEARLITOL® Flash, 0.4% of lu-bricant (magnesium stearate) and active substances from 2% up to 30%, display a very quick disintegra-tion time (Eur.Ph. method): 50-80 sec onds. This disintegration time is far below the “within 3 minutes” disin-tegration time required by the Eur.Ph. for orodispersible tablets.
