1. INTRODUCTION
Respiratory diseases are a common cause of illness in calves in all
countries with cattle industry. They are the principal cause of economic loss
both in dairy and beef industries (Smith, 2000; Van der Fels–Klerx et al.,
2001). Foreign and Lithuanian researches show that bronchopneumonia of
calves, otherwise enzootic pneumonia, occurs for a number of reasons why
this disease generally known as the bovine respiratory disease (BRD)
complex (Hagglund, 2005).
Studying the bovine respiratory diseases in calves researchers have
found out that parainfluenza-3 (PI-3) and respiratory syncytial (RS) viruses
and, somewhat more seldom, infectious bovine rhinothracheitis (IBR)
viruses, adenoviruses, coronaviruses and Mycoplasma spp. are the main
agents of the BRD complex (Storz et al., 2000; Šiugždaitė, 2002; Snowder
et al., 2006; Autio et al., 2007). Commonly viruses are found simultaneously
with bacteria, such as Manheimia hemolytica, Pasteurella multocida,
Haemophilus somnus, Salmonella dublin and Arcanobacterium pyogenes.
The BRD complex includes a few syndromes of different epidemiology.
This complex is characterised by different clinical symptoms and
epidemiological features (Radostits et al., 2000).
Respiratory diseases may occur in cattle of different age yet is most
common in young calves when the passive immunity is reduced and the
active immunity is not yet developed (Radostits et al., 2000). The BRD
complex is distinguished for its seasonal character: the respiratory diseases
usually occur in cold seasons. However, the diseases also may occur in the
summer, especially under the humid and cool weather conditions
(Mockeliūnas et al., 2005; Hagglund, 2005).
In the assessment of individual agents of BRD complex, parainfluenza-3
and respiratory syncytial viruses should be distinguished for their important
aetiological role (Elvander, 1996; Larsen, 2000; Autio et al., 2007).
The bovine parainfluenza-3 agents are single stranded RNA viruses from
Paramixoviridae family Respirovirus genus. The bovine PI-3 virus is
genetically comparable but not identical with human and sheep PI-3 viruses
(Henrickson, 2003). The circulating PI-3 viruses in a herd always pose
threat of the outbreak of respiratory diseases as viruses create favourable
conditions for the secondary bacterial infections; thus cattle may get
infected with mycoplasma and other pathogens. The presence of this agent
in a herd may not manifest in clinical signs until favourable ambient
conditions and other pathogens occur. Commonly animals infected with PI-
3 experience mild clinical signs; cases of animal death are rather rare. Often
PI-3 virus become active along with infectious bovine rhinotracheitis,

bovine viral diarrhoea and RS viruses, Pasteurella multocida and
Haemophilus somnus (Cusack et al., 2003; Autio et al., 2007; Calderon et al.,
2007; Griffin et al., 2010).
Bovine respiratory syncytial virus (RS) is a single stranded RNA virus
from the Paramixoviridae family Pneumovirus genus. The RS virus is
widespread over the world among cattle, sheep, goats and other animals
(Eatson et al., 2004). The same as PI-3 agent RS viruses prime the
respiratory diseases and is usually found together with other pathogens
(O’Neill et al., 2006; Valarcher et al., 2006). The disease commonly
manifests in calves before six months of age yet its outbreaks are possible in
herds of adult animals as well (Patel, Didlick, 2004).
The bovine respiratory syncytial and parainfluenza-3 viruses are
especially relevant for the Lithuanian cattle herds as they can induce the
disease outbreaks both in calves and adult animals (Šalomskas et al., 2008).
The diseases caused by the mentioned agents and the inflicted damage to the
Lithuanian farms have not been investigated and in the last ten years there
has appeared not a single publication on this issue. This is a weighty reason
to analyse and go deeper into the extent of the incidence of these viral
infections in the Lithuanian cattle farms and to identify the agents using
different diagnostic methods at the same time devoting special attention to
preventive measures and their practical application.
The main objective of the present dissertation
To carry out epidemiological investigations of PI-3 and RS viral
infections in the Lithuanian cattle farms, to establish the duration of passive
and active immunity and to find out the possibilities of immune response
using different routes of vaccination.
Goals of the study
1. To determine the prevalence of bovine PI-3 and RS viruses in the
Lithuanian cattle farms.
2. To compare the prevalence of bovine PI-3 and RS viral infections
with the prevalence of other cattle–relevant viral diseases at Lithuanian
cattle farms.
3. To determine the geographical distribution patterns for PI-3 and RS
viruses.
4. To conduct comparative analysis of laboratory diagnostic methods for
PI-3 and RS viral infections.
5. To investigate the dynamics and duration of colostral immunity to
bovine PI-3 and RS viruses and its influence on the morbidity of calves.

6. To conduct a comparative investigation of different routes of
vaccination – intramuscular (IM) and intradermal (ID) – and different doses.
Scientific importance and novelty of research
1. The prevalence and epidemiological peculiarities of PI-3 and RS
viruses in the Lithuanian cattle farms were determined.
2. A comparative analysis prevalence of bovine PI-3, RS, IBR and BVD
viral infections was conducted.
3. A comparative assessment of various diagnostic investigations was
performed.
4. The seroprevalence of bovine PI-3 and RS viral infections in the
Lithuanian counties was determined.
5. The duration of passive colostral immunity to bovine PI-3 and RS
viruses and its influence on morbidity of calves was determined.
6. It was determined that ID vaccination against PI-3 and RS viral
infections is effective and easily accomplished.
Practical importance
1. It was determined that the main agents of respiratory diseases in the
Lithuanian cattle herds are PI-3 and RS viruses.
2. It was proved that combined employment of Enzyme linked
immunosorbent assay (ELISA Ab) reaction and polymerase chain reaction
(PCR) methods can guarantee a prompt and effective identification of PI-3
and RS viral infections in cattle herds.
3. The duration of passive colostral immunity, which is important in
veterinary practice for development of vaccination programmes against PI-3
and RS infections, was determined.
4. It was found out that the intradermal (ID) vaccination against PI-3 and
RS viral infections may be as an alternative for the common vaccination
routes.
5. A memo of differential diagnostics of bovine respiratory diseases and
other economically important diseases was developed for veterinary
surgeons.
2. MATERIALS AND METHODS
The doctoral research was carried out in six phases. The first phase
included the epidemiological investigations of RS and PI-3 viruses for
determining the serological incidence of the mentioned agents in the
Lithuanian cattle population. Also the serological incidence of PI-3 and RS
viruses in two chosen cattle farms with similar cattle rearing technologies

was analysed in detail using Enzyme linked immunosorbent assay (ELISA)
and hemagglutination binding reactions. The blood samples for laboratory
analysis of bovine PI-3 and RS viruses were taken from the cattle herds of
15 Lithuanian farms in 9 districts. The chosen farms had histories of the
symptoms characteristic of bronchopneumonia. A total of 347 single blood
samples were examined for antibodies against PI-3 and RS viruses. For
determining the spread trends of PI-3 and RS viruses, the results obtained
during the investigations carried out in 2006–2007 were analysed. A total of
935 blood sample data from 36 farms were analysed for antibodies against
PI-3 viruses and 905 from 33 farms for RS viruses.
The second phase was devoted to comparison of bovine PI-3 and RS
viral infections versus other relevant bovine viral infections in Lithuania.
The investigation of the spread of IBR, PI-3, RS and BVD viruses was
conducted by ELISA. For this purpose, 553 blood samples were examined
for antibodies against PI-3 and BVD viruses and 538 samples for antibodies
against RS and IBR viruses. The incidence of these viruses was investigated
in 20 farms from 11 districts holding more than 200 head of cattle.
During the third phase of the present research, the geographical
distribution of PI-3 and RS viruses in the Lithuanian cattle farms was
determined. For this purpose, in 2006–2013, 2225 blood samples were
serologically tested for PI-3 virus and 2142 samples for RS virus.
In the fourth phase, the identification of PI-3 virus in the clinical
material was performed by ELISA whereas for RNA detection of RS virus
by reverse transcription polymerase chain reaction (RT–PCR). A total of 96
samples of pathological material (nasal discharges) were collected from
cattle of different age (treated or untreated by antimicrobials) held in the
farms in 13 Lithuanian districts.
The fifth investigation phase was devoted to determining the duration of
the passive immunity and its influence on occurrence of respiratory diseases
in calves. For this purpose the blood samples from the calves were taken and
the level of antibodies to PI-3 and RS viruses were assessed. At the same
time the health parameters of all groups of calves were monitored.
In the sixth phase, the immune response was investigated after
vaccination using different routes – intramuscular (IM) and intradermal (ID),
and different application doses (4 ml and 0.4 ml). For determining the
amount of antibodies, i.e. the dynamics and duration of post–vaccination
immunity, 126 blood samples were serologically analysed. The samples
were taken from cattle held in one farm. The trial included 21 calves from 3
weeks of age. The calves were respectively allocated into three groups – 7
animals in each. The first blood samples for serological analysis were taken
at the age of three weeks and, later, at the age of 6, 10, 18, 27 and 35 weeks.

2.1. Immunological and virusological research methods
2.1.1. Enzyme linked immunosorbent assay (ELISA) as a method for
determining antibodies against PI-3 and RS viruses in blood serum
Commercial standartized Enzyme linked immunosorbent assay (ELISA)
kits (Institute Pourquier, France) were used for the detection of antibodies in
blood serum against bovine PI-3 and RS viruses. These kits are designed for
determining the specific IgG antibodies against bovine PI-3 viruses and
antigens of RS viruses in blood serum. The diagnostic kit included plastic
microplates with wells covered with antigens of bovine PI-3 and RS viruses.
The analysed sera (control and tested) were diluted and poured into 96 wells
of plates. When a sample contained specific antibodies against PI-3 and RS
viruses, during the incubation there developed complexes of antigen and
antibody, i.e. the antibodies of the tested animals adhered to the plate. The
optical density was evaluated by reading using spectrophotometer. The
antibody titre was entered up using the system of pluses; from 1+ to 5 +.
2.1.2. Enzyme linked immunosorbent assay (ELISA) as a method for
determining antigens of PI-3 and RS viruses in blood serum
For analysis of antigens of PI-3 and RS viruses, IFA commercial
diagnostic kits BIO K Pulmotest (Belgium) were used. The microtitration
plates with 96 wells were sensibilised using specific antibodies against PI-3
and RS viruses. The reaction was evaluated by colour intensity when
measuring the optical density.
2.1.3. Hemagglutination binding reaction for determining the antibody
titres against PI-3 viruses
The antibody titres against PI-3 viruses were determined by standard
hemagglutination binding reaction using guinea–pig erythrocytes and
twofold sera dilutions (Rossi & Kiesel, 1971).
The reaction was performed using plates with 96 wells and V–shaped
bottom. The reaction in the wells without hemagglutination was regarded
positive because the specific antibodies, contained in the serum, inhibited
virus induced agglutination.
2.1.4. Reverse transcription polymerase chain reaction (RT–PCR) for
detection of RNA of RS virus
The samples also were analysed for RS viruses using polymerase chain
reaction technique. The extraction of the RNA of the virus was performed

using phenol chloroform isoamyl alcohol and genomic methods. For
diagnosing by polymerase chain reaction, the primers of oligonucleotides
were used allowing specific identifying of bovine RS viruses alone (Vilcek
at al., 1994).

2.1.5. Investigation of colostral immunity duration and health status of
calves
The passive colostral immunity was investigated via the evaluation of
the dynamics of antibody titres and number of seropositive calves. The
passive immunity level in calves aged 24–48 hours was determined by
semi–quantitative sodium sulphate precipitation method. 24 calves from one
farm were selected for the trial based on the IgG concentration in the blood
serum. The calves were allocated into three groups 8 individuals in each:
group I – IgG>15g/l, group II – 5–15 g/l and group III –

The confidence interval (CI) of serological distribution of PI-3 and RS
viruses in cattle population and reliability of distribution percentage
differences at 95 % of probability were calculated using “Dimension
Research” Inc. software.
The comparative statistical analysis of different vaccination routes also
was conducted using SPSS program „Statistic 20”. The differences between
the number of seropositive animals in the three groups (2 vaccinated and 1
unvaccinated) at different times were evaluated by the Fisher’s exact test.
The differences among the mentioned three groups at different times were
evaluated by the nonparametric Kruskal–Wallis test. When statistically
significant differences were established, the groups were compared in pairs
using the Mann–Whitney–Wilcoxon test.
The differences between the same groups at different times were
evaluated by the Friedman test. When statistically significant differences
were established by Friedman test, the groups were compared in pairs using
Wilcoxon Signed Ranks test (SPSS IBM, New York, USA, 2011).
3. RESULTS
The serological investigations carried out in 2006–2012 showed that RS
and PI-3 viruses are rather widespread in the Lithuanian cattle population. It
also is obvious that without preventive measures these viruses are able to
persist for a long time in a herd and permanently or sporadically cause
respiratory diseases in cattle.

The evaluation of the data for 2006, showed a rather high prevalence of
both viruses: PI-3 72.8 % (CI 68.5–77.1) and RS 56.8 % (CI 51.9–61.7). A
statistically significant decrease of seropositive animals in respect to RS
virus was observed in 2009 and 2012 whereas in respect to PI-3 virus in
2008, 2009 and 2012.

comparatively short duration of passive and active immunity (Hodgins et al.,
2002).
Analysis of the distribution of antibodies against RS virus showed that
the greater part of animals in farm 3 had no antibodies and were susceptible
to RS virus. Yet high antibody titres in some animals were indicative of the
recent history of disease. Such animals pose threat to susceptible individuals.
The age dynamics of antibodies against RS virus in farm 3 also showed that
some animals had a history of disease until the age of two years. It has been
established that after the termination of immunity, animals may get
reinfected in cow group (De Jong et al., 1996). Meanwhile, most of the
samples from farm 1 had high antibody titres what is indicative of recent
history of infection (Schrijver et al., 1996; Hagglund, 2005).
The dynamics of seropositive animals and antibody titres in different age
groups of cattle showed that RS viruses also pose danger for heifers and
cows over two years of age causing serious respiratory damage (De Jong et
al., 1996). The dynamics of the number of seropositive animals in different
age groups also showed that in farm 1 even 66.7 % young animals had
antibodies against PI-3 viruses, i.e. they already had a history of this disease
(Lazić et al., 2009). The higher titres of antibodies in cow group older than
four years are indicative of repeated infection yet usually PI-3 viruses are
less pathogenic in older animals (Graham et al., 1999). However in farm 3,
animals younger than 2 years of age had no antibodies what was indicative
of the terminated immunity. When included in the group of cows the
animals got infected repeatedly (Lazić et al., 2009).
The complex evaluation of the incidence of bovine viral infections
confirmed the hypothesis that in Lithuania not only PI-3 and RS viruses are
agents of respiratory diseases. It was determined that 95 % of investigated
farms had animals with antibodies against PI-3 and RS viruses. There were
fewer farms where IBR and BVD infections were detected serologically:
84.2 and 85 % respectively. Most farms (15 of 20; 75 %) had animals with a
history of all four viral diseases and only in five herds (25%) the animals
had histories of two or three viral diseases, i.e. in most of the herds mixed
viral diseases were prevalent.
The analysis of geographical distribution of PI-3 and RS viruses showed
high incidence of these viruses in all Lithuanian counties. The greatest
losses were inflicted by PI-3 virus in the Panevėžys, Kaunas and Telšiai
counties. Yet in other regions of the country the percentage of incidence is
rather high. The highest number of seropositive animals against RS virus
was identified in the Utena, Telšiai, Šiauliai, and Marijampolė counties.
Thus, we may state that the incidence of PI-3 and RS viruses is high all over
the country and, presumably, is the main cause of the recent frequent

outbreaks of endemic respiratory diseases. It should be pointed out that the
largest cattle breeding farms (with the highest density of animals) are
concentrated in North and Central Lithuania. Namely in these herds other
viral infections are extremely widespread (Mockeliūnienė et al., 2004;
Gulliksen et al., 2009; Jacevičius et al., 2010).
The tests for PI-3 and RS virus antigens and nucleic acids showed that
only some of the samples from infected calves contained viruses though the
serological analysis indicated that the majority of farms had animals with
antibodies against PI-3 viruses (91.7 %) and RS viruses (91.2 %). Animals
shedding PI-3 viruses were only identified in 23.1 % and shedding RS
viruses in 30.8 % of farms. Thus the positive results of single serological
tests are not necessarily indicative of virus–induced infection. The obtained
results partly correlate with the results of paired blood serum tests during
which it was determined that seroconversion (i.e. confirmed infection) was
characteristic only of 19.0–23.8 % of infected calves. Similar results have
been obtained by Swedish researchers who, investigating the outbreaks of
respiratory diseases, identified seroconversion of PI-3 and RS viruses in 17
and 22 % of cases respectively (Hagglund, 2005). This can be accounted for
by that the some tested calves still had colostral immunity which may
interfere with the induction of the active immunity after infection. The
absence of seroconversion during infection is characteristic of RS viruses
and it is associated with the interference of the residual passive immunity
(Hartel et al., 2004).
Yet the detection of PI-3 virus antigen and nucleic acids of RS virus in
the pathological material of infected calves showed that five samples were
PI-3 seropositive (5.2 %) and six samples were RS seropositive (6.25 %).
These data seem to be at variance with the results of serological tests. Yet it
is known that the viruses from the Paramyxoviridae family are extremely
labile, i.e. after infection, they are detected in the respiratory airway only at
the beginning of disease and for a very short time (Antonis et al., 2010).
Also it should be reminded that during the present study, the animals
shedding PI-3 viruses were identified in 3 (23.1 %) farms and RS viruses in
4 (30.8 %) farms. These data also are at variance with our data of single
serological test according to which even 95 % of all herds had seropositive
animals. This phenomenon can be explained by the fact that the majority of
seropositive animals is found in the adult group where PI-3 and RS viral
infections often manifest only sub–clinically. Also it should be born in mind
that bovine respiratory diseases are caused not only by viral infections but
also by plenty other factors (Härtel et al., 2004; Taylor et al., 2010).
The quality and duration of colostral immunity are a very important
index for prediction of the health status of calves and establishing the

optimal time for vaccination against respiratory diseases (Arthington et al.,
2000). The present complex investigation showed that the maximal duration
(age of seropositive calves) of passive immunity and antibody titres
depended on the quality of colostral immunity, i.e. on IgG content in the
blood serum of calves. The statistical analysis of the passive immunity
showed that the half–life of antibodies for PI-3 viruses was 28.55 days and
for RS viruses 35.18 days.
For establishing the optimal vaccination time, the statistical mean
duration of immunity is a very important index. The present research
revealed that this index for PI-3 and RS viruses was 41.19 and 50.75 days
respectively. Thus, the logical conclusion is that the optimal age of calves
for vaccination would be not less than 1.5 months, since it has been
determined that the huge amounts of antibodies acquired through the passive
immunity interfere with vaccine antigen and inhibit the formation of
humoral immunity (Parish et al., 2003; Fulton et al., 2004).
The trial for the influence of passive immunity on the health status of
calves showed that irrespective of the quality of the passive immunity in
general the morbidity and mortality caused by respiratory diseases were the
same (p>0.05). Yet in the calves who got sufficient amount of colostrum the
clinical signs were milder and lasted shorter. Concomitantly a smaller
number of treatment courses was necessary (p

against viral diseases and its effectiveness using different vaccines are
lacking (Meyer et al., 2008).
In the present research, the immune response of calves was evaluated
using the intradermal (ID) route of vaccination and compared with the
response after the standard intramuscular (IM) vaccination commonly
applied against PI-3 and bovine RS viruses.

The research revealed that calves vaccinated intradermally using 1/10
(0.4 ml) of the common doses (indicated in the approved instruction)
demonstrated immune response against bovine RS and PI-3 viruses.
However, the response was somewhat weaker if compared with the response
of calves vaccinated intramuscularly using the full dose (4 ml).
The obtained result could be explained by the fact that 0.4 ml is a rather
small dose responsible for weaker immune response and lower Ab titres
(Eble et al., 2009). Therefore it seems likely that a higher dose of vaccine
administered intradermally could produce a better immune response
(Bernardy et al., 2008; Eble et al., 2009). The high level of maternal
antibodies in the blood serum at the beginning of the trial also could be the
cause of lower immune response after the initial vaccination than expected
(Fulton et al., 2004).

The most important advantages of the intradermal administration are that
it is less invasive and painless, as well as quickly and easily performed.
Furthermore it should be emphasized that ID vaccination needs a reduced
volume of injection as compared to the more conventional intramuscular
route (Zhao et al., 2006; Goubier et al., 2008). The main disadvantage of the
ID technique is that the jet injection devices are not common equipments in
the veterinary practice therefore it remains the probability of injection faults
(Bernardy et al., 2008).
Recently, vaccination as a preventive measure against respiratory
diseases in the Lithuanian cattle farms has become especially relevant. The
main obstacle to its implementation refers to the necessary huge investments
into expensive vaccines. Therefore ID vaccination could be a perfect
preventive alternative as it requires minimal expenditures. It should be
pointed out that there are no data about cattle vaccination against PI-3 and
RS viruses yet this vaccination route is an object of great interest as a more
promising measure in comparison with the IM vaccination commonly
practiced for many years.

4. CONCLUSIONS
1. The respiratory syncytial and/or parainfluenza-3 infections are among
the main causes of respiratory diseases in cattle. In 91.7 % of cattle farms
animals with antibodies against PI-3 virus and in 91.2 % of farms animals
with antibodies against RS agents were identified. In the infected farms, the
numbers of PI-3 and RS seropisitive animals accounted on the average for
59.1 % and 53.1 % respectively.
2. The important role of PI-3 and RS viruses for bovine respiratory
diseases was confirmed by virus antigen and nucleic acids detection
methods: in 23.1 % of cattle farms animals shedding PI-3 viruses and in
30.8 % of farms animals shedding RS viruses were identified. Respectively,
PI-3 and RS viruses were detected in 5.2 % and 6.25 % of the total of
animals with clinical signs.
3. The several of viruses were identified as agents of respiratory diseases
occurring in the Lithuanian cattle farms. The analysis of PI-3, RS, IBR and
BVD incidence showed that in 75.0 % of cattle farms there were animals
with a history of all four viral diseases. Only in 25.0 % of cattle farms,
animals with a history of two or three viral diseases were identified.
4. The serological analysis of geographical distribution of RS and PI-3
viruses showed 53–79 % (PI-3) and 17–84 % (RS) seropositive animals in
different Lithuanian counties. The biggest number of seropositive animals
was determined in North and Central Lithuanian counties.
5. The half–time of colostral antibodies against PI-3 viruses was 28.55
days and against RS viruses 35.18 days. The average duration of the passive
immunity against PI-3 and RS viruses was 41.19 and 50.75 days
respectively.
6. Depending on the quality of colostral immunity, the duration of
respiratory diseases in young animals ranged from 3.25 to 6.13 days. In
general, the morbidity and mortality rates were comparable but the calves
who received sufficient amount of colostrum were sick for a shorter time
and suffered milder clinical signs.
7. The after vaccination indexes of immune response using different
vaccination routes (IM and ID) and different doses (4 ml and 0.4 ml) were
statistically comparable throughout the 35 weeks long trial. This implies that
ID vaccination can be a cheaper alternative to the commonly applied IM
vaccination.

1. ĮVADAS
Kvėpavimo takų ligomis galvijų prieauglis serga visose pramoninės
gyvulininkystės šalyse. Šie susirgimai daro didžiulių ekonominių nuostolių
tiek pieno, tiek mėsos pramonėje (Smith, 2000; Van der Fels–Klerx et al.,
2001). Užsienio ir Lietuvos mokslininkų tyrimai rodo, kad veršeliai
bronchopneumonija, kitaip dar vadinama enzootine pneumonija, suserga dėl
daugelio priežasčių, todėl apibendrintai šios ligos dar vadinamos galvijų
respiracinių ligų (GRL) kompleksu (Hagglund, 2005).
Tirdami galvijų prieauglio kvėpavimo takų ligas mokslininkai pastebėjo,
kad pagrindiniai bronchopneumonijų arba GRL komplekso sukėlėjai yra
paragripo-3 (PG-3) virusai ir respiraciniai sincitiniai (RS) virusai, rečiau –
galvijų infekcinio rinotracheito (GIR) virusai, adenovirusai, koronavirusai ar
mikoplazmos (Storz et al., 2000; Šiugždaitė, 2002; Snowder et al., 2006;
Autio et al., 2007). Dažniausiai virusai aptinkami kartu su tokiomis
bakterijomis kaip Manheimia hemolytica, Pasteurella multocida,
Haemophilus somnus, Salmonella dublin ir Arcanobacterium pyogenes.
GRL kompleksą sudaro keli skirtingos epidemiologijos sindromai. Šiam
kompleksui būdingi įvairūs klinikiniai simptomai ir epidemiologiniai
požymiai (Radostits et al., 2000).
Kvėpavimo takų ligomis serga įvairaus amžiaus galvijai, bet dažniausiai
– prieauglis tuo metu, kai susilpnėja pasyvusis imunitetas ir dar nėra
susidaręs aktyvusis imunitetas (Radostits et al., 2000). GRL būdingas
sezoniškumas – šie susirgimai dažniausi šaltuoju metų laiku, tačiau ligos
gali pasireikšti ir vasarą, ypač esant drėgnesniam ir vėsesniam orui
(Mockeliūnas ir kt., 2005; Hagglund, 2005).
Vertinant atskirus bronchopneumonijų sukėlėjus, dėl svarbios
etiologinės reikšmės GRL komplekse reikėtų išskirti paragripo-3 ir
respiracinius sincitinius virusus (Elvander, 1996; Larsen, 2000; Autio et al.,
2007).
Galvijų paragripą sukelia Paramixoviridae šeimos Respirovirus genties
viengubos spiralės RNR virusai. Galvijų PG-3 virusas antigeniškai panašus į
žmonių ir avių PG-3 virusus, tačiau nėra jiems identiškas (Henrickson,
2003). Dėl bandoje cirkuliuojančių PG-3 virusų visada yra respiracinių ligų
protrūkio grėsmė, nes virusai sudaro palankias sąlygas antrinėms
bakterinėms infekcijoms, ir galvijai gali užsikrėsti mikoplazmomis bei kitais
patogenais. Esant šiam sukėlėjui bandoje, klinika gali ir nepasireikšti, jei
nesusidarys tam tikros aplinkos sąlygos ar neprisidės kiti patogenai. PG-3
virusinė infekcija dažniausiai pasireiškia švelnia klinika, gyvuliai gaišta retai.
PG-3 dažnai pasireiškia kartu su infekcinio galvijų rinotracheito, galvijų
virusinės diarėjos ir RS virusais, Pasteurella multocida ir Haemophilus

somnus (Cusack et al., 2003; Autio et al., 2007; Calderon et al., 2007;
Griffin et al., 2010).
Galvijų respiracinis sincitinis (RS) virusas yra Paramixoviridae šeimos
Pneumovirus genties viengubos ribonukleorūgšties virusas. RS virusai
plačiai išplitęs visame pasaulyje tarp galvijų, avių, ožkų bei kitų gyvūnų
(Easton et al., 2004). Kaip ir PG-3 sukėlėjas, RS virusai tampa pirmine
respiracinių susirgimų priežastimi ir paprastai aptinkami kartu su kitais
patogenais (O’Neill et al., 2006; Valarcher et al.; 2006). Susirgimas
dažniausiai pasireiškia galvijams iki 6 mėn., tačiau ligos protrūkiai galimi ir
tarp suaugusių galvijų (Patel, Didlick, 2004).
Galvijų respiraciniai sincitiniai ir paragripo-3 virusai turi ypač didelę
reikšmę Lietuvos galvijų bandoms, nes gali sukelti kasmetinius ligos
protrūkius tiek prieaugliui, tiek ir suaugusiems galvijams (Šalomskas ir kt.,
2008). Šių sukėlėjų susirgimai ir jų daroma žala Lietuvos ūkiams nėra
tyrinėta ir pastaruosius dešimt metų apie šias ligas mokslinių publikacijų
nėra. Tai svari priežastis analizuoti ir gilintis į šių virusinių infekcijų
paplitimo mastą šalies galvijų ūkiuose bei identifikuoti sukėlėjus taikant
skirtingus diagnostikos metodus, kartu skiriant ypatingą dėmesį prevencijos
priemonių galimybėms ir jų praktiniam pritaikymui.
Darbo tikslas Atlikti PG-3 ir RS virusų infekcijų epidemiologinius tyrimus Lietuvos
galvijininkystės ūkiuose, nustatyti pasyvaus ir aktyvaus imuniteto trukmę
bei išaiškinti imuninio atsako galimybes, naudojant skirtingus vakcinacijos
būdus.
Darbo uždaviniai
1. Nustatyti galvijų PG-3 ir RS virusų paplitimą Lietuvos galvijų ūkiuose.
2. Palyginti galvijų PG-3 ir RS virusinių infekcijų paplitimą su kitomis
Lietuvos galvijų ūkiams aktualiomis virusinėmis ligomis.
3. Nustatyti galvijų paragripo-3 ir respiracinio sincitinio virusų paplitimo
geografinius ypatumus.
4. Atlikti PG-3 ir RS virusinių infekcijų laboratorinės diagnostikos
metodų palyginamąjį tyrimą.
5. Ištirti krekeninio imuniteto galvijų PG-3 ir RS virusams dinamiką ir
trukmę bei jo įtaka veršelių sergamumui.
6. Atlikti vakcinacijos palyginamąjį tyrimą naudojant skirtingus
įraumeninį (IM) ir įodinį (ID) vakcinacijos būdus ir bei skirtingas vakcinos
dozes.

Darbo mokslinė reikšmė ir naujumas

Darbo praktinė reikšmė
1. Nustatyta, kad Lietuvos galvijų bandose pagrindiniai kvėpavimo takų
ligų sukėlėjai yra PG-3 ir RS virusai.
2. Įrodyta, kad kartu naudojant imunofermentinės analizės (IFA Ak) ir
polimerazės grandininės reakcijos (PGR) metodus, galima greitai ir
efektyviai nustatyti PG-3 ir RS virusų infekcijas galvijų bandose.
3. Išaiškinta pasyvaus krekeninio imuniteto trukmė, svarbi veterinarinėje
praktikoje, kuriant efektyvias vakcinacijos programas nuo PG-3 ir RS virusų
infekcijų.
4. Nustatyta, kad įodinė (ID) vakcinacija nuo PG-3 ir RS virusinių
infekcijų gali tapti alternatyva įprastiems vakcinavimo būdams.
5. Sukurta ir pasiūlyta galvijų kvėpavimo takų ir kitų ekonomiškai
svarbių ligų diferencinės diagnostikos atmintinė praktikuojantiems
veterinarijos gydytojams.
2. MEDŽIAGOS IR METODAI
Tiriamasis darbas buvo atliekamas šešiais etapais. Pirmame etape buvo
atliekami RS ir PG-3 virusų epidemiologiniai tyrimai, kuriais išaiškintas
minėtų sukėlėjų serologinis paplitimas Lietuvos galvijų populiacijoje. Taip
pat atlikome detalų PG-3 ir RS virusų serologinio plitimo tyrimus,
pasirinktuose dviejuose ūkiuose su panašiomis galvijų auginimo
technologijomis, taikėme imunofermentinės analizės ir hemagliutinacijos
stabdymo reakcijas. Laboratoriniams galvijų PG-3 ir RS virusų paplitimo
Lietuvoje analizės tyrimams kraujo mėginiai buvo paimti iš 15 ūkių galvijų
bandų, esančių 9 rajonuose. Atrankai parinkti tie ūkiai, kuriuose galvijams
buvo pasireiškę bronchopneumonijoms būdingi simptomai. Iš viso dėl
antikūnų prieš PG-3 ir RS virusus buvo ištirti 347 vienkartiniai kraujo
mėginiai. Siekiant nustatyti RS ir PG-3 virusų plitimo tendencijas buvo

analizuojami ir 2006–2007 metais atliktų tyrimų rezultatai. Iš viso dėl
antikūnų prieš PG-3 virusus išanalizuoti 935 iš 36 ūkių, o dėl RS virusų –
905 kraujo mėginių tyrimo duomenys iš 33 ūkių.
Antrame darbo etape, norėdami palyginti galvijų PG-3 ir RS virusinių
infekcijų paplitimą galvijams aktualių virusinių ligų atžvilgiu Lietuvoje,
lygiagrečiai atlikome GIR, PG-3, RS ir GVD virusų paplitimo tyrimus
taikant imunofermentinės analizės reakciją. Tuo tikslu PG-3 ir GVD virusų
antikūnų atžvilgiu buvo ištirti 553 kraujo mėginiai, o dėl antikūnų prieš RS
ir GIR virusus – kraujo 538 mėginiai. Šių virusų paplitimas išanalizuotas 11
rajonų esančiuose 20 ūkių, kur buvo laikomos didesnės nei 200 galvijų
bandos.
Trečiame darbo etape buvo atliktas PG-3 ir RS virusų geografinio
paplitimo Lietuvos galvijų ūkiuose tyrimas. Tuo tikslu 2006–2013 metais
serologiškai buvo ištirta 2225 krajo mėginiai PG-3 viruso atžvilgiu ir 2142
mėginiai RS viruso atžvilgiu.
Ketvirtame etape PG-3 virusams identifikuoti klinikinėje medžiagoje
buvo atliekama IFA reakcija, o RS viruso genominės RNR išskyrimui
taikėme AT-PGR reakciją. Iš viso surinkti ir ištirti 96 patologinės medžiagos
mėginiai (šnervių išskyros), paimti iš 13 įvairiuose Lietuvos rajonuose
esančių ūkių, įvairaus amžiaus galvijų, gydytų ar negydytų
antimikrobinėmis medžiagomis.
Penktame darbo etape atlikome pasyvaus imuniteto trukmės tyrimus ir jo
įtaką veršelių sergamumui kvėpavimo takų ligomis.Vertinimui paimti ir
ištirti kraujo mėginiai, nustatytas antikūnų lygis PG-3 ir RS virusų atžvilgiu.
Tyrimo metu taip pat stebėti veršelių sergamumo parametrai.
Šeštame etape tyrėme imuninį atsaką, atliekant skirtingus įraumeninį
(IM) ir įodinį (ID) vakcinacijos būdus bei naudojant skirtingas vakcinos
dozes (4 ml ir 0,4 ml). Antikūnų nustatymui siekiant išaiškinti povakcininio
imunitento dinamiką ir trukmę, buvo surinkta ir serologiškai ištirta 126
kraujo mėginių iš viename ūkyje laikomų galvijų. Iš viso šiame tyrime
dalyvavo 21 veršelis nuo 3 savaičių. Veršeliai suskirstyti į tris grupes,
kiekvienoje grupėje po 7 gyvulius. Mėginiai serologiniams tyrimams pradėti
rinkti veršeliams 3 sav. amžiuje, toliau tęsiama: 6, 10, 18, 27 ir 35 savaičių
amžiuje.
2.1. Imunologiniai ir virusologiniai tyrimų metodai
2.1.1. Imunofermentinės analizės (IFA) tyrimo metodas galvijų PG-3 ir
RS virusų antikūnams kraujo serume nustatyti
Kraujo mėginių tyrimams dėl antikūnų galvijų PG-3 ir RS virusams
buvo naudojami komerciniai standartizuoti imunofermentinės analizės (IFA)

rinkiniai (Institute Pourquier, Prancūzija). Jie skirti specifiniams IgG
antikūnams galvijų PG-3 ir RS virusų antigenams kraujo serume nustatyti.
Diagnostinį rinkinį sudarė plastikinės mikroplokštelės, kurių šulinėliai
padengti galvijų RS ir PG-3 virusų antigenais. Analizuojami serumai
(kontrolės ir tiriamieji mėginiai) buvo praskiesti ir išpilstyti į plokšteles su
96 duobutėmis. Jei kuriame nors mėginyje buvo specifinių antikūnų prieš
RS ar PG-3 virusus, inkubacijos metu susidarė antigeno ir antikūno
kompleksai, t. y. tiriamųjų galvijų antikūnai prisitvirtino prie plokštelės.
Optinis tankis buvo nuskaitomas IFA spektrofotometru, antikūnų titras buvo
užrašomas pliusų sistema nuo 1+ iki 5+.
2.1.2. Imunofermentinės analizės (IFA) tyrimo metodas galvijų PG-3 ir
RS virusų antigenams kraujo serume nustatyti
PG-3 ir RS virusų antigeno tyrimams buvo naudojami IFA komerciniai
diagnostikumai (BIO K Pulmotest, BIOX, Belgija), kur 96 duobučių
mikrotitravimo ploštelės sensibilizuotos specifiniais antikūnais prieš PG-3 ar
RS virusą. Reakcija buvo vertinama pagal mėginio spalvos intensyvumą
matuojant optinį tankį.
2.1.3. Hemagliutinacijos stabdymo reakcija antikūnų titrams prieš PG-3
virusus nustatyti
Antikūnų titrai prieš PG-3 virusus taip pat buvo nustatomi standartine
hemagliutinacijos stabdymo reakcija, naudojant jūrų kiaulytės eritrocitus ir
dukartinius serumo atskiedimus (Rossi & Kiesel, 1971).
Reakcija buvo atliekama mikrometodu 96 duobučių plokštelėse su V
formos dugnu. Tose plokštelės įdubėlėse, kur visiškai nebuvo
hemagliutinacijos, reakcija laikyta teigiama, nes serume esantys specifiniai
antikūnai slopino virusų sukeliamą eritrocitų agliutinaciją.
2.1.4. Atvirkštinės transkripcijos polimerazinė grandininė reakcija (AT-
PGR) RS viruso genominei RNR nustatyti
Mėginiai dėl RS virusų buvo tiriami ir polimerazės grandinine reakcija.
Viruso RNR ekstrakcija buvo atliekama fenolo chloroformo izoamylo
alkoholio ir Genomic metodais. Polimerazės grandininei reakcijai RS
diagnozuoti naudoti oligonukleotidų pradmenys, leidžiantys specifiškai
identifikuoti tik galvijų RS virusus (Vilcek et al., 1994):

2.1.5. Krekeninio imuniteto trukmės ir veršelių sveikatingumo tyrimas
Pasyvaus krekeninio imuniteto tyrimo metu vertinome antikūnų titro ir
seroteigiamų veršelių skaičiaus dinamiką. Tyrimo pradžioje 24–48 val.
amžiaus veršelių pasyvaus imuniteto lygis nustatytas pusiau kiekybiniu
natrio sulfito precipitacijos metodu. Taip pagal kraujo serumo IgG kiekį
tyrimui atrinkti viename ūkyje laikomi 24 veršeliai, suskirstyti į tris grupes
po 8 individus: I grupė – IgG kiekis >15g/l; II grupė – 5–15 g/l ir III grupė

Kruskal-Wallis testu. Jeigu buvo nustatyta statistiškai reikšmingų skirtumų,
grupės buvo lyginamos poromis naudojant Mann-Whitney-Wilcoxon testą.
Skirtumai lyginant tas pačias grupes skirtingais laiko momentais buvo
įvertinti Friedman testu. Jeigu Friedman testu buvo nustatyta statistiškai
reikšmingų skirtumų, grupės buvo lyginamos poromis naudojant Wilcoxon
Signed Ranks testą (SPSS IBM, New York, JAV, 2011).
3. TYRIMŲ REZULTATAI
2006–2012 metais atlikti serologiniai tyrimai parodė, kad RS ir PG-3
virusai yra labai išplitę Lietuvos galvijų populiacijoje. Taip pat akivaizdu,
kad, nesiimant profilaktikos priemonių šie virusai gali ilgą laiką persistuoti
bandoje ir taip nuolat arba periodiškai sukelti galvijų kvėpavimo takų
susirgimus.

Tyrimų pradžioje, vertinant 2006 m. duomenis, nustatytas pakankamai
didelis abiejų virusų paplitimas – PG-3 – 72,8 proc. (CI 68,5–77,1) ir RS –
56,8 proc. (CI 51,9–61,7). Statistinis seroteigiamų gyvulių mažėjimas RS
viruso atžvilgiu stebimas 2009 ir 2012 m. tuo tarpu PG-3 atžvilgiu jis
fiksuojamas 2008, 2009 bei 2012 metais.

Seroteigiamų skaičiaus ir antikūnų titrų dinamika įvairaus amžiaus
galvijų grupėse rodo, kad RS virusai pavojingi ir vyresnėms nei 2 metų
amžiaus telyčioms bei karvėms, nes šie virusai sukelia pavojingus plaučių
pažeidimus suaugusiems galvijams (De Jong et al., 1996). PG-3 virusų
infekcijos seroteigiamų galvijų skaičiaus dinamika įvairaus amžiaus grupėse
rodo, kad pirmame ūkyje net 66,7 proc. jaunų galvijų turėjo antikūnus prieš
PG-3 virusus, t. y. jie jau buvo persirgę (Lazić et al., 2009). Padidėjęs
antikūnų titras vyresnių nei 4 metų karvių grupėje rodo pakartotinę infekciją,
tačiau paprastai PG-3 virusai vyresnio amžiaus galvijams yra mažai
patogeniški (Graham et al., 1999). Kitokia padėtis nustatyta trečiame ūkyje.
Čia galvijai iki 2 metų amžiaus antikūnų neturi. Veikiausiai dėl to, kad
galvijams jau pasibaigęs imunitetas, dėl ko jie pakartotinai užsikrečia
pervesti į karvių grupę (Lazić et al., 2009).
Kompleksinis galvijų virusinių infekcijų paplitimo tyrimas patvirtino
hipotezę, kad Lietuvoje kvėpavimo takų ligas galvijams sukelia ne tik PG-3
ar RS virusai. Nustatyta, kad 95 proc. visų tirtų ūkių buvo rasta galvijų,
turinčių antikūnų prieš PG-3 ir RS virusus. Ūkių, kur infekcinio galvijų
rinotracheito (GIR) ir galvijų virusinės diarėjos (GVD) virusų infekcija
nustatyta serologiškai, buvo kiek mažiau – atitinkamai 84,2 proc. ir 85 proc.
Daugumoje ūkių (15 iš 20; 75 proc.) rasta galvijų, persirgusių visomis
keturiomis virusinėmis ligomis, tik penkiose (25 proc.) bandose galvijai
buvo persirgę dviem arba trimis virusinėmis ligomis, t. y. daugumoje bandų
vyravo mišrios virusinės infekcijos.
Atlikus PG-3 ir RS virusų geografinio paplitimo galvijų ūkiuose analizę
nustatytas didelio masto virusų paplitimas visose Lietuvos apskrityse. PG-3
virusas galimai didžiausius nuostolius gyvulių augintojams kelia Panevėžio,
Kauno ir Telšių apskrityse, tačiau ir kituose šalies regionuose procentinio
paplitimo rodikliai yra pakankamai dideli. Tuo tarpu RS viruso didžiausias
procentas seroteigiamų galvijų nustatytas Utenos, Telšių, Šiaulių ir
Marijampolės apskrityse. Taigi, galime teigti, kad PG-3 ir RS virusai yra
labai išplitę visoje šalyje ir gali būti pastaruoju metu dažnai pasireiškiančių
galvijų endeminių kvėpavimo takų ligų priežastis. Reikia pažymėti, kad
Šiaurės ir Vidurio Lietuvoje yra sutelkta didžioji dalis stambių
galvijininkystės ūkių, čia yra didžiausias galvijų tankumas, ir būtent tokiose
bandose labai išplitę kitos virusinės ligos (Mockeliūnienė et al., 2004;
Gulliksen et al., 2009; Jacevičius et al., 2010).
PG-3 ir RS virusų antigeno ir nukleorūgščių nustatymas parodė, kad tik
dalis sergančių veršelių mėginių paėmimo metu išskyrė virusus, nors virusų
serologinis paplitimas rodė, kad daugumoje (91,7 proc.) ūkių buvo rasta
galvijų turinčių antikūnų prieš PG-3 virusus; 91,2 proc. ūkių buvo rasta
galvijų, turinčių antikūnų prieš RS sukėlėjus. PG-3 virusus išskiriančių buvo

rasta tik 23,1 proc. ūkių, o RSV išskiriantys galvijai buvo nustatyti 30,8 proc.
ūkių. Taigi, teigiami vienkartinių serologinių tyrimų rezultatai nebūtinai
rodo, kad tyrimo metu ligą sukelia būtent virusai. Šie tyrimų rezultatai iš
dalies sutampa su mūsų atliktais poriniais kraujo serumo tyrimų rezultatais,
kurių metu nustatyta, kad serokonversija (t. y. patvirtintas užsikrėtimas)
nustatytas tik 19,0–23,8 proc. sergančių veršelių. Panašius rezultatus yra
gavę švedų mokslininkai, kurie tyrė kvėpavimo takų ligų protrūkius ir PG-3
bei RS virusų serokonversiją, radę atitinkamai 17 proc. ir 22 proc. atvejų
(Hagglund, 2005). Tą galima paaiškinti tuo, kad daliai veršelių tyrimai buvo
atliekami dar esant neišnykusiam krekeniniam imunitetui, kuris gali
interferuoti su aktyvaus poinfekcinio imuniteto susidarymu. Serokonversijos
nebuvimas po užsikrėtimo yra būdinga RS virusų infekcijos savybė ir
siejama su likutinio pasyvaus imuniteto interferencija (Hartel et al., 2004).
Tačiau sergančių veršelių patologinėje medžiagoje nustatytas PG-3
viruso antigenas ir RS viruso nukleorūgštys parodė, kad tik penki mėginiai
buvo PG-3 teigiami (5,2 proc.), RS teigiamų buvo rasta šešiuose (6,25 proc.)
mėginiuose. Tokie duomenys, atrodytų, prieštarauja serologinių tyrimų
rezultatams, betgi žinoma, kad Paramyxoviridae šeimos virusai yra itin
labilūs, po infekcijos veršelių kvėpavimo takuose aptinkami labai trumpai ir
tik ligos pradžioje (Antonis et al., 2010). Reikia paminėti, kad mes PG-3
virusus išskiriančius galvijus nustatėme trijuose (23,1 proc.), o RS virusus –
keturiuose (30,8 proc.) ūkiuose. Šie duomenys taip pat prieštarauja mūsų
vienkartinio serologinio tyrimo duomenims, kurie rodo, kad net 95 proc.
visų bandų randami seroteigiami galvijai. Šį fenomeną galima būtų
paaiškinti tuo, kad didžioji dauguma seroteigiamų galvijų randama
suaugusiųjų grupėje, kur PG-3 ir RS virusų infekcija dažnai pasireiškia
subklinikine forma. Taip pat reikia nepamiršti, kad galvijų kvėpavimo takų
ligas nulemia ne tik virusų infekcijos, bet ir daugybė kitų veiksnių (Hartel et
al., 2004; Taylor et al., 2010).
Labai svarbus rodiklis, pagal kurį galima prognozuoti veršelių
sveikatingumą ir nustatyti optimalų vakcinacijos nuo kvėpavimo takų ligų
laiką – krekeninio imuniteto kokybė ir trukmė (Arthington et al., 2000).
Kompleksinio tyrimo metu nustatėme, kad pasyvaus imuniteto maksimali
trukmė (seroteigiamų veršelių amžius) ir antikūnų titrai priklausė nuo
krekeninio imuniteto kokybės, t. y. nuo IgG kiekio veršelių kraujo serume.
Statistinė pasyvaus imuniteto analizė parodė, kad antikūnų pusinio
eliminavimo laikas PG-3 virusams buvo 28,55 dienos, o RS virusams –
35,18 dienos.
Vakcinacijos laikui parinkti labai svarbi ir statistinė vidutinė imuniteto
trukmė. Mūsų tyrimai parodė, kad šis rodiklis PG-3 ir RS virusams buvo
atitinkamai 41,19 ir 50,75 dienos. Taigi, galime daryti išvadą, kad

optimaliausias veršelių vakcinavimo amžius būtų ne mažesnis nei 1,5
mėnesio, nes nustatyta, kad veršelių organizme pasyviai įgyti dideli
antikūnų kiekiai interferuoja su vakcinos antigenu ir aktyvus humoralinis
imunitetas nesusidaro (Parish et al., 2003; Fulton et al., 2004).
Pasyvaus imuniteto įtakos veršelių sveikatingumui tyrimas parodė, kad
nepriklausomai nuo gauto pasyvaus imuniteto kokybės, bendras veršelių
sergamumas ir gaištamumas nuo kvėpavimo takų ligų nesiskyrė (p>0,05).
Tačiau veršeliai, gavę pakankamai krekenų, sirgo trumpiau ir lengvesne
forma. Tai parodė mažesnis gydymo kursų skaičius (p

vakcinacijos (3 savaičių) nebuvo gautas visavertis imuninis atsakas, kokio
tikėtasi (Fulton et al., 2004).

Vienas svarbiausių ID vakcinacijos privalumų yra tas, kad atliekama
mažesnė invazija, procedūra neskausminga, ją atlikti yra greita ir paprasta.
Reikia pažymėti, kad palyginti su įprastine IM vakcinacija, ID reikalingas
mažesnis injekcijos tūris (Zhao et al., 2006; Goubier et al., 2008). Šio
metodo trūkumas yra tas, kad jis nėra įprastas, o veterinarijos specialistai
neturi tam tikslui pritaikytų tinkamų įrankių. Dėl šių priežasčių išlieka rizika
vakcinaciją atlikti netinkamai (Bernardy et al., 2008).
Pastaruoju metu vakcinacija, kaip prevencijos priemonė nuo respiracinių
susirgimų, Lietuvos galvijų ūkiuose yra labai aktuali, tačiau pagrindinė
įvardijama problema trukdanti tai įgyvendinti, yra šiam tikslui reikalingos
didžiulės piniginės investicijos dėl brangių vakcinų. Taigi ID vakcinacija
būtų puiki alternatyva siekiant įgyvendinti prevenciją, tam skiriant
minimalias išlaidas. Reikėtų pabrėžti, kad apie įodinę galvijų vakcinaciją
nuo PG-3 ir RSV sukėlėjų nėra apskritai jokių duomenų, tuo tarpu šis
vakcinavimo būdas kelia didžiulį susidomėjimą kaip perspektyvus ir
alternatyvus įprastiniams ir taikomiems jau daugelį metų.
4. IŠVADOS
1. Viena pagrindinių galvijų kvėpavimo takų ligų priežasčių yra respiracinių sincitinių ir (ar) paragripo-3 virusų infekcija. 91,7 proc. ūkių
buvo rasta galvijų turinčių antikūnų prieš PG-3 virusus. 91,2 proc. ūkių
išaiškinti galvijai, turintys antikūnų prieš RS sukėlėjus. Užkrėstuose ūkiuose
seroteigiamų PG-3 ir RS virusams galvijų vidutiniškai buvo atitinkamai 59,1
proc. ir 53,1 proc.
2. PG-3 ir RS virusų etiologinė reikšmė galvijų kvėpavimo takų susirgimuose patvirtinta virusų antigeno ir nukleorūgščių nustatymo
metodais: 23,1 proc. ūkiuose nustatėme PG-3 bei 30,8 proc. ūkiuose – RS
virusus išskiriančius galvijus. PG-3 ir RS virusai rasti atitinkamai 5,2 proc.
ir 6,25 proc. visų sirgusių veršelių.
3. Galvijų kvėpavimo takų ligoms Lietuvos ūkiuose būdingos keleto virusų infekcija. PG-3, RS, GIR ir GVD virusinių infekcijų paplitimo
analizė parodė, kad 75,0 proc. ūkių rasti galvijai, persirgę visomis
keturiomis virusinėmis ligomis. Tik 25,0 proc. bandų nustatyti galvijai,
persirgę dviem arba trimis virusinėmis ligomis.
4. Atlikus RS ir PG-3 virusų serologinio paplitimo geografinę analizę, nustatytas 53–79 proc. (PG-3) ir 17–84 proc.(RS) seroteigiamų galvijų
skaičius įvairiuose Lietuvos apskričių ūkiuose. Daugiausia seroteigiamų
galvijų rasta Šiaurės ir Vidurio Lietuvos apskrityse.

5. Krekeninių antikūnų pusinio eliminavimo laikas PG-3 virusams buvo 28,55 dienos, o RS virusams – 35,18 dienos. Vidutinė pasyvaus imuniteto
trukmė PG-3 ir RS virusams buvo atitinkamai 41,19 ir 50,75 dienos.
6. Priklausomai nuo krekeninio imuniteto kokybės, galvijų prieauglio kvėpavimo takų ligų trukmė svyravo nuo 3,25 iki 6,13 dienos, bendras
sergamumas ir gaištamumas nesiskyrė, tačiau veršeliai, gavę pakankamai
krekenų, sirgo trumpiau ir lengvesne forma.
7. Povakcininio imuninio atsako rodikliai, taikant skirtingus vakcinacijos būdus (IM ir ID) ir skirtingas vakcinos dozes (4 ml ir 0,4 ml),
vakcinuotose grupėse per visą 35 savaičių trukmės eksperimento laiką
statistiškai nesiskyrė. Tai įrodo, kad ID vakcinacija gali būti pigesnė
alternatyva įprastai taikomam IM vakcinavimo būdui.
5. PRAKTINĖS REKOMENDACIJOS
Parengta ir pateikta disertacijos prieduose:
1. Rekomendacijos galvijų kvėpavimo takų ligų diagnostikai ir
prevencijai.
2. Standartinis kvėpavimo takų ligų vakcinacijos planas.
3. Galvijų kvėpavimo takų ir kitų ekonomiškai svarbių ligų diferencinės
diagnostikos atmintinė.
4. Mokslinio tyrimo (metodo, išradimo, prototipo, prietaiso,
technologijos ar pan.) pristatymas LSMU: ID vakcinacija palyginti su IM
vakcinacija turi daug privalumų: yra pigesnė, mažiau skausminga, mažiau
dirginami audiniai, todėl ID vakcinacija gali tapti alternatyva įprastai
taikomam IM vakcinacijos būdui.
