Blood Vessel Staining Kit

CHEMICON’s Blood Vessel Staining Kit is a sensitive immunohistochemicaltool for the detection of vascular endothelial cells.

CHEMICON’s Blood Vessel Staining Kits utilize the two most widely accepted cell markers for the study of angiogenesis and neovascularization; von Willebrand factor (Factor VIII Related Antigen)and CD31 (PECAM-1) antibodies.

CHEMICON’s von Willebrand factor antibody reactsspecifically with the endothelial cells of blood vessels and is a useful marker for the identification of endothelial lineage in tumors.

CHEMICON’s monoclonal antibody to CD31 is very specific for the staining of vascular endothelial cells in normal and cancerous tissue.

Specimen Treatment andStaining Protocol:

Staining Procedure• Blocking Step

1. Apply about 3 drops (1 drop = ~45 µl) of the ready-to-use Blocking Reagent to cover the specimen.

2. Incubate 15-30 minutes in an enclosed container.

3. Gently rinse the specimen with 1X Rinse Buffer for a minimum of 15 seconds.

• Primary Antibody1. Apply ~150 µl of the diluted Primary Antibody

or a negative control reagent to cover the specimen. Incubate 1–2 hours in an enclosed container.

2. Rinse tissue section 3 x 5 minutes using 1X Rinse Buffer

• Secondary Antibodies1. Apply about 3 drops of the ready-to-use

Secondary Antibody solution to cover the specimen.

2. Incubate 10-15 minutes in an enclosed container.

3. Rinse tissue section 3 x 5 minutes with 1X Rinse Buffer.

• Streptavidin-HRP1. Apply about 3 drops of the ready-to-use

Streptavidin-HRP solution to cover the specimen.

2. Incubate 10-15 minutes in an enclosed container.

3. Rinse tissue section 3 x 5 minutes using 1X Rinse Buffer.

• Chromogen Reagent1. Apply ~150 µl of the freshly prepared

Chromogen Reagent to cover the specimen.2. Incubate 2-10 minutes in an

enclosed container.3. Rinse tissue section for 5-10 minutes under

tap water.
