Each target gene is ranked according to its estimated biological distance from TLR3 among the 12,009 genes in the TLR3 connectome. The sphere of each human

gene surrounding TLR3 is determined as follows: 0 for genes in the top 0.1% in terms of their proximity to TLR3, 1 for the top 1%, 2 for the top 5%, 3 for the top

10%, 4 for the top 25%, 5 for the top 50%, 6 for the top 75% and 7 for the lowest 25%. The P-value specifying the significance of the proximity of a gene to

TLR3 was determined by calculating the probability of a random gene being closer to TLR3 in the connectome. The ratio of the distance between a given gene

and TLR3 to the mean and median values of all TLR3 connectome genes was calculated. The numbers in square brackets in the route description specify the

direct biological distance between the two genes (Eq. 1). The degree of separation from TLR3 is the number of connections on the route predicted between the

gene and TLR3, and the weighted distance was calculated by summing the direct biological distances and multiplying by the degrees of separation (Eq. 2).
